Angiotensin II (Ang II), which is an important mediator of both vascular responsiveness and growth, has been shown to induce vascular smooth muscle cell (VSMC) hypertrophy via the activation of a complex series of intracellular signaling events. Heat shock protein 70 (Hsp70) has recently been shown to protect against Ang II-induced hypertension. In this study, we tested the hypothesis that Hsp70 can protect VSMC from Ang II-induced hypertrophy. We treated VSMCs with Ang II to induce hypertrophy and to activate MAPK signaling pathway. We observed that the augmentation of Hsp70 expression inhibited Ang II-stimulated VSMC hypertrophy. This inhibitory effect of Hsp70 appears to be partly due to extracellular signal-regulated kinase (ERK1/2) inactivation, which in turn, may possibly result from the accumulation of MAP kinase phosphatase-1 (MKP-1). 相似文献
Recently, it has been reported that curcumin, which is known as a potent antioxidant, acts as a non- stressful and non-cytotoxic inducer of the cytoprotective heme oxygenase (HO)-1. In this study, naturally occurring curcuminoids, such as pure curcumin, demethoxycurcumin (DMC) and bis-demethoxycurcumin (BDMC), were compared for their potential ability to modulate HO-1 expression and cytoprotective activity in human endothelial cells. All three curcuminoids could induce HO-1 expression and HO activity with differential levels. The rank order of HO activity was curcumin, DMC and BDMC. In comparison with endothelial protection against H2O2-induced cellular injury, cytoprotective capacity was found to be highest with curcumin, followed by DMC and BDMC. Interestingly, cytoprotective effects afforded by curcuminoids were considerably associated with their abilities to enhance HO activity. Considering that the main difference among the three curcuminoids is the number of methoxy groups (none for BDMC, one for DMC, and two for curcumin), the presence of methoxy groups in the ortho position on the aromatic ring was suggested to be essential to enhance HO-1 expression and cytoprotection in human endothelial cells. Our results may be useful in designing more efficacious HO-1 inducers which could be considered as promising pharmacological agents in the development of therapeutic approaches for the prevention or treatment of endothelial diseases caused by oxidative damages. 相似文献

The linear and polycatenar type 1H-imidazole amphiphiles showing a strong self-assembly tendency to build various supramolecular structures in bulk were synthesized by the esterification reaction of 4′-alkyloxy phenols (for 1-4) and hydroxyphenyl trialkyloxybenzoates (for 5-7) with 4-chlorocarbonyl imidazole. The linear 1H-imidazole amphiphiles formed thermotropic smectic phases, but compound 4 with an ethyl group instead of hydrogen on 1N did not show a mesophase. The polycatenar type 1H-imidazole amphiphiles (5-7) formed thermotropic hexagonal columnar and cubic phases. A phase transition was observed from a columnar phase to a cubic phase as the temperature increased on heating, and vice versa on cooling. In the POM study for compounds 5-7, optically isotropic phases first appeared on cooling from the isotropic melts, and then birefringent mesophases with a nonspecific texture appeared on further cooling. The X-ray analysis shows that the optically isotropic phases were very likely micellar cubic phases with Pm3n symmetry. The birefringent phases were confirmed as hexagonal columnar phases. 相似文献
We report for the first time the syntheses of electron-precise/deficient alloys, Ln5-xCaxGe4 (Ln = La, Ce; x = 3.37, 3.66, 3.82 for La; x = 3.00, 3.20, 3.26 for Ce), in the metal-rich R5Tt4 Zintl system (R = rare earth metal; Tt = Si, Ge). The new alloys extend the phase width from electron-rich to open-shell electron-deficient region in the metal-rich Zintl system and demonstrate possible occurrence of varied electron deficiencies in Zintl phases without structural changes, as a result of other existing structure-forming factors. 相似文献
The syntheses of arginine-containing hydroxamates using various peptide coupling methods are described. Fmoc-Arg(NO2)-Cl prepared at low temperature did not undergo intramolecular δ-lactam formation and effectively provided desired hydroxamates (8 and 10) in good yields. Fmoc and N-nitro protecting groups can be easily removed. Therefore, this report provides a facile synthesis of arginine-containing peptidomimetic compounds. 相似文献
The growth of gold nanoparticles (NPs) synthesized by sputter deposition on an ionic liquid surface is studied in situ in the bulk phase of the ionic liquids (ILs) 1-butyl-3-methylimidazolium dicyanamide [C(1)C(4)Im][N(CN)(2)], 1-butyl-3-methylimidazolium bis(trifluoromethanesulfonyl)amide [C(1)C(4)Im][Tf(2)N], 1-butyl-3-methylimidazolium tetrafluoroborate [C(1)C(4)Im][BF(4)], 1-butyl-3-methylimidazolium hexafluorophosphate [C(1)C(4)Im][PF(6)] and 1-butyl-3-methylimidazolium triflate [C(1)C(4)Im][TfO]. It is found that primary nanoparticles with a diameter smaller than 2.5 nm are present in the sample immediately after sputtering. Growth of these primary particles proceeds after the end of the sputtering process and stops when the nanoparticles reach a certain size. Depending on the viscosity of the ionic liquid this growth process can proceed several hours to several days. The growth speed is fastest for the least viscous ionic liquid and follows the trend [C(1)C(4)Im][N(CN)(2)] > [C(1)C(4)Im][Tf(2)N] > [C(1)C(4)Im][TfO] > [C(1)C(4)Im][BF(4)] > [C(1)C(4)Im][PF(6)]. It is also found that a higher concentration of sputtered gold results in faster growth of the gold nanoparticles. A discussion on the growth mechanism of sputtered gold NPs is included. 相似文献
Measuring true stress–strain curve over a large-strain-range is essential to understand mechanical behavior and simulate non-linear plastic deformation. The digital image correlation (DIC) technique, a non-contact full-field optical measurement technique, is a promising candidate to obtain a long-range true stress–strain curve experimentally.
Objective
This paper proposes a method for measuring true stress–strain curves over a large-strain-range during tensile testing using DIC.
Methods
The wide-strain-range true stress–strain curves of dual-phase and low carbon steels were extracted on the transverse direction in the neck region. The axial strain on the neck section was estimated by averaging the inhomogeneous deformation on the cross-section of the tensile specimen. The true stress was calculated from the engineering stress and the cross-sectional area of the neck.
Results
The validity of the proposed method was assessed by comparing the experimental load–displacement responses during tensile testing with the finite element method (FEM) simulation results. The stress and strain on the neck section estimated using the FEM and DIC, respectively, were proven to satisfy the uniaxial condition and successfully obtained.
Conclusions
The experimental results agree well with the FEM results. The proposed concept can be applied to various deformation modes for accurately measuring long-range true stress–strain curves.
