This study deals with the isolation of novel mutant of Bacillus and optimisation of media for the hyperproduction of cellulase. Cellulase-producing Bacillus PC-BC6 was subjected to physical and chemical mutagenesis to enhance the cellulolytic potential. Later, mutagenesis isolates were screened both qualitatively and quantitatively. Among all the tested isolates, Bacillus N3 yielded maximum (CMCase 1250 IU/mL/min and FPase 629 IU/mL/min) activity. The Bacillus N3 strain exhibited 1.7-fold more enzyme production as compared with the parental strain. Proximate analysis of untreated and pretreated Saccharum spontaneum was carried out to improve cellulase production. Three different media were tested for the production of cellulase, among which M2 medium containing MgSO4, pretreated S. spontaneum, K2HPO4, (NH4)2SO4 and peptone was found to be the best for maximum enzyme production by mutant Bacillus N3.  相似文献

Large doses of ionizing radiation can damage human tissues. Therefore, there is a need to investigate the radiation effects as well as identify effective and non-toxic radioprotectors. This study evaluated the radioprotective effects of Kelulut honey (KH) from stingless bee (Trigona sp.) on zebrafish (Danio rerio) embryos. Viable zebrafish embryos at 24 hpf were dechorionated and divided into four groups, namely untreated and non-irradiated, untreated and irradiated, KH pre-treatment and amifostine pre-treatment. The embryos were first treated with KH (8 mg/mL) or amifostine (4 mM) before irradiation at doses of 11 Gy to 20 Gy using gamma ray source, caesium-137 (137Cs). Lethality and abnormality analysis were performed on all of the embryos in the study. Immunohistochemistry assay was also performed using selected proteins, namely γ-H2AX and caspase-3, to investigate DNA damages and incidences of apoptosis. KH was found to reduce coagulation effects at up to 20 Gy in the lethality analysis. The embryos developed combinations of abnormality, namely microphthalmia (M), body curvature and microphthalmia (BM), body curvature with microphthalmia and microcephaly (BMC), microphthalmia and pericardial oedema (MO), pericardial oedema (O), microphthalmia with microcephaly and pericardial oedema (MCO) and all of the abnormalities (AA). There were more abnormalities developed from 24 to 72 h (h) post-irradiation in all groups. At 96 h post-irradiation, KH was identified to reduce body curvature effect in the irradiated embryos (up to 16 Gy). γ-H2AX and caspase-3 intensities in the embryos pre-treated with KH were also found to be lower than the untreated group at gamma irradiation doses of 11 Gy to 20 Gy and 11 Gy to 19 Gy, respectively. KH was proven to increase the survival rate of zebrafish embryos and exhibited protection against organ-specific abnormality. KH was also found to possess cellular protective mechanism by reducing DNA damage and apoptosis proteins expression.  相似文献

Melamine‐formaldehyde (MF) resins of improved environmental compatibility were synthesized at different molar ratios, temperature and pH. Five molar ratios of melamine and formaldehyde: 35:65, 36.5:63.5, 38.3:61.7, 41.1:58.9 and 42.4:58.6 were used to synthesize the corresponding resins. The prepared samples were characterized by using molecular weight determination viscometery, field emission scanning electron microscopy and thermogravimetric analysis. It was noticed that the solid content in the resin increases with an increase in pH. The maximum percentage of the solid content (51 %) was obtained for pH of 8.5 and temperature range of 95–98 oC. The refractive index of the final product was found increasing with an increase of melamine concentration in the resin. A similar trend was noticed in molecular weight of the resins with an increase in monomer yield. The maximum molecular weight of 114218 was obtained from the sample synthesized with MF molar ratio of 42.4:58.6.  相似文献

Abstract

The widespread and large scale use of platinum group metals, especially palladium, in a wide variety of industrial applications has seen their levels in wastewater streams, roadside dust and even pharmaceuticals significantly rise over recent years. Due to the possible environmental damage and potential health risk this may cause, there is now substantial demand for inexpensive, efficient and robust methods for the detection of palladium. Based upon self-immolative linker technologies, we have designed and synthesised a number of allyl ether-functionalised electrochemical probes to determine the optimum probe structure required to deliver a ratiometric electrochemical detection method capable of achieving a limit of detection of <1 mg/mL within 20 min through the use of disposable screen-printed carbon electrodes. Combined with an enzymatic assay, this method was then used to achieve a proof-of-principle ratiometric electrochemical molecular logic gate.  相似文献

A new metabolite,named seimatoric acid(1),representing a new oxobutanoic acid derivative has been isolated from Seimatosporium sp., in addition to four known compounds viz.,2-hydroxymethyl-4β,5α,6β-trihydroxycyclohex-2-enone(2),(-)-phyllostine(3),(+)-epiepoxydon(4) and(+)-epoxydon monoacetate(5).Similarly one new benzoic acid derivative,named colletonoic acid(6) was isolated from the ethyl acetate fraction of Colletotrichum sp.The structures of the new compounds were elucidated by detailed ~1 H NMR,~(13)C NMR,COSY,HMQC.HMBC spectroscopic analysis,and HR-E1-MS.Seimatoric acid(1)was also isolated from another taxonomical unidentified fungal strain 4295 in ourgroup.The structures of the known compounds were elucidated by their spectral data comparison to literature data.Preliminary studies showed that colletonoic acid(6) showed good antibacterial,antifungal,and antialgal activities.  相似文献

The kinetics of the oxidation of [Ni(II)(H2L1)](ClO4)2, (H2L1 = 3,8-dimethyl-4,7-diaza-3,7-decadiene-2,9-dione dioxime) and [Ni(II)(HL2)]ClO4, (H2L2 = 3,9-dimethyl-4,8-diaza-3,8-undecadiene-2,10-dione dioxime) by peroxodisulfate anion (PDS) in aqueous media at 298.0 K have been studied. The kinetics of oxidation of both Ni(II) complexes was found to be first order in the complex concentration. The dependence of the pseudo-first-order rate constant, k obs, for both complexes showed first-order dependence on PDS concentration. The kinetics of oxidation of [Ni(II)(H2L1)]2+ complex showed a complex dependence on [H+] over the pH range of 4.98–7.50, whereas that of [Ni(II)(HL2)]+ is independent of pH over the pH range of 5.02–7.76. The value of k obs, for both complexes, decreased with increasing ionic strength consistent with the involvement of oppositely charged ions in the rate-determining step. The effect of ionic strength is more pronounced for [Ni(II)(H2L1)]2+–PDS reaction than for [Ni(II)(HL2)]+–PDS reaction, confirming the higher charges of the latter.  相似文献

This study was designed to determine the adaptation capability of bitter melon (Momordica charantia L.), which is widely grown in tropical and subtropical climates, in northern parts of Turkey. In this study, plant height, number of fruits, fruit length, fruit width, number of seeds and fruit weight of bitter melon grown in field conditions were determined. The antimicrobial effect of the ethanol extract of fruit and seeds against Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Salmonella typhi, Aspergillus niger and Candida albicans microorganisms was tested in vitro by the disc diffusion method. In conclusion, plant height (260 cm), number of fruits (16 per plant), number of seeds (30.2 per fruit), fruit width (3.8 cm), fruit length (10.6 cm) and fruit weight (117.28 g fruit? 1) were determined; fruits were found to have antimicrobial activity against A. niger; oil and seeds were found to have antimicrobial activity against A. niger and E. coli.  相似文献

Extreme states of matter such as Warm Dense Matter “WDM” and Dense Strongly Coupled Plasmas “DSCP” play a key role in many high energy density experiments, however, creating WDM and DSCP in a manner that can be quantified is not readily feasible. In this paper, isochoric heating of matter by intense heavy ion beams in spherical symmetry is investigated for WDM and DSCP research: the heating times are long (100 ns), the samples are macroscopically large (millimeter-size) and the symmetry is advantageous for diagnostic purposes. A dynamic confinement scheme in spherical symmetry is proposed which allows even ion beam heating times that are long on the hydrodynamic time scale of the target response. A particular selection of low-Z target tamper and X-ray probe radiation parameters allows to identify the X-ray scattering from the target material and use it for independent charge state measurements Z* of the material under study.  相似文献
