C-terminal processing of yeast Spt7 occurs in the absence of functional SAGA complex

Background  

Spt7 is an integral component of the multi-subunit SAGA complex that is required for the expression of ~10% of yeast genes. Two forms of Spt7 have been identified, the second of which is truncated at its C-terminus and found in the SAGA-like (SLIK) complex.     

A simple method for extracting both active oily and water soluble extract (WSE) from Nigella sativa (L.) seeds using a single solvent system

The current study provides a way of extraction for both active NSO and WSE from Nigella sativa seeds using 98% methanol. About 1?kg of ground seeds was macerated by 1:2.5 w/v (g/mL) for 72?hours. After rotary evaporation and 7 days of continuous drying and chilling at 50 and 4?°C, NSO and WSE were obtained at the same instant. Solubility tests of 24 solvents and 11 thin layer chromatographic analyses while 2, 2-diphenyl-1-picrylhydrazyl free radical scavenging assay of NSO (73.66) , WSE (33.32) and NSO?+?WSE (78.22) against ascorbic acid (IC50?=?4.28?mg/mL) was performed. WSE was found to be highly soluble in water and 5% NaOH exhibiting the same Rf value of 0.95 for EtOH:DMSO (9:1) against the honey. WSE has revealed more than twofold higher anti-oxidant activity than others. Formulation of WSE with Tualang honey may provide better targeted hydrophilic drug delivery systems.     

Investigation of the structure of detonation waves in a non-premixed hydrogen–air rotating detonation engine using mid-infrared imaging

The structure of detonation waves propagating through the annular channel of an optically accessible non-premixed rotating detonation engine (RDE) are investigated using mid-infrared imaging. The RDE is operated on hydrogen–air mixtures for a range of air mass flow rates and equivalence ratios. Instantaneous images of the radiation intensity from water vapor are acquired using a mid-infrared camera and a band-pass filter (2.890?±?0.033?µm). The instantaneous mid-infrared images reveal the stochastic nature of the detonation wave structure, position and angle of oblique and reflected shock waves, presence of shear layer separating products from the previous and current cycles, and extent of mixing between the reactants and products in the reactant fill zone in front of the detonation wave. The images show negligible signal directly in front of the detonation waves suggesting that there is minimal mixing between the reactants and products from the previous cycle ahead of the detonation wave for most operating conditions. The mid-infrared images provide insights useful for improving fundamental understanding of the detonation structure in RDEs and benchmark data for evaluating modeling and simulation results of RDEs.     

Solvent isotope effects on interfacial protein electron transfer in crystals and electrode films

D(2)O-grown crystals of yeast zinc porphyrin substituted cytochrome c peroxidase (ZnCcP) in complex with yeast iso-1-cytochrome c (yCc) diffract to higher resolution (1.7 A) and pack differently than H(2)O-grown crystals (2.4-3.0 A). Two ZnCcP's bind the same yCc (porphyrin-to-porphyrin separations of 19 and 29 A), with one ZnCcP interacting through the same interface found in the H(2)O crystals. The triplet excited-state of at least one of the two unique ZnCcP's is quenched by electron transfer (ET) to Fe(III)yCc (k(e) = 220 s(-1)). Measurement of thermal recombination ET between Fe(II)yCc and ZnCcP+ in the D(2)O-treated crystals has both slow and fast components that differ by 2 orders of magnitude (k(eb)(1) = 2200 s(-1), k(eb)(2) = 30 s(-1)). Back ET in H(2)O-grown crystals is too fast for observation, but soaking H(2)O-grown crystals in D(2)O for hours generates slower back ET, with kinetics similar to those of the D(2)O-grown crystals (k(eb)(1) = 7000 s(-1), k(eb)(2) = 100 s(-1)). Protein-film voltammetry of yCc adsorbed to mixed alkanethiol monolayers on gold electrodes shows slower ET for D(2)O-grown yCc films than for H(2)O-grown films (k(H) = 800 s(-1); k(D) = 540 s(-1) at 20 degrees C). Soaking H(2)O- or D(2)O-grown films in the counter solvent produces an immediate inverse isotope effect that diminishes over hours until the ET rate reaches that found in the counter solvent. Thus, D(2)O substitution perturbs interactions and ET between yCc and either CcP or electrode films. The effects derive from slow exchanging protons or solvent molecules that in the crystal produce only small structural changes.

Extending shelling orders and a hierarchy of functions of unimodal simple polytopes

Let P be a simplicial d-polytope with n facets ((d − 1)-dimensional faces) in R^d. A shelling of P is an ordering of the facets of P such that the intersection of each facet F with the union of all facets that precede it the ordering is a nonempty union of (d − 2)-faces of F. The following open question was raised by Tverberg and is recorded in [4]. Suppose for some k < n, there is an ordering of k of the facets of P so that the intersection of each of these facets with the union of all of the facets that precede it in the ordering is a nonempty union of (d − 2)-faces. Can this initial “segment” be extended to a shelling of all the facets? This question is open even in the case that P is the dual of the d-dimensional hypercube. The question in this case has resurfaced several times since G. Danaraj and V. Klee (1978) in a variety of forms. It is related to the hierarchies of completely unimodal pseudo-Boolean functions studied in P.L. Hammer et al. (1988), the author (1988) and D. Wiedemann (1986). (A pseudo-Boolean function is a function mapping the vertices of the d-dimensional hypercube into the reals). In this paper, the hierarchies are compared and combined. This hierarchy is then extended to general simple polytopes, and the relationship to the above open question is explained.
