Formulation study for lansoprazole fast-disintegrating tablet. II. Effect of triethyl citrate on the quality of the products

The purpose of this study was to develop enteric-coated microgranules for the lansoprazole fast-disintegrating tablet (LFDT), which is a rapidly disintegrating tablet containing enteric-coated microgranules. In our previous study, it was clarified that sufficient flexibility of the enteric layer was achieved by the optimized combined ratio of methacrylic acid copolymer dispersion to ethyl acrylate-methyl methacrylate copolymer dispersion and adding the optimized concentration of triethyl citrate to reduce the damage during the compression process. However, since triethyl citrate has an unpleasant bitter taste and is especially incompatible with lansoprazole, it adversely affects the taste and stability of lansoprazole in the enteric-coated microgranules. The enteric layer containing macrogol 6000 was proven useful to improve the unpleasant bitter taste and stability of lansoprazole, because macrogol 6000 does not have an unpleasant bitter taste and is more compatible than triethyl citerate. By covering the inner (first enteric layer) and outer side (third enteric layer) of the enteric layer containing triethyl citrate (second enteric layer) with the enteric layer containing macrogol 6000, we resolved the stability problem of lansoprazole and the unpleasant bitter taste. Finally, we developed enteric-coated microgranules comprising seven layers: 1) core, 2) active compound layer, 3) intermediate layer, 4) first enteric layer, 5) second enteric layer, 6) third enteric layer, and 7) over coating layer. The enteric-coated microgranules have the multiple functions of reducing the damage to the enteric layer during the compression process, improving the stability of lansoprazole, and masking the unpleasant bitter taste.

Efficient algorithms for solving rank two and rank three bilinear programming problems

Finitely convergent algorithms for solving rank two and three bilinear programming problems are proposed. We show that a variant of parametric simplex algorithm can solve large scale rank two bilinear programming problems efficiently. The microwave spectrum of thioformaldehyde,CD2S,and CH2S: Average structure,dipole moments,and 33S quadrupole coupling

Microwave transitions up to J = 53 in the ground vibrational state of deuterothioformaldehyde, CD₂S, were studied between 8 and 40 GHz. A detailed centrifugal distortion analysis yields accurate constants for comparison with force field values. The isotopic species ¹³CH₂S, CH₂³⁴S, CH₂³³S, ¹³CD₂S, CD₂³⁴S, and CD₂³³S were studied in natural abundance. Accurate average zero-point structures were determined for both CD₂S and CH₂S:

Changes in the zero-point geometry for deuterium substitution were established. Quadrupole fine structure arising from the ³³S nucleus has been measured in CH₂³³S and CD₂³³S. Analysis gives the following coupling constants (for both molecules) as χ_aa = ?11.7 and χ_bb - χ_cc = 88.1 MHz. The dipole moment of CD₂S was measured to be 1.6588(8)D and an accurate comparison with CH₂S was made; the ratio of dipole moments $\text{CD}{}_2\text{S}\text{CH}{}_2\text{S}$ was found to be 1.0062(4). The spectroscopic and bonding properties of CH₂S will be compared with formaldehyde and other molecules.     

An automated analyzer for vancomycin in plasma samples by column-switching high-performance liquid chromatography with UV detection

An automated analyzer for vancomycin in rat plasma by column-switching high-performance liquid chromatography (HPLC) with UV detection was developed. The method includes in-line extraction of vancomycin by ion-exchange cartridge column and a separation on a reversed-phase column with UV detection at 215 nm. Plasma samples were diluted by mobile phase solution and directly injected to HPLC. Vancomycin was quantitatively recovered from rat plasma samples. The separation was completed within 15 min. The calibration curve was linear over the range from 0.5 to 100 microg/mL with the detection and quantification limits of 0.5 microg/mL (2.5 ng on column; signal-to-noise ratio = 3). The values of precision in intra- and inter-day assays (n = 3) were less than 1.92 and 3.69%, respectively. This method does not require time-consuming pre-treatment and is suitable for the routine assay of plasma samples.     

Total synthesis of (+)-benzastatin E via diastereoselective Grignard addition to 2-acylindoline

[reaction: see text] A stereoselective total synthesis of (+)-benzastatin E (1) is described. The synthesis involves a diastereoselective Grignard addition to 2-acylindoline 2, which is derived from commercially available (S)-2-indolinecarboxylic acid (3). The unknown absolute configuration of (+)-1 is determined as (9S,10R).     

Synthesis of di-branched heptasaccharide by one-pot glycosylation using seven independent building blocks

[reaction: see text] We describe an efficient synthesis of di-branched heptasaccharide 1 having phytoalexin elicitor activity in soybeans by one-pot glycosylation. The synthesis involves chemo- and regioselective sequential six-step glycosylations using seven independent building blocks and sequential removal of acyl- and benzyl ether-type protecting groups. The coupling of seven building blocks requires only four chemoselective activitable leaving groups of glycosyl donors. Both the glycosylation and deprotection reactions can be achieved utilizing a parallel manual synthesizer.     

Ring opening reaction of gem-difluorocyclopropyl ketones with nucleophiles

Syntheses of gem-difluorocyclopropyl ketones (3a–d) and their reactions with nucleophiles are described. Ring opening reactions of 3a,c and d with a methanolate and a thiolate anion took entirely different courses of bond scission of the cyclopropane ring.     

Diethylhydrogensilyl cyclic diethylsilylene derivatives in gas chromatography/mass spectrometry of hydroxylated steroids. III-Fragmentations of 5β-pregnane-17,20,21-triol derivatives

A reaction of N,O-bis-(diethylhydrogensilyl) trifluoroacetamide with 5β-pregnane-17,20,21-triols afforded diethylhydrogensilyl (DEHS) cyclic diethylsilylene (DES) derivatives. 5β-Pregnane-3α,17α,20β,21-tetraol yielded the 3α,21-bis-DEHS-17α,20β-DES compound, whereas the structure of the major product from its 20α isomer was assigned as the 3α,17α-bis-DEHS-20α,21-DES derivative, which would undergo facile isomerization to the unstable 3α,21-bis-DEHS-17α,20α-DES structure under electron ionization. Fragmentations of these DEHS-DES compounds are discussed on the basis of isotope labelling experiments, linked scanning data, and accurate mass measurements.     

Mononuclear five-coordinate molybdenum(IV) and -(V) monosulfide complexes coordinated with dithiolene ligands: reversible redox of Mo(V)/Mo(IV) and irreversible dimerization of [MoVS]- cores to a dinuclear [MoV2(mu-S)2]2- Core

A mononuclear five-coordinate molybdenum(IV) monosulfide complex, (Et4N)2[MoS(L)2] (L = cyclohexene-1,2-dithiolate) (1), was obtained and characterized by IR, UV-vis spectroscopic methods, and X-ray crystallography. 1 was oxidized by an equivalent ferrocenium cation to give the corresponding mononuclear molybdenum(V) complex, (Et4N)[MoS(L)2] (2), which was stable for a few minutes under a lower concentration than 0.3 mM and then further dimerized to (Et4N)2[Mo(L)2]2(mu-S)2 (3).
