A strategy for clustering of native lipid membranes is presented. It relies on the formation of complexes between hydrophobic chelators embedded within the lipid bilayer and metal cations in the aqueous phase, capable of binding two (or more) chelators simultaneously Fig. 1. We used this approach with purple membranes containing the light driven proton pump protein bacteriorhodopsin (bR) and showed that patches of purple membranes cluster into mm sized aggregates and that these are stable for months when incubated at 19 °C in the dark. The strategy may be general since four different hydrophobic chelators (1,10-phenanthroline, bathophenanthroline, Phen-C10, and 8-hydroxyquinoline) and various divalent cations (Ni2+, Zn2+, Cd2+, Mn2+, and Cu2+) induced formation of membrane clusters. Moreover, the absolute requirement for a hydrophobic chelator and the appropriate metal cations was demonstrated with light and atomic force microscopy (AFM); the presence of the metal does not appear to affect the functional state of the protein. The potential utility of the approach as an alternative to assembled lipid bilayers is suggested. 相似文献

Synthesis of grafting silane on a hydro soluble cellulose ether (HPMC) was described. In alkaline medium, this derivate is under gel form. With a decrease of the pH, a self-hardening occurs due to the silanol condensation. For potential biomedical use, we described the silated-HPMC synthesis, the gel behavior after steam sterilization and the parameters of the silanol condensation i.e. pH, silane percentage and temperature. Minimum kinetic of the condensation was observed for pH between 5.5 and 6.5. So temperature catalyzed the reaction and the self-hardening speed was increased by silane percentage. 相似文献

We investigated how the duration of short laser pulses evolves in a dispersive material, using rms widths and a propagation law based on a pulse quality factor. Experiments were carried out with femtosecond pulses (10 to 25fs at the temporal waist) propagating in bulk fused silica. Excellent agreement was found between theory and experiment. This approach does not require complete characterization of laser pulses and eliminates the need for any assumption regarding the interpretation of autocorrelation traces. Despite the use of high resolution magic angle spinning NMR, the NMR linewidth of anchored molecules on the commonly used Merrifield solid phase resins remains larger than that of the corresponding molecules in solution. We investigate the different mechanisms that might be at the origin of this line broadening.
