The complex [Ru(bpy)2(dafo)](PF6)2, where bpy is 2,2-bipyridine and dafo is diazafluorenone crystallizes in the space group P21/n witha=9.505(3) Å,b=14.002(4) Å andc=25.783(8) Å. The coordination geometry of the Ru atom is that of a distorted octahedron with a RuN6 core. The two Ru-N bond distances to the dafo ligand are 2.13(1) and 2.15(1) Å; the four Ru-N bond distances to the bipyridine ligands are 2.03(1), 2.05(1), 2.06(1), and 2.07(1) Å. The three shortest Ru-N distances aretrans to the three longest Ru-N distances. The complex is oxidized and reduced reversibly at 1.41 and –0.65 V vs. SSCE, respectively. It displays absorptions at 438 nm (1.6×104), 285 nm (6.2×104), and 240 nm (4.1×104) and a broad emission centered at 626 nm in water at room temperature. The emission lifetime is 420 ns and the emission quantum yield is 5.3×10–4.  相似文献

The physical and photophysical properties of a series of monometallic, [Ru(bpy)(2)(dmb)](2+), [Ru(bpy)(2)(BPY)](2+), [Ru(bpy)(Obpy)](2+) and [Ru(bpy)(2)(Obpy)](2+), and bimetallic, [{Ru(bpy)(2)}(2)(BPY)](4+) and [{Ru(bpy)(2)}(2)(Obpy)](4+), complexes are examined, where bpy is 2,2'-bipyridine, dmb is 4,4'-dimethyl-2,2'-bipyridine, BPY is 1,2-bis(4-methyl-2,2'-bipyridin-4'-yl)ethane, and Obpy is 1,2-bis(2,2'-bipyridin-6-yl)ethane. The complexes display metal-to-ligand charge transfer transitions in the 450 nm region, intraligand pi --> pi transitions at energies greater than 300 nm, a reversible oxidation of the ruthenium(II) center in the 1.25-1.40 V vs SSCE region, a series of three reductions associated with each coordinated ligand commencing at -1.3 V and ending at approximately -1.9 V, and emission from a (3)MLCT state having energy maxima between 598 and 610 nm. The Ru(III)/Ru(II) oxidation of the two bimetallic complexes is a single, two one-electron process. Relative to [Ru(bpy)(2)(BPY)](2+), the Ru(III)/Ru(II) potential for [Ru(bpy)(2)(Obpy)](2+) increases from 1.24 to 1.35 V, the room temperature emission lifetime decreases from 740 to 3 ns, and the emission quantum yield decreases from 0.078 to 0.000 23. Similarly, relative to [{Ru(bpy)(2)}(2)(BPY)](4+), the Ru(III)/Ru(II) potential for [{Ru(bpy)(2)}(2)(Obpy)](4+) increases from 1.28 to 1.32 V, the room temperature emission lifetime decreases from 770 to 3 ns, and the room temperature emission quantum yield decreases from 0.079 to 0.000 26. Emission lifetimes measured in 4:1 ethanol:methanol were temperature dependent over 90-360 K. In the fluid environment, emission lifetimes display a biexponential energy dependence ranging from 100 to 241 cm(-)(1) for the first energy of activation and 2300-4300 cm(-)(1) for the second one. The smaller energy is attributed to changes in the local matrix of the chromophores and the larger energy of activation to population of a higher energy dd state. Explanations for the variations in physical properties are based on molecular mechanics calculations which reveal that the Ru-N bond distance increases from 2.05 ? (from Ru(II) to bpy and BPY) to 2.08 ? (from Ru(II) to Obpy) and that the metal-to-metal distance increases from approximately 7.5 ? for [{Ru(bpy)(2)}(2)(Obpy)](4+) to approximately 14 ? for [{Ru(bpy)(2)}(2)(BPY)](4+).  相似文献

An analytical method is described for detection of endogenous disease-associated prion protein in the buffy coat fraction from the blood of sheep infected with scrapie. The method has been improved and evaluated for its performance in the preclinical diagnosis of ovine transmissible spongiform encephalopathies. The test system uses a protocol for sample preparation that includes extraction and concentration and a test method that uses a liquid-phase competitive immunoassay for prion protein. Antibodies directed to a peptide sequence at the C-terminus of the prion protein (PrP) and a fluorescein-labeled peptide conjugate are used in the assay. Free zone capillary electrophoresis with laser-induced fluorescence for detection is used to separate the antibody-bound fluorescently labeled peptide and free labeled peptide. In this assay, the PrP competes with the fluorescently labeled peptide for limited antibody binding sites, which results in a reduction of the peak representing the immunocomplex of the antibody bound to the fluorescently labeled peptide. When blood samples from scrapie-infected sheep aged 7-12 months and of the scrapie-susceptible PrP genotypes VRQ/VRQ and VRQ/ARQ were analyzed, the abnormal PrP was found in blood samples. These results correlated with the post-mortem diagnosis of scrapie. The sheep were preclinical and appeared normal at the time of testing but later died with clinical disease approximately 12 months after testing. In older animals, and those with clinical signs, a smaller percentage of animals tested positive. This study has demonstrated that this technology can be used as a sensitive, rapid preclinical test to detect the disease-associated PrP in the blood of scrapie-infected sheep. Improvements in the extraction protocol and capillary electrophoresis conditions will enhance the robustness of this test.  相似文献

Dual, independently addressable thin-film resistive heaters fabricated in a multilayer geometry on the surface of an optical fiber provide a new, flexible means for thermally tuning the properties of intracore gratings. In particular, control of the current that is applied to each of these heaters permits the chirp and the central wavelength of the grating to be adjusted independently. The designs and simple fabrication procedures for these types of device, the important physics of heat flow in them, and a tunable add-drop filter that demonstrates essential aspects of their operation are described.  相似文献

Background: Two complementary approaches to studying the cellular function of proteins involve alteration of function either by mutating protein-encoding genes or by binding a small molecule to the protein. A mutagen can generate millions of genetic mutations; correspondingly, split-pool synthesis can generate millions of unique ligands attached to individual beads. Genetic screening of mutations is relatively straightforward but, in contrast, split-pool synthesis presents a challenge to current methods of screening for compounds that alter protein function. The methods used to screen natural products are not feasible for large libraries composed of covalently immobilized compounds on synthesis beads. The sheer number of compounds synthesized by split-pool synthesis, and the small quantity of individual compound attached to each bead require assay miniaturization for efficient screening.Results: We present a miniaturized cell-based technique for the screening of ligands prepared by split-pool synthesis. Spatially defined droplets with uniform volumes of approximately 50–150 nanoliters (depending on well dimensions) are arrayed on plastic devices prepared using a combination of photolithography and polymer molding. Using this microtechnology, approximately 6,500 assays using either yeast cells or mammalian tissue culture can be performed within the dimensions of a standard 10 cm petri dish. We demonstrate that the biological effect of a small molecule prepared by split-pool synthesis can be detected in this format following its photorelease from a bead.Conclusions: The miniaturized format described here allows uniformly sized nanodroplets to be arrayed on plastic devices. The design is amenable to a large number of biological assays and the spatially arrayed format ensures uniform and controlled ligand concentrations and should facilitate automation of assays. The screening method presented here provides an efficient means of rapidly screening large numbers of ligands made by split-pool synthesis in both yeast and mammalian cells.  相似文献

Thin oxide layers on (110) and (100) Si have been studied by ion scattering experiments in a channeling grazing exit angle geometry. Oxides are found to be stoichiometric SiO2 to within 10 Å of the (100) substrate surface, and 7 Å of the (110). The transition interface region between single crystal Si and the SiO2 layer is abrupt and is characterized by approximately one to two monolayers of Si which is out of registration with the substrate lattice for the (110) case. The possibility of a layer of O-deficient oxide is also explored.  相似文献
