A confirmatory method is described for the determination of the illegal antibiotic chloramphenicol using a specifically developed molecularly imprinted polymer (MIP) as the sample clean-up technique. The newly developed MIP was produced using an analogue to chloramphenicol as the template molecule. Using an analogue of the analyte as the template avoids a major traditional drawback associated with MIPs of residual template leeching or bleeding. The MIP described was used as a solid-phase extraction phase for the extraction of chloramphenicol from various sample matrices including honey, urine, milk and plasma. A full analytical method with quantification by LC-MS/MS is described. The method was fully validated according to the European Union (EU) criteria for the analysis of veterinary drug residues.  相似文献

Alterations in the polyamine and amino acid (tyrosine) moieties of philanthotoxin-343 (PhTX-343) were investigated for their effects on the antagonism of nicotinic acetylcholine receptors (nAChRs) isolated from the locust (Schistocerca gregaria) mushroom body. Through whole-cell patch-clamp recordings, the philanthotoxin analogues in this study were shown to cause inhibition of the inward current when co-applied with acetylcholine (ACh). PhTX-343 (IC50 = 0.80 μM at −75 mV) antagonised locust nAChRs in a use-dependent manner, suggesting that it acts as an open-channel blocker. The analogue in which both the secondary amine functionalities were replaced with methylene groups (i.e., PhTX-12) was ~6-fold more potent (IC50 (half-maximal inhibitory concentration) = 0.13 μM at −75 mV) than PhTX-343. The analogue containing cyclohexylalanine as a substitute for the tyrosine moiety of PhTX-343 (i.e., Cha-PhTX-343) was also more potent (IC50 = 0.44 μM at −75 mV). A combination of both alterations to PhTX-343 generated the most potent analogue, i.e., Cha-PhTX-12 (IC50 = 1.71 nM at −75 mV). Modulation by PhTX-343 and Cha-PhTX-343 fell into two distinct groups, indicating the presence of two pharmacologically distinct nAChR groups in the locust mushroom body. In the first group, all concentrations of PhTX-343 and Cha-PhTX-343 inhibited responses to ACh. In the second group, application of PhTX-343 or Cha-PhTX-343 at concentrations ≤100 nM caused potentiation, while concentrations ≥ 1 μM inhibited responses to ACh. Cha-PhTX-12 may have potential to be developed into insecticidal compounds with a novel mode of action.  相似文献

In the beginning of the 1980’s Large Eddy BreakUp (LEBU) devices, thin plates or airfoils mounted in the outer part of turbulent boundary layers, were shown to be able to change the turbulent structure and intermittency as well as reduce turbulent skin friction. In some wind-tunnel studies it was also claimed that a net drag reduction was obtained, i.e. the reduction in skin-friction drag was larger than the drag on the devices. However, towing-tank experiments with a flat plate at high Reynolds numbers as well as with an axisymmetric body showed no net reduction, but instead an increase in total drag. Recent large-eddy simulations have explored the effect of LEBUs on the turbulent boundary layer and evaluations of the total drag show similar results as in the towing tank experiments. Despite these negative results in terms of net drag reduction, LEBUs manipulate the boundary layer in an interesting way which explains why they still attract some interest. The reason for the positive results in the wind-tunnel studies as compared to drag measurements are discussed here, although no definite answer for the differences can be given.  相似文献

This study thoroughly examines various higher-order three and four-node beam elements for use in the absolute nodal coordinate formulation (ANCF). The paper carefully investigates which potential benefits and drawbacks the utilization of higher-order ANCF beam elements without in-slope vectors has in the case of the usage of full three-dimensional elasticity. When the elastic forces for shear-deformable ANCF beam elements are calculated using full three-dimensional elasticity—especially in the form of the St. Venant–Kirchhoff material law—Poisson locking severely deteriorates the accuracy of the numeric results. As shown in this paper, an existing approach to preventing this locking phenomenon for three-node beam elements can still produce unsatisfying results in load cases involving bidirectional bending. The results of this study show that enriching the polynomial basis used to approximate the beam kinematics provides a natural solution to this issue. As will be seen, these findings for three-node elements can also be extended to four-node elements. When using a sufficient approximation order in transverse directions, satisfying accuracy can be achieved both in conventional one-dimensional bending and in the above-mentioned bidirectional load case.  相似文献

We describe an one-step 11-min magnetic permeability based two-site immunoassay for C-reactive protein (CRP) utilizing polyclonal anti-canine CRP antibody conjugated dextran iron oxide nanoparticles (79 nm) as superparamagnetic labels and polyclonal anti-canine CRP conjugated silica microparticles (15 to 40 μm) as carriers. An inductance based magnetic permeability reader was used to detect the target analyte, CRP, in 10 μL whole blood samples, by measuring the magnetic permeability increase of the silica microparticle sediment due to immuno complex superparamagnetic nanoparticles. Measurements on standards showed a linear response between 0 and 17.5 mg/L CRP. Measurements performed on 16 whole blood samples from mixed breeds showed good correlation with a commercially available ELISA assay.  相似文献

The peptide main chains are connected by hydrogen bonds in two‐dimensional layers. The peptide mol­ecules in each layer are related only by translation, and generate a very rare pattern. Chemical yield determination can be a limiting source to the combined standard uncertainty in measurements of radionuclides. Therefore, the combined standard uncertainties in yield determination in the measurement of radioactive strontium using two methods were evaluated. The two methods compared were the measurement of stable strontium by atomic absorption spectrometry (AAS) and the gamma spectrometric measurement of 85Sr. The evaluation showed that using gamma spectrometry for yield determination can reduce the combined standard uncertainty by a factor of three compared to AAS. The expanded uncertainties of AAS and gamma spectrometry were calculated to be 10% and 3.3% (k = 2), respectively.  相似文献
