CRANAD-2 is a fluorogenic curcumin derivative used for near-infrared detection and imaging in vivo of amyloid aggregates, which are involved in neurodegenerative diseases. We explore the performance of CRANAD-2 in two super-resolution imaging techniques, namely stimulated emission depletion (STED) and single-molecule localization microscopy (SMLM), with markedly different fluorophore requirements. By conveniently adapting the concentration of CRANAD-2, which transiently binds to amyloid fibrils, we show that it performs well in both techniques, achieving a resolution in the range of 45–55 nm. Correlation of SMLM with atomic force microscopy (AFM) validates the resolution of fine features in the reconstructed super-resolved image. The good performance and versatility of CRANAD-2 provides a powerful tool for near-infrared nanoscopic imaging of amyloids in vitro and in vivo.  相似文献

It is shown that a vorticity, constructed from the spin field of a quantum spinning plasma, combines with the classical generalized vorticity (representing the magnetic and the velocity fields) to yield a new grand generalized vorticity that obeys the standard vortex dynamics. Expressions for the quantum or spin vorticity and for the resulting generalized helicity invariant are derived. Reduction of the rather complex spinning quantum system to a well known and highly investigated classical form opens familiar channels for the delineation of physics peculiar to dense plasmas spanning solid state to astrophysical objects. A simple example is worked out to show that the magnetics of a spinning plasma can be much richer than that of the corresponding classical system.  相似文献

A new derivative of the previously reported 1,2‐bis(benzimidazol‐2‐yl)ethane motif, cation [1H2]2+, was synthesized under microwave irradiation and fully characterized by solution NMR, high‐resolution mass spectrometry, cyclic voltammetry and X‐ray crystallography. This cation presents a linear geometry and incorporates nitro substituents as electrochemical handles. In solution, cation [1H2]2+, is capable of threading the cavity of dibenzo‐24‐crown‐8 ether host (DB24C8) giving rise to a [2]pseudorotaxane complex [1H2?DB24C8]2+, regardless of the counterion, [CF3SO3]? or [CF3COO] ?. The interpenetrated structure of [1H2?DB24C8]2+ was proven by solution NMR and X‐ray crystallography. This host–guest complex is held together by several non‐covalent interactions, such as hydrogen bonding and ion‐dipole. An electrochemical study of [1H2]2+ in the presence of variable amounts of DB24C8 was performed; due to the irreversible redox behavior of cation [1H2]2+, it was not possible to electrochemically control the association/dissociation process with DB24C8. β-Carboline imine reductions mediated by Saccharomyces bayanus have been described achieving moderate to good enantiomeric excesses of the amine products. The enantiomeric excesses of the bioreduction showed a dependence on the imine substituents. Compounds presenting C1–C11 aliphatic substituent groups afforded amines with an (S)-configuration, whereas C15 and higher aliphatic- and aromatic-substituted β-carboline imines achieved inversion of the configuration in the final (R)-2 amine products. Based on this data, a model for the Saccharomyces reduction is proposed.  相似文献
