The synthesis of the enantiomerically pure bis(hydroxymethyl)-branched cyclohexenyl and cyclohexyl purines is described. Racemic trans-4,5-bis(methoxycarbonyl)cyclohexene [(+/-)-6] was reduced with lithium aluminum hydride to give the racemic diol (+/-)-7. Resolution of (+/-)-7 via a transesterification process using lipase from Pseudomonas sp. (SAM-II) gave both diols in enantiomerically pure form. The enantiomerically pure diol (S,S)-7was benzoylated and epoxidized to give the epoxide 9. Treatment of the epoxide 9 with trimethylsilyl trifluoromethanesulfonate and 1,5-diazabicyclo[5.4.0]undec-5-ene followed by dilute hydrochloric acid gave (1R,4S,5R)-4,5-bis[(benzoyloxy)methyl]-1-hydroxycyclohex-2-ene (10). Acetylation of 10 gave (1R,4S,5R)-1-acetoxy-4,5-bis[(benzoyloxy)methyl]cyclohex-2-ene (11). (1R,4S,5R)-1-Acetoxy-4,5-bis[(benzoyloxy)methyl]cyclohex-2-ene (11) was converted to the adenine derivative 12 and guanine derivative 13 via palladium(0)-catalyzed coupling with adenine and 2-amino-6-chloropurine, respectively. Hydrogenation of 12 and 13 gave the correspondning saturated adenine derivative 14 and guanine derivative 15. (1R,4S,5R)-4,5-Bis[(benzoyloxy)methyl]-1-hydroxycyclohex-2-ene (10) was converted to the adenine derivative 16 and guanine derivative 17 via coupling with 6-chloropurine and 2-amino-6-chloropurine, respectively, using a modified Mitsunobu procedure. Hydrogenation of 16 and 17 gave the corresponding saturated adenine derivative 18 and guanine derivative 19. Compounds 12-19 were evaluated for activity against human immunodeficiency virus (HIV), but were found to be inactive. Further biological testings are underway. 相似文献

An optical method is described, by means of which immunological and enzymatic reactions can be followed at a primary level on a solid surface, without labelling procedures. When plane-polarized light is reflected at a solid surface, there is a minimum in reflectance at a certain angle of incidence, the pseudo-Brewster angle. For example, a layer of protein adsorbed on a silicon surface increases the reflectance with increasing amount of adsorbed material. High sensitivity is obtained because of the large difference in refractive index between silicon and organic material; about 0.1 μg cm?2 adsorbed protein can be detected. In a model system of human IgG and anti-human IgG, the primary adsorption of IgG on a hydrophobic surface is first measured, and on this IgG-coated surface the binding kinetics of anti-IgG could be measured. The kinetics of proteolytic degradation of IgG-coated surfaces by trypsin was also investigated. 相似文献

The synthesis of [4,5-bis(hydroxymethyl)-1,3-oxathiolan-2-yl]nucleosides is described. 2,3-Epoxy alcohol 10 was converted in one pot into thioacetate 11. Treatment of 11 under mild alkaline conditions gave thiirane 12 with inversion of configuration at C-2. We also found that thioacetate 11 rearranges into thiirane 14 under mild acidic conditions. This rearrangement reaction was shown by independent synthesis to proceed with net retention of configuration at C-2. We have proposed a tentative mechanism which may explain the results obtained. Opening of thiiranes 12 and 14 followed by deprotection gave (2R,3R)-2-thiothreitol (23) and (2S,3R)-2-thioerythritol (25), respectively. Regioselective silylation of the primary hydroxyl groups of 23 followed by treatment with trimethyl orthoformate gave 2-methoxy-1,3-oxathiolanes 26 and 27. Condensation with silylated bases followed by deprotection and separation of the anomers gave the oxathiolanylnucleosides. Compounds 29-31, 34, and 35 were found to be inactive when tested for inhibition of HIV-1 activity in vitro. 相似文献

Global glycosylation changes of serum proteins in type 1 diabetic patients have in this paper been investigated based on the interaction of the saccharide moiety of serum proteins with different lectins. Lectins are proteins, which bind carbohydrates specifically and reversibly. Panels with lectins of various carbohydrate specificities were immobilized on gold surfaces. Sera from healthy individuals, newly diagnosed type 1 diabetes patients and type 1 diabetes patients having had the disease for 4-6 years, respectively, were applied to the lectin panel. The biorecognition was evaluated with null ellipsometry. Data obtained were related to an internal standard of lactoferrin. Multivariate data analysis (MVDA) techniques were used to analyze data. Principal component analysis showed that the lectin panel enabled discrimination between sera from the three different above-mentioned groups. Using an artificial neuronal net (ANN), it was possible to correctly categorize unknown serum samples into one of the three groups. 相似文献

The Hessian of the entropy function can be thought of as a metric tensor on the state space. In the context of thermodynamical fluctuation theory Ruppeiner has argued that the Riemannian geometry of this metric gives insight into the underlying statistical mechanical system; the claim is supported by numerous examples. We study this geometry for some families of black holes. In many areas of scientific research and development, for example in the pharmaceutical industry, it is important to prepare
and to characterize crystals of pure organic compounds which are thermodynamically stable. The formation of crystals from
the gas phase is technically less straightforward than crystallisation from solution, but sublimation techniques can have
several important features. In the present paper we report the design and testing of a novel apparatus for small scale sublimation
and fractional deposition of crystals. The instrument has been developed with special reference to the needs in the pharmaceutical
industry. A few mg of the samples are enclosed, under reduced pressure, in thin-walled glass tubes, along which a well defined
temperature gradient can be formed. During an experiment the substance will sublime from the hot end of the glass tube and
crystals will be deposited along the temperature gradient. The applicability of the instrument has been verified by experiments
with several test compounds. Results from experiments with carbamazepine, are reported in some detail. Carbamazepine single
crystals of high quality were obtained and the transition temperature between the triclinic (Form I) and the monoclinic (Form

Interest in the use of the optical properties of chemical indicators is growing steadily. Among the optical methods that can
be used to capture changes in sensing layers, those producing images of large-area devices are particularly interesting for
chemical sensor array development. Until now, few studies addressed the characterization of image sensors from the point of
view of their chemical sensor application. In this paper, a method to evaluate such performance is proposed. It is based on
the simultaneous measurement of absorption events in a metalloporphyrin layer with an image sensor and a quartz microbalance
(QMB). Exploiting the well-known behaviour of QMB, comparison of signals enables estimation of the minimum amount of absorbed
molecules that the image sensor can detect. Results indicate that at the single pixel level a standard image sensor (for example
a webcam) can easily detect femtomoles of absorbed molecules. It should therefore be possible to design sensor arrays in which
the pixels of images of large-area sensing layers are regarded as individual chemical sensors providing a ready and simple
