We propose a method of optical waveguiding, which relies on Bragg diffractions from a 1D grating that gives rise to waveguiding in the direction normal to the grating wave vector. The waveguide structure consists of a shallow 1D grating that has a bell- or trough-shaped amplitude in the confinement direction. The 2'-hydroxyl group contributes inextricably to the functional behavior of many RNA molecules, fulfilling numerous essential chemical roles. To assess how hydroxyl groups impart functional behavior to RNA, we developed a series of experimental strategies using an array of nucleoside analogs. These strategies provide the means to investigate whether a hydroxyl group influences function directly (via hydrogen bonding or metal ion coordination), indirectly (via space-filling capacity, inductive effects, and sugar conformation), or through interactions with solvent. The nucleoside analogs span a broad range of chemical diversity, such that quantitative structure activity relationships (QSAR) now become possible in the exploration of RNA biology. We employed these strategies to investigate the spliced exons reopening (SER) reaction of the group II intron. Our results suggest that the cleavage site 2'-hydroxyl may mediate an interaction with a water molecule. 相似文献

A minimum thickness gate valve design for mass spectrometry is described in detail. The ion optics required to transmit ions from the source to the ICR cell are integrated into the design to minimize fringe field effects on the ions as they travel through the gate valve. The total thickness of the complete gate valve assembly is 1.03 in. (26.2 mm) with a maximum fringe field distance of 0.065 in. (1.7 mm). The gate valve is able to maintain a vacuum of <10(-10) mbar at the ICR cell when the source is vented to atmosphere and the estimated ion transfer efficiency is >95%. 相似文献

In this work the separation of eighteen flavonoids was attempted using reduced-flow micellar elektrokinetic chromatography (RF-MEKC) electrolytes modified by selected solvents with differing properties: methanol (MeOH), acetonitrile (ACN) and tetrahydrofuran (THF). Structural aspects such as unsaturation of the C ring, number and position of OH groups, methylation and glycosylation as well as solvent effects and their impact on the electrophoretic behavior of flavonoids were addressed. By evaluating the electropherograms obtained from mixture-designed electrolytes and searching for changes in the critical pairs, a favorable separation condition was achieved using 20 mmol/L phosphate buffer at pH 2.5 containing 50 mmol/L sodium dodecyl sulfate (SDS), 15% ACN and 5% THF (one critical pair) in less than 12 min with 1.5% coefficient of variation (CV) for retention factor and 3% CV for peak area (n = 5). Alumina-supported Au particles (1.16 wt %) were prepared by a deposition-precipitation method involving a HAuCl4 precursor. X-ray absorption spectroscopy at the Au L(III) edge was used to monitor the evolution of the Au oxidation state and atomic structure during pretreatment in He up to 623 K. Although the as-prepared material had Au present in a +3 oxidation state, thermal treatment at 623 K facilitated autoreduction of Au cations to metal particles. Analysis of the EXAFS revealed a coordination number (Au-Au) of 7.2, which is consistent with spherical particles of 1.2 nm in average diameter. Steady-state isotopic transient kinetic analysis was used to evaluate the intrinsic turnover frequency (TOF intr) and the surface coverage of carbon-containing species (theta COx) on the gold catalyst during CO oxidation at 1.2 atm total pressure and 296 K. The artifacts in the kinetic parameters caused by re-adsorption of product carbon dioxide were removed by varying the total flow rate. The values of TOF intr and theta COx determined from the intrinsic lifetime of surface intermediates at infinite flow rate were 1.6 s(-1) and 4.9%, respectively. The intrinsic turnover frequency was nearly independent of temperature, indicating a very low activation energy for the reaction. Singers are extremely conscious of health problems that affect their voices and well-being and often take an active role in seeking care for these problems. They frequently seek treatment from providers or with modalities considered “alternative” to traditional medical care. A survey of singers was completed to elucidate their attitudes and practices regarding “alternative modalities” of medical care. Frequently singers will self-medicate or take advice from people not well versed in the special needs of a professional voice user. They will fail to share this information with the physician when seeking “traditional” medical care. These practices may predispose the singer to suboptimal or even dangerous care. These results are discussed, as well as the implications for the medical physician treating the singer. The possible detrimental pharmacologic effects of “natural therapies” widely used by singers are presented, with special attention to the particular concerns for the professional singer 相似文献
