Human neural stem cell growth and differentiation in a gradient-generating microfluidic device 总被引:9,自引:0,他引:9
This paper describes a gradient-generating microfluidic platform for optimizing proliferation and differentiation of neural stem cells (NSCs) in culture. Microfluidic technology has great potential to improve stem cell (SC) cultures, whose promise in cell-based therapies is limited by the inability to precisely control their behavior in culture. Compared to traditional culture tools, microfluidic platforms should provide much greater control over cell microenvironment and rapid optimization of media composition using relatively small numbers of cells. Our platform exposes cells to a concentration gradient of growth factors under continuous flow, thus minimizing autocrine and paracrine signaling. Human NSCs (hNSCs) from the developing cerebral cortex were cultured for more than 1 week in the microfluidic device while constantly exposed to a continuous gradient of a growth factor (GF) mixture containing epidermal growth factor (EGF), fibroblast growth factor 2 (FGF2) and platelet-derived growth factor (PDGF). Proliferation and differentiation of NSCs into astrocytes were monitored by time-lapse microscopy and immunocytochemistry. The NSCs remained healthy throughout the entire culture period, and importantly, proliferated and differentiated in a graded and proportional fashion that varied directly with GF concentration. These concentration-dependent cellular responses were quantitatively similar to those measured in control chambers built into the device and in parallel cultures using traditional 6-well plates. Summary: N,N‐Diphenylacrylamide was polymerized in a living fashion with triisobutylaluminum in THF at 0 °C. The polymerization results showed an increase of molecular weight proportional to the amount of monomer consumed and a first‐order kinetics at −78 °C. The intermediates obtained with excess initiator at −78 °C revealed that the polymerization was initiated through 1,4‐addition of hydride from a triisobutyl group in the triisobutylaluminum and then proceeded through aluminum‐oxygen bond interchange.

We describe our full report of the catalytic asymmetric addition of simple and functionalized dialkylzinc reagents to a broad range of saturated ketones and enones. The functionalized organozinc reagents contain esters, silyl ethers, alkyl chlorides, and alkyl bromides. In general, the resulting tertiary alcohol products are isolated with high ee's. With some substrates, yields are low as a result of the formation of aldol byproducts. Most substrates undergo additions with good yields reaching as high as 91%. 相似文献

Doped mesoporous carbons comprising nitrogen, boron, and phosphorus (N, B, and P, respectively) were prepared as non-Pt catalysts for oxygen reduction reaction (ORR) in an acidic solution. The N-doped carbons were varied to increase their catalytic activity through by additionally doping of B and P. All the mesoporous carbons were synthesized by carbonizing polyaniline at 900 °C for the N species, while the B and P species were inserted into the carbon structure at the carbon growth step. The linear sweep voltammogram recorded in the acidic solution showed that the ORR activity of the N-doped carbon catalysts increased significantly after the addition of B. An approximately 19 % increase in the pyridinic N content at the carbon surface was observed, along with B-N-C moieties with a binding energy of 399.5 eV. The non-precious metal ORR catalysts were prepared via pyrolysis, with the insertion of an additional transition metal (iron, Fe). The deconvoluted X-ray photoelectron spectroscopy (XPS) results showed that the Fe-N peak was generated after the pyrolysis. The peak intensity of the quaternary N also increased compared with the pyridic and pyrrolic N, which indicates that Fe serves to catalyze the modification of N species. The numerical examinations showed that N- and B-doped mesoporous carbon (NBC) 1.5 % Fe had the highest limited current (4.94 mA/cm2), with the B-doped carbon still the most active mesoporous carbon catalyst for ORR. As a result, it can be said that Fe positively contributes to the formation of graphitic N, which is known to be an active site for ORR. The cyclic voltammetry results showed that the peak area of the NBC 1.5 % Fe catalyst was larger than that of the N-doped mesoporous carbon (NC) 1.5 % Fe catalyst. It was concluded that B doping enhances the ORR activity and the stability of carbon materials even after 1000 cycles under acidic conditions. 相似文献

Axin is a negative regulator of the Wnt/beta-catenin pathway and is involved in the regulation of axis formation and proliferation. Involvement of Axin in the regulation of other signaling pathways is poorly understood. In this study, we investigated the involvement of Akt in growth regulation by Axin in L929 fibroblasts stimulated by EGF. Akt activity was increased by EGF treatment and Ras activation, respectively. Both the EGF- and Ras-induced Akt activations were abolished by Axin induction, as revealed by both Western blot and immunocytochemical analyses. The proliferation and Akt activation induced by EGF were decreased by Axin induction, and the effects of EGF were abolished by treatment of an Akt-specific inhibitor. Therefore, Axin inhibits EGF-induced proliferation of L929 fibroblasts by blocking Akt activation. 相似文献

A novel dielectrophoresis switching with vertical electrodes in the sidewall of microchannels for multiplexed switching of objects has been designed, fabricated and tested. With appropriate electrode design, lateral DEP force can be generated so that one can dynamically position particulates along the width of the channel. A set of interdigitated electrodes in the sidewall of the microchannels is used for the generation of non-uniform electrical fields to generate negative DEP forces that repel beads/cells from the sidewalls. A countering DEP force is generated from another set of electrodes patterned on the opposing sidewall. These lateral negative DEP forces can be adjusted by the voltage and frequency applied. By manipulating the coupled DEP forces, the particles flowing through the microchannel can be positioned at different equilibrium points along the width direction and continue to flow into different outlet channels. Experimental results for switching biological cells and polystyrene microbeads to multiple outlets (up to 5) have been achieved. This novel particle switching technique can be integrated with other particle detection components to enable microfluidic flow cytometry systems. 相似文献
