Radio-high-performance liquid chromatography for ecotoxicity assessment of insect growth regulators

The described radiochromatographic method permits fast and high-sensitivity monitoring of soil biodegradation products of an insect growth regulator for its environmental risk assessment. We analyzed and compared two diastereoisomers of ethyl N-(2-(4-[(2-hydroxycyclohexyl) methyl]phenoxy)ethyl)carbamate, namely its cis-(1S,2S) isomer JN-W330 and a trans-(1R,2S) isomer JN-W331. Microbial conversion of the cis-isomer to the trans-isomer was proved by mass spectrometry analyzer. Among the chromatographic columns tested, the best separation was found with a 125 mm x 4 mm i.d. column packed with Supersphere 100 RP-C18, 5 microm and an acetonitrile-water gradient. The detection limit for the both isomers was in the range of 120-250 Bq (0.3-0.8 ng) at a concentration of 2 ng/ml with radiometric detection. The calibration curves for standard solutions were linear in the range of 150Bq-150kBq (r = 0.996). The method enabled us to compare the analyzed juvenoids with biologically active oostatic peptides in terms of their environmental safety.     

Photoinduced metalation of nonactivated C-Cl bonds with samarium diiodide: synthesis of alkenes with high (Z)-selectivity through beta-elimination reactions

[reaction: see text] The photoinduced metalation of nonactivated C-Cl bonds of O-acetyl chlorohydrins is promoted by samarium diiodide. As a result of this, beta-elimination of O-acetyl chlorohydrins is achieved, affording the corresponding (Z)-alkenes with total or high stereoselectivity.     

The first fluorogenic assay for detecting a Baeyer-Villigerase activity in microbial cells

The first fluorogenic assay allowing for detection of microbial enzymes able to perform Baeyer-Villiger oxidation is described. This is based on the use of 4-oxopentyl umbelliferyl ether 1 as a fluorogenic substrate. When Baeyer-Villigerases active against this test ketone are present in the selected whole cells, 1 is transformed into 3-hydroxypropyl umbelliferyl ether 3, which, in a subsequent step, releases the fluorescent product umbelliferone. Different microorganisms, known to be endowed with Baeyer-Villigerase activity, were assayed.     

Determination of Low Contents of Fenpiverine Bromide by Extraction Spectrophotometry

An ion-pair extraction spectrophotometry method was developed for the determination of fenpiverine bromide in tablets. To determine this substance it was necessary to find experimental conditions that would allow eliminating the influence of other components of the tablets. Attention was paid to the fact that a suitable pH of water phase was necessary to achieve high selectivity.Received December 11, 2002; accepted April 24, 2003 Published online July 28, 2003     

Dynamic contact angle and spreading rate measurements for the characterization of the effect of dentin surface treatments

A new methodology capable of providing reliable and reproducible contact angle (theta) data has been employed to study the effect of clinical treatments grinding, acid etching, and deproteinization on medial dentin tissue. It is based on the application of the ADSA-CD algorithm to the determination of low-rate dynamic contact angles, obtained from slowly growing drops, and on contact angle measurement, as well as spreading behavior analysis, during the relaxation of the system (water on treated dentin) after initial drop growth. The theta data obtained were substantially more reproducible than those obtained with classical methods. A net effect of the treatment on theta was found, increasing dentin wettability: theta (polished) >theta (etched) >theta (deproteinized). The spreading rates correlate with the angles and are adequate for the dentin surface characterization. ANOVA and SNK tests show that for advancing contact angles the means corresponding to all treatments are significantly different. In the relaxing phase, mean angle and spreading rates on polished dentin differ significantly from those on etched and deproteinized dentin, but the latter do not differ significantly from each other.     

An advantageous route to oxcarbazepine (trileptal) based on palladium-catalyzed arylations free of transmetallating agents

[reaction: see text] A new route to oxcarbazepine (Trileptal), the most widely prescribed antiepileptic drug, starting from commercially available 2'-aminoacetophenone and 1,2-dibromobenzene, is reported. The sequentially accomplished key steps are palladium-catalyzed intermolecular alpha-arylation of ketone enolates and intramolecular N-arylation reactions. After several experiments to establish the best conditions for both arylation processes, the target oxcarbazepine is obtained in a satisfactory overall yield, minimizing the number of steps and employing scalable catalytic procedures developed in partially aqueous media.     

Host-guest complexes of crown ethers with HN-proton-donor molecules

Crown ethers were reacted with HN-proton-donor molecules to obtain crystalline molecular host-guest complexes. Molecular Beam Mass Spectrometry System for Characterization of Thermal Plasma Chemical Vapor Deposition

A molecular beam mass spectrometry system for in situ measurement of the concentration of gas phase species including radicals impinging on a substrate during thermal plasma chemical vapor deposition (TPCVD) has been designed and constructed. Dynamically controlled substrate temperature was achieved using a variable thermal contact resistance method via a backside flow of an argon/helium mixture. A high quality molecular beam with beamtobackground signal greater than 20 was obtained under film growth conditions by sampling through a small nozzle (75 m) in the center of the substrate. Mass discrimination effects were accounted for in order to quantify the species measurements. We demonstrate that this system has a minimum detection limit of under 100 ppb. Quantitative measurements of hydrocarbon species (H, H₂, C, CH₃, CH₄, C₂H₂, C₂H₄) using Ar/H₂/CH₄ mixtures and silicon species (Si, SiH, SiH₂, SiCl, SiCl₂, Cl, HCl) using Ar/H₂/SiCl₄ mixtures were obtained under thermal plasma chemical vapor deposition conditions.
