The negative impacts on the ecosystem of antibiotic residues in the environment have become a global concern. However, little is known about the transformation mechanism of antibiotics by manganese peroxidase (MnP) from microorganisms. This work investigated the transformation characteristics, the antibacterial activity of byproducts, and the degradation mechanism of tetracycline (TC) by purified MnP from Phanerochaete chrysosporium. The results show that nitrogen-limited and high level of Mn2+ medium could obtain favorable MnP activity and inhibit the expression of lignin peroxidase by Phanerochaete chrysosporium. The purified MnP could transform 80% tetracycline in 3 h, and the threshold of reaction activator (H2O2) was about 0.045 mmol L−1. After the 3rd cyclic run, the transformation rate was almost identical at the low initial concentration of TC (77.05–88.47%), while it decreased when the initial concentration was higher (49.36–60.00%). The antimicrobial potency of the TC transformation products by MnP decreased throughout reaction time. We identified seven possible degradation products and then proposed a potential TC transformation pathway, which included demethylation, oxidation of the dimethyl amino, decarbonylation, hydroxylation, and oxidative dehydrogenation. These findings provide a novel comprehension of the role of MnP on the fate of antibiotics in nature and may develop a potential technology for tetracycline removal.  相似文献

Background: Danshen (DS), the dry root of Salvia miltiorrhiza Bge., has been used in traditional Chinese medicine (TCM) for many years to promote blood circulation and to inhibit thrombosis. However, the active ingredients responsible for the anti-thrombotic effect and the underlying mechanisms are yet to be fully elucidated. Methods: Molecular docking was used to predict the active ingredients in DS and their potential targets by calculating the scores of docking between DS ingredients and thrombosis-related proteins. Then, a chemical-induced zebrafish thrombosis model was applied to confirm their anti-thrombotic effects. Result: The molecular docking results indicated that compared to the control ligand, higher docking scores were observed for several compounds in DS, among which salvianolic acid B (SAB), lithospermic acid (LA), rosmarinic acid (MA), and luteolin-7-O-β-d-glucoside (LG) could attenuate zebrafish caudal vein thrombosis and recover the decrease in heart red blood cells (RBCs) in a dose-dependent manner. Conclusions: Our study showed that it is possible to screen the potential active components in natural products by combining the molecular docking method and zebrafish in vivo model.  相似文献

It is well known that oxidative stress induces muscle atrophy, which decreases with the activation of Nrf2/HO-1. Fermented oyster extracts (FO), rich in γ-aminobutyric acid (GABA) and lactate, have shown antioxidative effects. We evaluated whether FO decreased oxidative stress by upregulating Nrf2/HO-1 and whether it decreased NF-κB, leading to decreased IL-6 and TNF-α. Decreased oxidative stress led to the downregulation of Cbl-b ubiquitin ligase, which increased IGF-1 and decreased FoxO3, atrogin1, and Murf1, and eventually decreased muscle atrophy in dexamethasone (Dexa)-induced muscle atrophy animal model. For four weeks, mice were orally administered with FO, GABA, lactate, or GABA+Lactate, and then Dexa was subcutaneously injected for ten days. During Dexa injection period, FO, GABA, lactate, or GABA+Lactate were also administered, and grip strength test and muscle harvesting were performed on the day of the last Dexa injection. We compared the attenuation effect of FO with GABA, lactate, and GABA+lactate treatment. Nrf2 and HO-1 expressions were increased by Dexa but decreased by FO; SOD activity and glutathione levels were decreased by Dexa but increased by FO; NADPH oxidase activity was increased by Dexa but decreased by FO; NF-κB, IL-6, and TNF-α activities were increased by Dexa were decreased by FO; Cbl-b expression was increased by Dexa but restored by FO; IGF-1 expression was decreased by Dexa but increased by FO; FoxO3, Atrogin-1, and MuRF1 expressions were increased by Dexa but decreased by FO. The gastrocnemius thickness and weight were decreased by Dexa but increased by FO. The cross-sectional area of muscle fiber and grip strength were decreased by Dexa but increased by FO. In conclusion, FO decreased Dexa-induced oxidative stress through the upregulation of Nrf2/HO-1. Decreased oxidative stress led to decreased Cbl-b, FoxO3, atrogin1, and MuRF1, which attenuated muscle atrophy.  相似文献

Biomimetic electrochemical sensors are very promising not only due to their lower expense and longer stability than conventional enzymatic ones, but they also often suffer from simultaneously achieving high sensitivity and good selectivity. Here we present a well-defined Au@Co3O4/CeO2 yolk-shell nanostructure (YSN) that is first synthesized and exploited as highly efficient electrocatalysts for hydrogen peroxide (H2O2) detection. The introduced CeO2 in Co3O4 matrix greatly facilitates the migration of lattice oxygen, which increases the concentration of surface oxygen vacancies (Oa), remarkably enhancing the adsorption ability of H2O2 and promoting the decomposition of H2O2 for faster electron transfer than pristine Au@Co3O4 core-shell nanostructure (CSN). The abundant Oa of Au@Co3O4/CeO2 YSN is confirmed by X-ray photoelectron spectroscopy (XPS) and electron paramagnetic resonance (EPR). The as-prepared biomimetic sensor delivers a wide dynamic range (5.0 nM to 5.4 μM), a low limit of detection (LOD) (2.74 nM), and a high sensitivity (35.67 μA μM−1 cm−2), paving a new way to construct an ultrasensitive and selective enzyme-free biomimetic electrochemical sensor. Furthermore, the sensor is used to real-time monitor H2O2 released from human cervical cancer cells (HeLa) and human umbilical vein endothelial cells (HUVEC), demonstrating its great potential in practical applications.  相似文献

Lithium-sulfur batteries (LSBs) with high energy density and low cost have been recognized as one of the most promising next-generation energy storage systems. Although it has taken decades of development, the practical application of LSBs has been hindered by several inherent obstacles, particularly the severe shuttle effect and sluggish reaction kinetics in the sulfur cathode. Various strategies have been proposed to address these problems via rational design of electrode materials and configurations. Freestanding sulfur cathode could be a promising strategy to improve the sulfur mass loading at the cathode level and energy density of LSBs. This minireview will briefly summary the recent advances in freestanding cathodes for LSBs. The advantages and disadvantages of various freestanding cathodes are discussed and the prospects for the development of flexible cathodes are envisioned.  相似文献

Understanding the physicochemical properties and heterogeneous processes of aerosols is key not only to elucidate the impacts of aerosols on the atmosphere and humans but also to exploit their further applications, especially for a healthier environment. Experiments that allow for spatially control of single aerosol particles and investigations on the fundamental properties and heterogeneous chemistry at the single-particle level have flourished during the last few decades, and significant breakthroughs in recent years promise better control and novel applications aimed at resolving key issues in aerosol science. Here we propose graphene oxide (GO) aerosols as prototype aerosols containing polycyclic aromatic hydrocarbons, and GO can behave as two-dimensional surfactants which could modify the interfacial properties of aerosols. We describe the techniques of trapping single particles and furthermore the current status of the optical spectroscopy and chemistry of GO. The current applications of these single-particle trapping techniques are summarized and interesting future applications of GO aerosols are discussed.  相似文献

The effects of surface treatment of a carbon fiber (CF) by supersonic atmosphere plasma spraying (SAPS) on the interfacial adhesion behavior and morphology of polypropylene/polystyrene (PP/PS) matrix blends filled CF composites were investigated. Effects of surface treated a commercial CF on mechanical properties are studied. Contact angle was measured to examine the changes in wettability of the CF. The chemical and morphological changes were characterized by using X‐ray photoelectron spectroscopy and scanning electron microscopy. PP/PS/CF composites were fabricated with and without SAPS treatment, and their interlaminar fracture toughnesses were compared. The results showed that the interlaminar shear strength of composites has been greatly improved filled SAPS modification CF. The water contact angle of resin sample decreased 50% after addition of SAPS surface‐treated CF. Scanning electron microscopy results on the fractured surface exhibited PP/PS blends adhered well around the CFs of the SAPS‐treated specimen compared with that of the untreated specimen. This attributed to the CF interlock, and it improves the wetting between fibers and resins. Copyright © 2017 John Wiley & Sons, Ltd.  相似文献
