For enhancing immunogenicity and develop vaccine strategies using peptide based constructs against HIV-1, a chimeric peptide
containing V3 loop and transmembrane sequence of gp41 with two glycine motifs as spacer was constructed. The V3-gp41, gp41
peptide and p17 and p24 peptides separately or in a cocktail were entrapped with or without MA729 as an immunoadjuvant in
liposomes or ISCOMs. The immunogenicity, antigen induced T-cell proliferation and cytokine profiles of various formulations
were studied in four different inbred strains of mice of H-2d, H-2b, H-2k and H-2q haplotypes, keeping alum as a control adjuvant. Both liposomes and ISCOM preparations elicited high titer and long lasting
antibody response (60 days and above). When compared to the alum formulation, the liposomes co-entrapped with MA729 produced
high antibody levels, comparable with that induced by ISCOMs. Peptide in alum, liposomes and ISCOMs enhanced both antigen
specific IgG2a and IgG2b isotypes and high T-cell stimulation index. Peptide formulations also induced antibodies with high
affinity and in vitro neutralizated the formation of HIV-1 syncytia. T-cell supernatants contained high levels of IFN-γ and IL-2. Thus formulation
in these adjuvants induced a predominant Th1 like response with MA729 as a versatile novel delivery vehicle for stimulating
the appropriate arm of the immune response that can selectively modulate MHC class I or MHC class II response. The above peptide
can be of wide vaccination interest as a means to improve immune responses to several other HIV-1 antigens and may serve as

An ensemble of random particle-hole matrices is studied as a function of the signs of the matrix elements. When all matrix elements have the same sign, a theorem due to Perron ensures the existence of a coherent state: the collective particle-hole state. The example of Gamow-Teller transitions between208Bi(1+) and208Pb(0+) is considered and the strength of the Gamow-Teller giant resonance as well as the fluctuations in the ensemble are studied. When a fractionp of matrix elements are of opposite sign, we find that a giant resonance may still exist providedp is small enough. Its magnitude and position in the spectrum are studied. The method proposed is valid for any particle hole excitation. The two body matrix elements of several interactions are analysed and are shown to fit into a unique valuep=0.2 for the 1 + states. 相似文献

We present experimentally measured absolute values of the photoionization
cross sections from the 5s5p 1P1 and 5s5p 3P1 excited
states of strontium at the first ionization threshold as 11.4±1.8 Mb
and 10.7±1.7 Mb respectively using saturated absorption technique
along with a thermionic diode ion detector in conjunction with a Nd:YAG
pumped dye laser system. These threshold photoionization cross sections
values have been utilized to determine the oscillator strengths of the
5s5p 1P1↦5snd 1D2 and 5s5p 3P1↦5snd 3D2 Rydberg transitions. The oscillator strength densities in the
continuum corresponding to the 5s5p 3P1 excited state have also
been determined by measuring the photoionization cross sections at five
ionizing wavelengths above the first ionization threshold. Smooth merging of
the discrete f-values into the oscillator strength densities has been
observed for the 5s5p 3P1↦5snd 3D2 series across the

We study the effects of quantum interference in the spontaneous emission spectrum of a four-level driven atomic system. We use three strong laser fields to drive the atom and a weak laser field to prepare the initial state of the atom. The atomic system exhibits Autler-Townes triplet in the spectrum. The single Lorentzian peak splits into triplet and their widths are controlled by the relative strengths of the laser fields. 相似文献

Some new pyrimidine derivatives have been synthesised by electrochemical oxidation of catechol (1a) in the existence of 2-mercapto-6-(trifluoromethyl) pyrimidine-4-ol (3) as a nucleophile in aqueous solution using Cyclic Voltammetric and Controlled Potential Coulometry. The catechol has been oxidised to o-quinone through electrochemical method and participative in Michael addition reaction, leading to the development of some new pyrimidine derivatives. The products were achieved in good yield with high pureness. The mechanism of the reaction has been conformed from the Cyclic Voltammetric data and Controlled Potential Coulometry. After purification, the compounds were characterised using modern techniques. The synthesised materials were screened for antimicrobial actions using Gram positive and Gram negative strain of bacteria. These new synthesised pyrimidine derivatives showed very good antimicrobial activity. 相似文献

This paper describes the design and synthesis of a polymerizable lipid capable of complexing lanthanide ions. The lipid has been successfully incorporated into liposomes and then polymerized. Fluorescence studies indicate that the diacetylene (unpolymerized lipid) and the conjugated alkenes (after polymerization) can be used as sensitizers for the lanthanide ion. 相似文献

This work describes the application and usefulness of the focused ion beam (FIB) technique for the preparation of transmission electron microscopy (TEM) samples from metal matrix composite materials. Results on an Aldiamond composite, manufactured by the squeeze casting infiltration process, were chosen for demonstration. It is almost impossible to prepare TEM specimens of this material by any other conventional method owing to the presence of highly inhomogeneous phases and reinforcement diamond particles. The present article gives a detailed account of the salient features of the FIB technique and its operation. One of the big advantages is the possibility to prepare site-specific TEM specimens with high spatial resolution. The artifacts occurring during the specimen preparation, for example, Ga-ion implantation, curtain effects, amorphous layers, bending of the lamella, or different milling behaviors of the materials have been discussed. Furthermore, TEM examination of the specimens prepared revealed an ultrafine amorphous layer of graphite formed at the interface between the Al and diamond particles that may affect the interfacial properties of the composite materials. This may not have been feasible without the successful application of the FIB technique for production of good quality site-specific TEM specimens. 相似文献

Equilibrium colloid stability measurements with nonionic surfactant (C12E8) stabilised polybutyl methacrylate (PBMA) latex dispersions indicate a sudden temperature induced destabilisation coinciding with the glass transition temperature,Tg, of the polymer. In control experiments with polystyrene latex particles of similar size, for whichTg was not approached, the flocculation temperature was significantly higher. The effect is interpreted in terms of a reduced adsorbed layer thickness aboveTg caused by mixing of part of the surfactant molecule with the polymer. This interpretation is supported by DSC, elastic modulus and mechanical damping measurements on films made from dispersions of the same latex containing commercial nonionic surfactants. These measurements indicate a shift inTg in the presence of surfactant consistent with partial penetration of the polymer surface by the surfactant. In addition, C12E8 adsorption measurements show increased adsorption (or absorption) onto PBMA aboveTg which is irreversible on both dilution and temperature reduction. 相似文献
