Background  

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by the selective loss of motor neurons (MN) in the brain stem and spinal cord. Intracellular disruptions of cytosolic and mitochondrial calcium have been associated with selective MN degeneration, but the underlying mechanisms are not well understood. The present evidence supports a hypothesis that mitochondria are a target of mutant SOD1-mediated toxicity in familial amyotrophic lateral sclerosis (fALS) and intracellular alterations of cytosolic and mitochondrial calcium might aggravate the course of this neurodegenerative disease. In this study, we used a fluorescence charged cool device (CCD) imaging system to separate and simultaneously monitor cytosolic and mitochondrial calcium concentrations in individual cells in an established cellular model of ALS.  相似文献

Specific and sensitive analysis to reveal and monitor the wide variety of chemical contaminants polluting all environment compartments, feed, and food is urgently required because of the increasing attention devoted to the environment and health protection. Our research group has been involved in monitoring the presence and distribution of agrochemicals by monitoring beehives distributed throughout the area studied. Honeybees have been used both as biosensors, because the pesticides affect their viability, and as “contaminant collectors” for all environmental pollutants. We focused our research on the development of analytical procedures able to reveal and quantify pesticides in different samples but with a special attention to the complex honeybee matrix. Specific extraction and purification procedures have been developed and some are still under optimization. The analytes of interest were determined by gas or liquid chromatographic methods and by compound-specific or group-specific immunoassays in the ELISA format, the analytical performance of which was improved by introducing luminescence detection. The range of chemiluminescent immunoassays developed was extended to include the determination of completely different pollutants, for example explosives, volatile organic compounds (including benzene, toluene, ethylbenzene, xylenes), and components of plastics, for example bisphenol A. An easier and portable format, a lateral flow immunoassay (LFIA) was added to the ELISA format to increase application flexibility in these assays. Aspects of the novelty, the specific characteristics, the analytical performance, and possible future development of the different chromatographic and immunological methods are described and discussed.

UDP-galactofuranose (UDP-Galf) is a substrate for two types of enzymes, UDP-galactopyranose mutase and galactofuranosyltransferases, which are present in many pathogenic organisms but absent from mammals. In particular, these enzymes are involved in the biosynthesis of cell wall galactan, a polymer essential for the survival of the causative agent of tuberculosis, Mycobacterium tuberculosis. We describe here the synthesis of derivatives of UDP-Galf modified at C-5 and C-6 using a chemoenzymatic route. In cell-free assays, these compounds prevented the formation of mycobacterial galactan, via the production of short "dead-end" intermediates resulting from their incorporation into the growing oligosaccharide chain. Modified UDP-furanoses thus constitute novel probes for the study of the two classes of enzymes involved in mycobacterial galactan assembly, and studies with these compounds may ultimately facilitate the future development of new therapeutic agents against tuberculosis.  相似文献

The paper reports on new determinations at INRIM of the triple point temperature of pure neon isotopes 20Ne and 22Ne, obtained on samples sealed in cryogenic cells and measured with an uncertainty much lower than that of the previous determinations. The experimental technique is the same used at INRIM for recent studies on neon of natural-isotopic composition, showing an expanded uncertainty of ≈30 μK for a single cell and ≈50 μK for the comparison of sample pairs. The determinations were mainly intended to obtain a more accurate value of the temperature difference for the two pure isotopes, found to be 0.14660 K with an expanded uncertainty of 0.00007 K. The temperature values were found to be 24.5422 K for 20Ne and 24.6888 K for 22Ne on ITS-90, but each with a larger expanded uncertainty, 0.00032 K. This is mainly caused, contrarily to the difference, by each of these values being affected by the present ambiguity of the ITS-90 definition. This definition refers to neon generically to ‘natural’ composition, while its uncorrected variability heavily affects the uncertainty of the thermometer calibrations. These results for pure isotopes are compared with those recently obtained at INRIM on samples of commercial neon of natural composition and with literature data. The problems involved in the correction for the residual content of isotopic impurities and for chemical impurities are discussed.  相似文献

All aerobic organisms have developed different mechanisms for neutralising the free radicals, mostly produced by the monoelectronic reduction of O(2), and preventing the severe damages these can provoke. The efficiency of these mechanisms can be assessed, in different matrices, by a simple and direct chemiluminescent assay (CL) based on luminol oxidation catalysed by horseradish peroxidase. Light emission is mediated by the production of free radicals and it is inhibited after a sample addition in a way that is directly proportional to the sample total content of molecules displaying antioxidant activity. The performances of this chemiluminescent assay were compared with those of two spectrophotometric methods already applied in clinical practice. First spectrophotometric method measures, like CL assay, the total antioxidant capacity, whereas the second one determines free thiol groups content. The chemiluminescent assay has a linearity interval between 0.60 and 9.46 mumol l(-1) of Trolox (y=34.91x+3.10; r=0.999; n=5) with an imprecision, expressed as CV, of 3.8% and an inaccuracy, expressed as percentage recovery, of 109%. The first spectrophotometric method, based on the same reference standard, the Trolox molecule, has a linearity interval between 0.2 and 2.5 mmol l(-1) of Trolox (y=-0.01x+4.54; r=0.95; n=5); the thiol groups assay has a linearity interval between 0.1 and 1 mmol l(-1) of l-cysteine (y=1.68x-47.09; r=0.998; n=5). Different clinical samples of plasma from healthy individuals, obese subjects and patients with liver diseases were tested. Interesting correlations were obtained among the three methods, but no significant correlations emerged between antioxidant capacity and clinical parameters. Significant differences were there only between men and women among obese subjects and between drinkers and non-drinkers among liver disease patients.  相似文献

Knowledge of the structure of active sites is a prerequisite for the rational design of solid catalysts. Using site-selective blocking by CO and isotope labeling combined with in situ attenuated total reflection infrared (ATR-IR) spectroscopy, we were able to discriminate the different sites involved in the liquid-phase oxidation of benzyl alcohol on Pd/Al(2)O(3). The main reaction, that is, the oxidative dehydrogenation of the alcohol to the corresponding aldehyde, showed only little dependence on structure and occurred on all exposed Pd faces, whereas the undesired product decarbonylation occurred preferentially on hollow sites on (111) Pd faces. This explains why specific blocking of the latter sites, as realized in the industrially used Pd-Bi/Al(2)O(3) catalysts, leads to improved catalytic performance.  相似文献

A bacterial alpha-d-glucopyranosyl-1-phosphate thymidylyltransferase was found to couple four hexofuranosyl-1-phosphates, as well as a pentofuranosyl-1-phosphate, with deoxythymidine 5'-triphosphate, providing access to furanosyl nucleotides. The enzymatic reaction mixtures were analyzed by electrospray ionization mass spectrometry and NMR spectroscopy to determine the anomeric stereochemistry of furanosyl nucleotide products. This is the first demonstration of a nucleotidylyltransferase discriminating between diastereomeric mixtures of sugar-1-phosphates to produce stereopure, biologically relevant furanosyl nucleotides.  相似文献

The response of interfacial layers to deformations in size and shape depends on their composition. The corresponding main mechanical quantities are elasticity and viscosity of dilation and shear, respectively. Hence, the interfacial rheology represents a kind of two-dimensional equivalent to the traditional bulk rheology. Due to growing interest in the quantitative understanding of foams and emulsions, more works are dedicated to studies on interfacial rheology. This overview presents the theoretical basis for traditional and recently developed experimental tools and discusses their application to different interfacial systems. While dilational rheology provides information on the composition of mixed interfacial layers, the shear rheology gives answers essentially on structures formed at an interface. The most frequently used methods at present are the oscillating drop and bubble tensiometry methods for dilational deformations and oscillating ring/bicone rheometers for shear deformations.  相似文献
