Temperature and solvent dependent NMR studies on mangiferin and complete NMR spectral assignments of its acyl and methyl derivatives

By employing concerted 1 and 2D NMR techniques, exact NMR spectral assignments have been made of the acyl (2-7) and methyl (8 and 9) derivatives of mangiferin (1) isolated from the leaves of Bombax ceiba. Derivatives 2, 8 and 9 have been reported in literature, while 3-7 represent new compounds. The acetates 2 and 3 were found to be unstable and were converted into the same penta-acetate 4 at room temperature. Extensive NMR studies on mangiferin (1) and its derivatives showed that H-4 exchanges with deuterium of the solvent molecule more easily. This exchange under acidic conditions occurred at that position (C-4) where electrophilic substitution reactions can easily take place. This is the first report describing the exchange of C-4 proton of mangiferin (1), or any other xanthone, with deuterium of solvent molecules.     

Von Neumann's Problem and Large Cardinals

It is a well-known problem of Von Neumann to discover whetherthe countable chain condition and weak distributivity of a completeBoolean algebra imply that it carries a strictly positive probabilitymeasure. It was shown recently by Balcar, Jech and Pazák,and by Velikovi, that it is consistent with ZFC, modulo theconsistency of a supercompact cardinal, that every ccc weaklydistributive complete Boolean algebra carries a contiuous strictlypositive submeasure – that is, it is a Maharam algebra.We use some ideas of Gitik and Shelah and implications fromthe inner model theory to show that some large cardinal assumptionsare necessary for this result. 2000 Mathematics Subject Classification03E55, 28A60 (primary), 03E75 (secondary).

In vitro Photo‐Oxidation of Pyruvate to Acetyl: A Main Source for the Formation of Acetyl Coenzyme A in the Citric Acid Cycle to Perform Biochemical Reactions

The kinetics of photooxidation of pyruvate was investigated in presence of zinc oxide catalyst under illumination of visible light. The influence of different parameters such as concentrations of reactants, amount of catalyst and irradiation time was studied on the redox reaction under pseudo‐first order conditions. The results indicated that amount of catalyst, presence of electron accepter in solution and irradiation time was the key factors influencing the efficiency of photo‐oxidation of pyruvate to acetyl.     

Development and validation of a stability‐indicating assay including the isolation and characterization of degradation products of metaxalone by LC‐MS

A stability‐indicating reverse‐phase high‐performance liquid chromatography–mass spectrometric method was developed and validated for the assay of metaxalone through forced degradation under acidic, alkaline, photo, oxidative and peroxide stress conditions. Separation of degradation products was accomplished on a reverse‐phase Phenomenex C₁₈ (250 × 4.6 mm, 5 µm) column thermostated at 25°C using 10 mM aqueous ammonium acetate: methanol (35:65 v/v) as mobile phase in an isocratic mode of elution. The eluents were detected at 275 nm by photo diode array detector and mass detectors connected in series. Two unknown base hydrolysis products of metaxalone were identified and characterized as (a) methyl 3‐(3,5‐dimethylphenoxy)‐2‐hydroxypropylcarbamate and (b) 1‐(3,5‐dimethylphenoxy)‐3‐aminopropan‐2‐ol by MS, ¹H NMR and FTIR spectroscopy. The method was validated as per International Conference on Harmonization guidelines and metaxalone was selectively determined in presence of its degradation impurities, demonstrating its stability‐indicating nature.
