Fructosamine-6-kinases (FN6Ks) that catalyze phosphorylation of glycated amino acids, i.e., fructosyl amino acids (FAs), have been shown as a potential recognition element for glycated protein detection. However, there are only two available FN6Ks: those from Escherichia coli which is specific for ε-fructosyl lysine (ε-FK) and Bacillus subtilis which recognizes both ε-FK and α-FA as substrates. In this study, we characterized an FN6K homologue isolated from Arthrobacter, some of whose species are reported to assimilate FA. The BLAST searches of Arthrobacter genomic database, using the bacterial FN6K primary structure information, revealed the presence of an FN6K homologue in Arthrobacter aurescens TC1 strain. Indeed, enzymatic assays confirmed that the putative FN6K from A. aurescens is an FN6K that is specific for ε-FK, although the primary sequence alignments showed similarity of A. aurescens FN6Ks with FN6Ks from B. subtilis and E. coli at the same level. In this study, we describe for the first time the presence of FN6K in Arthrobacter spp. and ε-FK-specific degradation pathway from Gram-positive bacteria, providing important information for the development of FA-recognizing molecules as well as for the FA assimilation system in bacteria.  相似文献

UDP-galactofuranose (UDP-Galf) is a substrate for two types of enzymes, UDP-galactopyranose mutase and galactofuranosyltransferases, which are present in many pathogenic organisms but absent from mammals. In particular, these enzymes are involved in the biosynthesis of cell wall galactan, a polymer essential for the survival of the causative agent of tuberculosis, Mycobacterium tuberculosis. We describe here the synthesis of derivatives of UDP-Galf modified at C-5 and C-6 using a chemoenzymatic route. In cell-free assays, these compounds prevented the formation of mycobacterial galactan, via the production of short "dead-end" intermediates resulting from their incorporation into the growing oligosaccharide chain. Modified UDP-furanoses thus constitute novel probes for the study of the two classes of enzymes involved in mycobacterial galactan assembly, and studies with these compounds may ultimately facilitate the future development of new therapeutic agents against tuberculosis.  相似文献

The paper reports on new determinations at INRIM of the triple point temperature of pure neon isotopes 20Ne and 22Ne, obtained on samples sealed in cryogenic cells and measured with an uncertainty much lower than that of the previous determinations. The experimental technique is the same used at INRIM for recent studies on neon of natural-isotopic composition, showing an expanded uncertainty of ≈30 μK for a single cell and ≈50 μK for the comparison of sample pairs. The determinations were mainly intended to obtain a more accurate value of the temperature difference for the two pure isotopes, found to be 0.14660 K with an expanded uncertainty of 0.00007 K. The temperature values were found to be 24.5422 K for 20Ne and 24.6888 K for 22Ne on ITS-90, but each with a larger expanded uncertainty, 0.00032 K. This is mainly caused, contrarily to the difference, by each of these values being affected by the present ambiguity of the ITS-90 definition. This definition refers to neon generically to ‘natural’ composition, while its uncorrected variability heavily affects the uncertainty of the thermometer calibrations. These results for pure isotopes are compared with those recently obtained at INRIM on samples of commercial neon of natural composition and with literature data. The problems involved in the correction for the residual content of isotopic impurities and for chemical impurities are discussed.  相似文献

The response of interfacial layers to deformations in size and shape depends on their composition. The corresponding main mechanical quantities are elasticity and viscosity of dilation and shear, respectively. Hence, the interfacial rheology represents a kind of two-dimensional equivalent to the traditional bulk rheology. Due to growing interest in the quantitative understanding of foams and emulsions, more works are dedicated to studies on interfacial rheology. This overview presents the theoretical basis for traditional and recently developed experimental tools and discusses their application to different interfacial systems. While dilational rheology provides information on the composition of mixed interfacial layers, the shear rheology gives answers essentially on structures formed at an interface. The most frequently used methods at present are the oscillating drop and bubble tensiometry methods for dilational deformations and oscillating ring/bicone rheometers for shear deformations.  相似文献

The realization of a reliable receptor biosensor requires stable, long-lasting, reconstituted biomembranes able to supply a suitable biomimetic environment where the receptor can properly work after incorporation. To this end, we developed a new method for preparing stable biological membranes that couple the biomimetic properties of BLMs (bilayer lipid membranes) with the high stability of HBMs (hybrid bilayer membranes); this gives rise to an innovative assembly, named MHBLM (mixed hybrid bilayer lipid membrane). The present work deals with the characterization of biosensors achieved by embedding an ionotropic glutamate receptor (GluR) on MHBLM. Thanks to signal (transmembrane current) amplification, which is typical of natural receptors, the biosensor here produced detects glutamate at a level of nmol L(-1). The transmembrane current changes linearly vs glutamate up to 100 nmol L(-1), while the limit of detection is 1 nmol L(-1). In addition, the biosensor response can be modulated both by receptor agonists (glycine) and antagonists (Mg(2+)) as well, and by exploiting the biosensor response, the distribution of different kinds of ionotropic GluR present in the purified sample, and embedded in MHBLM, was also evaluated. Finally, one of the most important aspects of this investigation is represented by the high stability of the biomimetic system, which allows the use of biosensor under flowing conditions, where the solutions flow on both biomembrane faces.  相似文献

The phenomenology of the α- and β-relaxation processes in an amorphous and a crystallized specimen of a side-chain LC polyacrylate, based on the azobenzene mesogenic unit, was investigated by thermal and dynamic mechanical methods. The activation energy of the β relaxation of the crystallized sample is equal to that of the amorphous sample. The α-transition process of the amorphous sample was described by the WLF equation. In contrast, the α-relaxation behavior of the amorphous part of the semicrystalline sample was described by a double Arrhenius law broken at T = Tg. This peculiar behavior has been tentatively related to the decrease of the motion characteristic length of the macromolecules confined in the multidomain structure of the semicrystalline state. © 1996 John Wiley & Sons, Inc.  相似文献

The present paper deals with the speciation of selenium in potatoes (enriched or not in selenium). The study was carried out by using differential pulse cathodic stripping voltammetry (DPCSV) for quantifying selenium. Results obtained provide evidence that the selenium content in the protein fraction is rather independent from the selenium added to the plants during their growth. On the contrary, the amount of Se in the non-protein fraction (water and starch) in Se-enriched sample is significantly higher than in non-enriched one, suggesting that it is the main selenium-storing site. In this fraction the Se(VI)/Se(IV) ratio seems independent from selenium application but it may be related to the redox conditions. The accumulation of selenium in the non-protein fraction is tentatively ascribed to the “Se–starch interaction” that should be able to modulate both the Se absorption into proteins and, possibly, its toxic effect for the plant itself.  相似文献

Liquid crystalline block copolymers are new materials in which multiple molecular interactions can provide the driving force for complex phase behaviors and states of order. Block copolymers containing both liquid crystalline main-chain polyester and side-group polymethacrylate blocks were investigated. They phase separated in the liquid crystalline state and their individual mesophases coexisted. The copolymers responded very differently when either a mechanical or a magnetic field was used for alignment. In the fibers the orientations of the side-group and main-chain smectic planes with respect to the fiber axis depended critically on the block lengths and on their distinct tendencies to align, whereas under a magnetic field the mesogens aligned collectively with their long molecular axis parallel to the field, independent of the copolymer structure.  相似文献
