[reaction: see text] The organometallic intermediate obtained from halogen-metal exchanges of 4-iodo-6-phenylthieno[2,3-d]pyrimidine under Barbier-type conditions was reacted with aldehydes to form the corresponding alcohols in moderate yields. The reaction involving an organolithium intermediate proceeded only at low temperature, whereas the reaction involving a magnesium ate intermediate also proceeded at room temperature. A crystal structure confirms that the expected constitutional alcohol isomer is formed, where no migration has taken place. The conditions were also suitable for 9-benzyl-6-iodopurine. 相似文献

Photoinitiated radical carbonylation with [(11)C]carbon monoxide at low concentration was employed in syntheses of carbonyl-(11)C-labeled amides using alkyl iodides and amines as precursors. Eleven (11)C-amides were synthesized in up to 74% decay-corrected radiochemical yields with reaction times of 400 s and with up to 95% conversion of carbon monoxide. Starting with 26.3 GBq of [(11)C]carbon monoxide, 10.6 GBq of 1-cyclohexane [(11)C]carbonyl-4-phenyl-piperazine (15) was obtained within 35 min from the end of bombardment (33 microA) and with a specific radioactivity of 192 GBq/micromol at the same time point. The influence of solvents was investigated. The described procedure extends the range of accessible labeling methods. The method may also be useful for preparation of (13)C- and (14)C-substituted compounds. 相似文献

The spectroscopic properties for seven different commercial resins used in solid-phase synthesis were investigated with (19)F NMR spectroscopy. A fluorine-labeled dipeptide was synthesized on each resin, and the resolution of the (19)F resonances in CDCl(3), DMSO-d(6), benzene-d(6) and CD(3)OD were measured with a conventional NMR spectrometer, i.e. without using magic angle spinning. In general, resins containing poly(ethylene glycol) chains (ArgoGel, TentaGel and PEGA) were found to be favorable for the (19)F NMR spectral quality. Three serine containing tri-, penta-, and heptapeptides were then prepared on an ArgoGel resin functionalized with a fluorine-labeled linker. The resin bound peptides were glycosylated utilizing a thiogalactoside glycosyl donor carrying fluorine-labeled protective groups. Monitoring of the glycosylations with gel-phase (19)F NMR spectroscopy allowed each glycopeptide to be formed in similar 80% yield, using a minimal amount of glycosyl donor (3 x 2 equivalents). Electron capture dissociation (ECD) of polypeptide cations was obtained with pencil and hollow electron beams for both sidekick and gas-assisted dynamic ion trapping (GADT) using Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS) with an electrostatic ion transfer line. Increasing the number of trapped ions by multiple ICR trap loads using GADT improved the ECD sensitivity in comparison with sidekick ion trapping and ECD efficiency in comparison with single ion trap load by GADT. Furthermore, enhanced sensitivity made it possible to observe ECD in a wide range of electron energies (0-50 eV). The degree, rate and fragmentation characteristics of ECD FTICR-MS were investigated as functions of electron energy, electron irradiation time, electron flux and ion trapping parameters for this broad energy range. The results obtained show that the rate of ECD is higher for more energetic (>1 eV) electrons. Long electron irradiation time with energetic electrons reduces average fragment ion mass and decreases efficiency of formation of c- and z-type ions. The obtained dependencies suggest that the average fragment ion mass and the ECD efficiency are functions of the total fluence of the electron beam (electron energy multiplied by irradiation time). The measured electron energy distributions in low-energy ECD and hot ECD regimes are about 1 eV at full width half maximum in employed experimental configurations. 相似文献

2,2-Dibutyl-2-stanna-1,3-dioxepane 1 (or 1,3-dioxepene 2) were prepared from 1,4-butane (or 1,4-butene) diol and dibutyltin dimethoxide. They were polycondensed at 80°C in n-heptane with adipoyl-, suberoyl, sabacoyl chloride and with decane-1,10-dicarbonyl chloride. In the case of suberoyl chloride and 2,2-dibutyl-2-stanna-1,3-dioxepane reaction time, temperature and stoichiometry were varied to optimize both the molecular weight and the fraction of cyclic polyesters. With a slight excess of the dicarboxylic acid chlorides, only macrocyclic polyesters were obtained in all cases. We explored the application of electron detachment dissociation (EDD) and infrared multiphoton dissociation (IRMPD) tandem mass spectrometry to fluorescently labeled sialylated oligosaccharides. Standard sialylated oligosaccharides and a sialylated N-linked glycan released from human transferrin were investigated. EDD yielded extensive glycosidic cleavages and cross-ring cleavages in all cases studied, consistently providing complementary structural information compared with infrared multiphoton dissociation. Neutral losses and satellite ions such as C-2H ions were also observed following EDD. In addition, we examined the influence of different fluorescent labels. The acidic label 2-aminobenzoic acid (2-AA) enhanced signal abundance in negative-ion mode. However, few cross-ring fragments were observed for 2-AA-labeled oligosaccharides. The neutral label 2-aminobenzamide (2-AB) resulted in more cross-ring cleavages compared with 2-AA-labeled species, but not as extensive fragmentation as for native oligosaccharides, likely resulting from altered negative charge locations from introduction of the fluorescent tag. 相似文献

A candidate certified reference material (CRM) for multiple shellfish toxins (domoic acid, okadaic acid and dinophysistoxins,
pectenotoxins, yessotoxin, azaspiracids and spirolides) has been prepared as a freeze-dried powder from mussel tissues (Mytilus edulis). Along with the certified values, the most important characteristics for a reference material to be fit-for-purpose are
homogeneity and stability. Acceptable between-bottle homogeneity was found for this CRM. Within-bottle homogeneity was assessed
using domoic acid, and it was shown that repeated subsampling of the CRM can be performed precisely down to 0.35 g. Both short-
and long-term stability studies carried out under isochronous conditions demonstrated excellent stability of the various toxins
present in the material. While degradation of some analytes was observed at +60°C in short-term studies, it was determined
that shipping at ambient temperature is adequate. No instability was detected in long-term stability studies, and it was shown
that the material can be held at +18°C safely for up to 1 year. To guarantee stability of the CRM over its lifetime the stock
will be maintained at −20°C. The results of the homogeneity and stability testing show that CRM–FDMT1 is appropriate for its
intended use in quality assurance and quality control of shellfish toxin analysis methods. 相似文献

The deprotected nucleoside4 undergoes aDimroth rearrangement on reflux for 24 h in water to furnish the 4-arylamino nucleoside7.

The creep behavior of nanocellulose films and aerogels are studied in a dynamic moisture environment, which is crucial to
their performance in packaging applications. For these materials, the creep rate under cyclic humidity conditions exceeds
any constant humidity creep rate within the cycling range, a phenomenon known as mechanosorptive creep. By varying the sample
thickness and relative humidity ramp rate, it is shown that mechanosorptive creep is not significantly affected by the through-thickness
moisture gradient. It is also shown that cellulose nanofibril aerogels with high porosity display the same accelerated creep
as films. Microstructures larger than the fibril diameter thus appear to be of secondary importance to mechanosorptive creep
in nanocellulose materials, suggesting that the governing mechanism is found between molecular scales and the length-scales
