Exhaled breath condensate (EBC) collection is a simple and non-invasive method to sample airway secretions, but analysis is limited by extensive and variable dilution of airway secretions within the condensate. To overcome this limitation, we developed a sensitive and specific liquid chromatography/tandem mass spectrometry (LC/MS/MS) method to simultaneously detect adenyl purines as biomarkers of inflammation and urea as a dilution marker in EBC. Separation prior to mass spectrometry was achieved using a C18 column with methanol and formic acid as the mobile phase, and characteristic precursor to product ion transitions of m/z 268 to 136 (for adenosine), m/z 348 to 136 (for AMP), and m/z 61 to 44 (for urea) were monitored for quantification. To correct for matrix effects, isotopically labeled adenosine, AMP, and urea were used as internal standards. Using these methods, we detected urea and the adenyl purines adenosine and AMP in EBC from seven subjects with cystic fibrosis (CF) and seven healthy controls and found that the AMP/urea ratio was elevated in the CF samples. These results demonstrate that mass spectrometry can be used successfully in EBC analysis to simultaneously detect a biomarker for airway inflammation and control for variable dilution.  相似文献

The usefulness of neural networks for the classification of signal-time curves from dynamic MR mammography was recently demonstrated by our group. The multi-layer perceptron under study consists of 28 input, 4 hidden, and 3 output nodes, and was trained to classify signal-time curves into three tissue classes: "carcinoma," "benign lesion," and "parenchyma." Extending this approach, it was the aim of the present study to evaluate the performance of the developed network in the segmentation of dynamic MR mammographic images in comparison to a pixel-by-pixel two-compartment pharmacokinetic analysis. The population investigated in this pilot study comprised 15 women with suspicious lesions in the breast, which were confirmed histologically after the MR examination. The neural network classified the same areas as malignant as those which were marked as being highly suspicious by the pharmacokinetic mapping approach but with the advantage that no a priori knowledge on tissue microcirculation was needed, that computation proved to be much faster, and that it yielded a unique classification into just three tissue classes.  相似文献

Chitosan (CS) and poly(acrylic acid) (PAA) were crosslinked by an ionic gelation method to form super absorbent polymers (SAPs). CS and PAA form amide bonds between the amino and carboxyl groups. The CS-PAA copolymers were synthetically engineered by varying the feed ratios of the prepolymer units. The copolymer materials possess tunable sorption and mucoadhesive properties with a backbone structure resembling proteinaceous materials. The sorption properties of the copolymers toward methylene blue (MB) in aqueous solution were studied using UV-Vis spectrophotometry at ambient pH and 295 K. The copolymers showed markedly varied interactions with MB, from physisorption- to chemisorption-like behavior, in accordance with their composition, surface area, and pore structure characteristics. The sorption isotherms were evaluated with the Sips model to provide estimates of the sorption properties. The sorbent surface area (271 and 943 m2/g) and the sorption capacity (Qm = 1.03 and 3.59 mmol/g) were estimated for the CS-PAA copolymer/MB systems in aqueous solution.  相似文献

The improved synthesis of the antitumor compound (+)‐tedanolide is described through an aldol coupling of bis‐ketone 7 . This modification shortens the synthetic steps in the endgame and provides rapid access to this biologically important natural product. Additionally, it serves as a probe in order to unravel the conformational effects that impede or enable its successful synthesis. Having this way access to des‐epoxy‐tedanolide, its biological characterization surprisingly unravelled the mode of action to resemble candidaspongiolide rather than deoxytedanolide.  相似文献

Bacteria have developed cell-to-cell communication mechanisms, termed quorum sensing (QS), that regulate bacterial gene expression in a cell population-dependent manner. Autoinducer-2 (AI-2), a class of QS signaling molecules derived from (4S)-4,5-dihydroxy-2,3-pentanedione (DPD), has been identified in both Gram-negative and Gram-positive bacteria. Despite considerable interest in the AI-2 QS system, the biomolecular communication used by distinct bacterial species still remains shrouded. Herein, we report the synthesis and evaluation of a new class of DPD analogues, C4-alkoxy-5-hydroxy-2,3-pentanediones, termed C4-alkoxy-HPDs. Remarkably, two of the analogues were more potent QS agonists than the natural ligand, DPD, in Vibrio harveyi. The findings presented extend insights into ligand-receptor recognition/signaling in the AI-2 mediated QS system.  相似文献

The first total synthesis of the (+)-tedanolide is described. Pivotal steps are the Felkin-selective aldol coupling between C12 and C13 and an efficient Mitsunobu macrolactonization. Selective protecting group transformations and subsequent oxidations generate the macrocyclic triketone. In the endgame of the synthesis, four TBS groups are removed in one reaction and a chemo- and stereoselective final step epoxidation generates (+)-tedanolide.  相似文献
