Four molecules comprising a phenol moiety and a distal pyridine base connected by an intermediary H-bonding and an H-bonded alcohol group have been synthesized and their electrochemistry has been investigated by means of cyclic voltammetry. The molecules differ by the substituent at the alcohol functional carbon and by methyl groups on the pyridine. The reaction follows a concerted proton-electron transfer pathway as confirmed by the observation of a significant H/D kinetic isotope effect in all four cases. The standard rate constants characterizing each of the four compounds are analyzed in terms of reorganization energy and pre-exponential factor. Intramolecular and solvent reorganization energies appear as practically constant in the series, in which a previously investigated aminophenol is included, whereas significantly different pre-exponential factors are observed. That the latter, which is a measure of the efficiency of proton tunneling concerted with electron transfer, be substantially smaller with the H-bond relay molecules than with the aminophenol is related to the fact that two protons are moved in the first case instead of one in the second. Within the H-bond relay molecules, the pre-exponential factor varies with the substituent present at the alcohol functional carbon in the order CF(3) > H > CH(3), presumably as the result of a fine tuning of the balance between the H-bond accepting and H-bond donating properties of the central OH group. The kinetic H/D kinetic isotope effect increases accordingly in the same order.  相似文献

The carbon dioxide reforming of methane to synthesis gas was investigated in a dielectric barrier discharge reactor at room temperature. The influence of dilution of reactants by helium was studied. We showed that, at a fixed contact time, the conversions of CH4 and CO2 increase when the amount of helium in the gas mixture increases. This result is attributed to the “penning ionization” phenomenon, which corresponds to an energy transfer from excited He to molecules in ground state (CH4, CO2). The selectivity to products is affected by the dilution factor. As soon as helium is present in a large amount the formation of products resulting from recombination of methyl radicals (such as C2, C3 and C4) is less favourable due to the lowest probability of collisions to proceed. A kinetic model is proposed based on the assumption that the reactant molecules CH4 or CO2 are attacked by active species produced by the plasma discharges, and the production of this active species are function of the plasma power. This model which takes into account the dilution by helium fits particularly well the experimental data we obtained.  相似文献

The incidence of fungal infections, in particular candidiasis and aspergillosis, has considerably increased during the last three decades. This is mainly due to advances in medical treatments and technologies. In high risk patients (e.g. in haematology or intensive care), the prognosis of invasive candidiasis is relatively poor. Therefore, a rapid and correct identification of the infectious agent is important for an efficient and prompt therapy. Most clinical laboratories rely on conventional identification methods that are based on morphological, physiological and nutritional characteristics. However, these have their limitations because they are time-consuming and not always very accurate. Moreover, molecular methods may be required to determine the genetic relationship between the infectious strains, for instance in Candida outbreaks. In addition, the latter methods require time, expensive consumables and highly trained staff to be performed adequately. In this study, we have applied the FTIR spectroscopic approach to different situations encountered in routine mycological diagnosis. We show the potentials of this phenotypic approach, used in parallel with routine identification methods, for the differentiation of 3 frequently encountered Candida species (C. albicans, C. glabrata and C. krusei) by using both suspensions and microcolonies. This approach, developed for an early discrimination, may help in the initial choice of antifungal treatment. Furthermore, we demonstrate the feasibility of the method for intraspecies comparison (typing) of 3 Candida species (C. albicans, C. glabrata and C. parapsilosis), particularly when an outbreak is suspected.  相似文献

We have established that polyhydroxylated styrylquinolines are potent inhibitors of HIV-1 integrase (IN). Among them, we have identified (E)-8-hydroxy-2-[2-(4,5-dihydroxy-3-methoxyphenyl)-ethenyl]-7-quinolinecarboxylic acid (1) as a promising lead. Previous molecular dynamics simulations and docking procedures have shown that the inhibitory activity involves one or two metal cations (Mg2+), which are present in the vicinity of the active center of the enzyme. However, such methods are generally based on a force-field approach and still remain not as reliable as ab initio calculations with extended basis sets on the whole system. To go further in this area, the aim of the present study was to evaluate the predictive ability of the electron density and electrostatic properties in the structure-activity relationships of this class of HIV-1 antiviral drugs. The electron properties of the two chemical progenitors of 1 were derived from both high-resolution X-ray diffraction experiments and ab initio calculations. The twinning phenomenon and solvent disorder were observed during the crystal structure determination of 1. Molecule 1 exhibits a planar s-trans conformation, and a zwitterionic form in the crystalline state is obtained. This geometry was used for ab initio calculations, which were performed to characterize the electronic properties of 1. The electron densities, electrostatic potentials, and atomic charges of 1 and its progenitors are here compared and analyzed. The experimental and theoretical deformation density bond peaks are very comparable for the two progenitors. However, the experimental electrostatic potential is strongly affected by the crystal field and cannot straightforwardly be used as a predictive index. The weak difference in the theoretical electron densities between 1 and its progenitors reveals that each component of 1 conserves its intrinsic properties, an assumption reinforced by a 13C NMR study. This is also shown through an excellent correlation of the atomic charges for the common fragments. The electrostatic potential minima in zwitterionic and nonzwitterionic forms of 1 are discussed in relation with the localization of possible metal chelation sites.  相似文献

In the class of NADH:acceptor oxidoreductases, the diaphorase from Bacillus stearothermophilusis a particularly promising enzyme for sensing NADH, and indirectly a great number of analytes, when coupled with a NAD-dependent dehydrogenase as well as for the design of mono- and multienzyme affinity sensors. The design and rational optimization of such systems require devising immobilization procedures that prevent dramatic losses of the enzymatic activity and a full kinetic characterization of the immobilized enzyme system. Two immobilization procedures are described, which involve recognition of the biotinylated diaphorase by a monolayer of neutravidin adsorbed on the electrode surface either directly or through the intermediacy of a monolayer of biotinylated rabbit immunoglobulin. Thorough kinetic characterization of the two systems is derived from cyclic voltammetric responses. A precise estimate of the enzyme coverages is obtained after comparing the enzyme kinetics of the immobilized and the homogeneous system.  相似文献

The phosphorylation of the neuronal Tau protein modulates both its physiological role of microtubule binding and its aggregation into paired helical fragments observed in Alzheimer's diseased neurons. However, detailed knowledge of the role of phosphorylation at specific sites has been hampered by the analytical difficulties to evaluate the level of site-specific phosphate incorporation. Even with recombinant kinases, mass spectrometry and immunodetection are not evident for determining the full phosphorylation pattern in a qualitative and quantitative manner. We show here that heteronuclear NMR spectroscopy on a 15N labeled Tau sample modified by the cAMP dependent kinase allows identification of all phosphorylation sites, measures their level of phosphate integration, and yields kinetic data for the enzymatic modification of the individual sites. Filtering through the 15N label discards the necessity of any further sample purification and allows the in situ monitoring of kinase activity at selected sites. We finally demonstrate that the NMR approach can equally be used to evaluate potential kinase inhibitors in a straightforward manner.  相似文献

Dichloro and chloromethyl Ga(III) complexes of general formulae [XClGa-eta2-{R2P(E)NP(E'R'2-E,E'}](X = Cl, R, R'= Ph, E, E'= O (1), S (2), Se (3); R = Ph, R'= OEt, E = O, E'= S (4); R = Me, R'= Ph, E, E'= S (5) and X = Me, E, E'= O (6), S (7), Se (8)) were synthesised by either metathesis reactions between GaCl3 and the potassium salt of the ligand (X = Cl) or by methane eliminations from in situ prepared GaMe2Cl and the protonated ligands LH (X = Me). Redistribution reaction of (3) in either CDCl3 or THF afforded the solvent-free tetracoordinate gallium spirocycle cation [Ga-{eta2-{Ph2P(Se)NP(Se)Ph2-Se,Se'})2]+ (9+). The molecular structures of complexes 2, 4, 5, 7 and 9(+) show non-planar gallacycle rings.  相似文献

To study the photobleaching of the main fluorescent compounds of the arterial wall, we repeatedly measured the time-resolved fluorescence of elastin, collagen and cholesterol during 560 s of excitation with nitrogen laser pulses. Three fluence rate levels were used: 0.72, 7.25 and 21.75 microW/mm2. The irradiation-related changes of the fluorescence intensity and of the time-resolved fluorescence decay constants were characterized for the emission at 390, 430 and 470 nm. The fluorescence intensity at 390 nm decreased by 25-35% when the fluence delivered was 4 mJ/mm2, a common value in fluorescence studies of the arterial wall. Cholesterol fluorescence photobleached the most, and elastin fluorescence photobleached the least. Photobleaching was most intense at 390 nm and least intense at 470 nm such that the emission spectra of the three compounds were markedly distorted by photobleaching. The time-resolved decay constants and the fluorescence lifetime were not altered by irradiation when the fluence was below 4 mJ/mm2. The spectral distortions associated with photobleaching complicate the interpretation of arterial wall fluorescence in terms of tissue content in elastin, collagen and cholesterol. Use of the time-dependent features of the emission that are not altered by photobleaching should increase the accuracy of arterial wall analysis by fluorescence spectroscopy.  相似文献

As an illustration of how cyclic voltammetry can be used to unravel the mechanisms and kinetics of redox enzymes, the reductive dechlorination of trichloroethylene and tetrachloroethylene by a typical reductive dehalogenase, the tetrachloroethene reductive dehalogenase of Sulfurospirillum multivorans (formerly called Dehalospirillum multivorans), was investigated by means of several electrochemically generated cosubstrates. They comprised the monocation and the neutral form of methylviologen, the neutral form of benzylviologen, and cobaltocene. Cyclic voltammetry is used to produce the active form of the cosubstrate under controlled potential conditions. It shows large plateau-shaped catalytic responses, which are used to measure the kinetics of the enzymatic reaction as a function of the substrate and cosubstrate concentrations. The variation of the rate constant for the cosubstrate reaction with its standard potential shows the transition between two asymptotic behaviors, one in which the reaction is under diffusion control and the other in which it is under counter-diffusion control. Simple fitting of this plot allows an estimation of the standard potential of the electron acceptor center in the enzyme (E degrees = -0.57 V vs NHE).  相似文献
