Type 2 diabetes (T2D) is a chronic metabolic disease, which could affect the daily life of patients and increase their risk of developing other diseases. Synthetic anti-diabetic drugs usually show severe side effects. In the last few decades, plant-derived drugs have been intensively studied, particularly because of a rapid development of the instruments used in analytical chemistry. We tested the efficacy of Gundelia tournefortii L. (GT) in increasing the translocation of glucose transporter-4 (GLUT4) to the myocyte plasma membrane (PM), as a main strategy to manage T2D. In this study, GT methanol extract was sub-fractionated into 10 samples using flash chromatography. The toxicity of the fractions on L6 muscle cells, stably expressing GLUTmyc, was evaluated using the MTT assay. The efficacy with which GLUT4 was attached to the L6 PM was evaluated at non-toxic concentrations. Fraction 6 was the most effective, as it stimulated GLUT4 translocation in the absence and presence of insulin, 3.5 and 5.2 times (at 250 μg/mL), respectively. Fraction 1 and 3 showed no significant effects on GLUT4 translocation, while other fractions increased GLUT4 translocation up to 2.0 times. Gas chromatography–mass spectrometry of silylated fractions revealed 98 distinct compounds. Among those compounds, 25 were considered anti-diabetic and glucose disposal agents. These findings suggest that GT methanol sub-fractions exert an anti-diabetic effect by modulating GLUT4 translocation in L6 muscle cells, and indicate the potential of GT extracts as novel therapeutic agents for T2D.  相似文献

The electronic structure of [TiCl4]n systems (n = 0, ?1, ?2, ?3, and ?4) has been investigated by the INDO method. Calculations show the stability of the tetrahedral structure over the square-planar one, for any value of n, in accord with experimental observation. The occupation of the 4p orbitals of the titanium atom was found to increase regularly as the dihedral angle decreases and reaches a maximum at the square-planar conformation. Energy partitioning analysis enabled much deeper understanding of bonding characteristics of the studied systems. It has been shown that the one-atom energy component may be a very sensitive measure of the stability of the central metal atom in its various oxidation states.  相似文献

Chromatographic studies on the EtOAc soluble portion of the MeOH extract of Geranium lasiopus led to the isolation of eight flavonoids (kaempferol (1), quercetin (2), quercetin 3-O-β-glucopyranoside (3), quercetin 3-O-β-galactopyranoside (4), kaempferol 3-O-α-rhamnopyranosyl-(1?→?6)-β-glucopyranoside (5), quercetin 3-O-α-rhamnopyranosyl-(1?→?6)-β-glucopyranoside (6), kaempferol 3-O-α-rhamnopyranosyl-(1?→?2)-β-glucopyranoside (7) and quercetin 3-O-α-rhamnopyranosyl-(1?→?2)-β-glucopyranoside (8)), two simple phenolic compounds (gallic acid (9) and its methyl ester (10)) and a hydrolysable tannin (pusilagin (11)). The structures of the compounds were elucidated by 1- and 2-dimensional NMR techniques ((1)H, (13)C, COSY, HMBC, HMQC) and ESI-TOF-MS spectrometry. Inhibitory effects on H(2)O(2)-induced lipid peroxidation in human red blood cells of the different extracts of G. lasiopus, as well as isolated compounds, were investigated. All tested compounds showed comparable or higher activity than that of ascorbic acid and trolox.  相似文献

Non-contact atomic force microscopy has been used to investigate the surface pore structure of Cyclopore and Anopore microfiltration membranes in air. Three Cyclopore membranes and three Anopore membranes of different pore sizes were studied. Excellent high resolution images were obtained. Analysis of the images gave quantitative information on the surface pore structure, in particular the pore size distribution. Non-contact AFM is an excellent means of obtaining such information for microfiltration membranes.  相似文献

The past few years have shown rapid growth of NMR imaging in both image quality and diagnostic usefulness. It has become apparent, as the images have been published, that both inter- and intra-group imaging of the same underlying pathology produces images which can have vastly differing appearance. This effect is mainly due to imaging techniques which use different pulse sequence types and timings thus varying the relative contribution of the protpn density, T1, and T2 properties of the tissues. In this paper we investigate the contrast manipulation effects and methods for SNR optimization for the saturation recovery, inversion recovery, spin echo, and inversion recovery spin echo pulse sequences when applied to three clinically relevant imaging tasks.  相似文献

The complexes of orotic acid with Co(II), Ni(II), Fe(III), Cu(II), and Cd(II) were prepared and their stoichiometry were determined by elemental analysis. Co(II) and Ni(II) give complexes with orotic acid of 1:1 ratio whereas that of the remaining transition metals give complexes of 1:2 ratio. The stereochemistry of the studied metal complexes has been established by analyses of their electronic spectra and magnetic susceptibilities. The mode of bonding in the studied series of metal complexes was established via, analysis of their infrared spectra. The present analysis leads to the conclusion that all metal ions studied coordinate to orotic acid via N(1) and the adjacent carboxylate group. Thermal decomposition studies of orotic acid complexes have been carried out as to understand the status of water molecules present in these complexes as well as to know their general decomposition pattern. Theoretical investigation of the electronic structure of the studied metal complexes has been carried out. MO computations at the HF-level were performed. Charge density distribution, extent of distortion from regular geometry, dipole moment, and orientation were computed and discussed.  相似文献

Computer simulations of colloidal suspensions are discussed. The simulations are based on the Langevin equations, pairwise interaction between colloidal particles and take into account Brownian, hydrodynamic and colloidal forces. Comparison of two models, one taking into account inertial term in Langevin equation and another based on diffusional approximation proposed in [D.L. Ermak, J.A. McCammon, J. Chem. Phys. 69 (1978) 1352], has shown that both models enable the prediction of the correct values of the diffusion coefficient and residence time of particle in a doublet and are therefore suitable to study the dynamics of formation and breakage of clusters in colloidal suspensions. It is shown that the appropriate selection of the time step and taking into account inertia of particles provides also the correct value of the average kinetic energy of each particle during the simulations, what allows to use the model based on full Langevin equations as a reference model to verify the validity of the numerical scheme for simulation using diffusion approximation.  相似文献

A nanocomposite (Ho2O3NPs/BNT) was synthesized by decorating holmium(III)oxide nanoparticles (H2O3NPs) on bentonite (BNT) through a realizable sonochemical approach for the electrochemical detection of haloperidol (Hlp). A glassy carbon electrode was modified with this nanocomposite. The Ho2O3NPs/BNT modified electrode outperformed bare and other modified electrodes in terms of electrochemical performance for Hlp detection in a pH 8.0 phosphate buffer. The proposed electrochemical platform showed a wide linear range (0.01 μM–24 μM), low detection limit (2.4 nM), and high sensitivity by square wave voltammetry. In addition, the proposed electrochemical sensor met the clinical criteria in terms of stability, selectivity, and repeatability.  相似文献

Two-dimensional fuzzy entropy, dispersion entropy, and their multiscale extensions (MFuzzyEn2D and MDispEn2D, respectively) have shown promising results for image classifications. However, these results rely on the selection of key parameters that may largely influence the entropy values obtained. Yet, the optimal choice for these parameters has not been studied thoroughly. We propose a study on the impact of these parameters in image classification. For this purpose, the entropy-based algorithms are applied to a variety of images from different datasets, each containing multiple image classes. Several parameter combinations are used to obtain the entropy values. These entropy values are then applied to a range of machine learning classifiers and the algorithm parameters are analyzed based on the classification results. By using specific parameters, we show that both MFuzzyEn2D and MDispEn2D approach state-of-the-art in terms of image classification for multiple image types. They lead to an average maximum accuracy of more than 95% for all the datasets tested. Moreover, MFuzzyEn2D results in a better classification performance than that extracted by MDispEn2D as a majority. Furthermore, the choice of classifier does not have a significant impact on the classification of the extracted features by both entropy algorithms. The results open new perspectives for these entropy-based measures in textural analysis.  相似文献

A 2-D computer simulation of a coaxial plasma device depending on the conservation equations of electrons, ions and excited atoms together with the Poisson equation for a plasma gun is carried out. Some characteristics of the plasma focus device (PF) such as critical wave numbers a c and voltages U c in the cases of various pressures Pare estimated in order to satisfy the necessary conditions of traveling particle densities (i.e. plasma patterns) via a linear analysis. Oscillatory solutions are characterized by a nonzero imaginary part of the growth rate (σ) for all cases. The model also predicts the minimal voltage ranges of the system for certain pressure intervals.   相似文献
