Ammonia-Borane Derived BN Fragments Trapped on Bi- and Trimetallic Titanium(III) Systems

Titanium(III) complexes containing unprecedented (NH₂BH₂NHBH₃)²⁻ and {N(BH₃)₃}³⁻ ligands have been isolated, and their structures elucidated by a combination of experimental and theoretical methods. The treatment of the trimethyl derivative [TiCp*Me₃] (Cp*=η⁵-C₅Me₅) with NH₃BH₃ (3 equiv) at room temperature gives the paramagnetic dinuclear complex [{TiCp*(NH₂BH₃)}₂(μ-NH₂BH₂NHBH₃)], which at 80 °C leads to the trinuclear hydride derivative [{TiCp*(μ-H)}₃{μ₃-N(BH₃)₃}]. The bonding modes of the anionic BN fragments in those complexes, as well as the dimethylaminoborane group trapped on the analogous trinuclear [{TiCp*(μ-H)}₃(μ₃-H)(μ₃-NMe₂BH₂)], have been studied by X-ray crystallography and density functional theory (DFT) calculations.     

pH‐Responsive Pharmacological Chaperones for Rescuing Mutant Glycosidases

A general approach is reported for the design of small‐molecule competitive inhibitors of lysosomal glycosidases programmed to 1) promote correct folding of mutant enzymes at the endoplasmic reticulum, 2) facilitate trafficking, and 3) undergo dissociation and self‐inactivation at the lysosome. The strategy is based on the incorporation of an orthoester segment into iminosugar conjugates to switch the nature of the aglycone moiety from hydrophobic to hydrophilic in the pH 7 to pH 5 window, which has a dramatic effect on the enzyme binding affinity. As a proof of concept, new highly pH‐responsive glycomimetics targeting human glucocerebrosidase or α‐galactosidase with strong potential as pharmacological chaperones for Gaucher or Fabry disease, respectively, were developed.     

Bayesian Nonparametric Analysis for a Generalized Dirichlet Process Prior

This paper considers a generalization of the Dirichlet process which is obtained by suitably normalizing superposed independent gamma processes having increasing integer-valued scale parameter. A comprehensive treatment of this random probability measure is provided. We prove results concerning its finite-dimensional distributions, moments, predictive distributions and the distribution of its mean. Most expressions are given in terms of multiple hypergeometric functions, thus highlighting the interplay between Bayesian Nonparametrics and special functions. Finally, a suitable simulation algorithm is applied in order to compute quantities of statistical interest.
