An important feature of the synthesis of salinosporamide A, a potent proteasome inhibitor, is the establishment of the quaternary stereocenter at C3. A new route has been developed based on the methylation of a functionalized pyrrolidinone. Direct methylation reaction led to the unwanted diastereomer; however, by means of a Corey–Chaykovsky reaction followed by LiAlH4 epoxide opening, the desired alcohol was obtained. The pyrrolidinone was elaborated through a key allylation reaction between a tertiary allyltitanium reagent and an aldehyde bearing a spiroketal moiety in α-position.  相似文献

An experimental study on the interplay of sorbent structure and fluid phase conditions (pH) has been carried out examining adsorption and transport of bovine serum albumin (BSA) and a monoclonal antibody (IgG 2a) on SP Sepharose Fast Flow and SP Sepharose XL. SP Sepharose Fast Flow is characterised by a relatively open pore network, while SP Sepharose XL is a composite structure with ligand-carrying dextran chains filling the pore space. Both adsorbents have similar ionic capacity. Protein transport and adsorption profiles were evaluated using confocal laser scanning microscopy. Under all investigated conditions, BSA uptake could be adequately explained by a pore diffusion mechanism. The adsorption profiles obtained for IgG 2a, however, indicated that changes in fluid phase conditions as well as a change in the solid phase structure could result in a more complex uptake mechanism as compared to pore diffusion alone. This mechanism results in a fast transport of proteins into the adsorbent, followed by an overshoot of protein in the center of the sorbent and a setback towards a homogeneous adsorption profile.  相似文献

Organometallic Compounds of the Lanthanides. 139 Mixed Sandwich Complexes of the 4 f Elements: Enantiomerically Pure Cyclooctatetraenyl Cyclopentadienyl Complexes of Samarium and Lutetium with Donor‐Functionalized Cyclopentadienyl Ligands The reactions of [K{(S)‐C5H4CH2CH(Me)OMe}], [K{(S)‐C5H4CH2CH(Me)NMe2}] and [K{(S)‐C5H4CH(Ph)CH2NMe2}] with the cyclooctatetraenyl lanthanide chlorides [(η8‐C8H8)Ln(μ‐Cl)(THF)]2 (Ln = Sm, Lu) yield the mixed cyclooctatetraenyl cyclopentadienyl lanthanide complexes [(η8‐C8H8)Sm{(S)‐η5 : η1‐C5H4CH2CH(Me)OMe}] ( 1 a ), [(η8‐C8H8)Ln{(S)‐η5 : η1‐C5H4CH2CH(Me)NMe2}] (Ln = Sm ( 2 a ), Lu ( 2 b )) and [(η8‐C8H8)Ln{(S)‐η5 : η1‐C5H4CH(Ph)CH2NMe2}] (Ln = Sm ( 3 a ), Lu ( 3 b )). For comparison, the achiral compounds [(η8‐C8H8)Ln{η5 : η1‐C5H4CH2CH2NMe2}] (Ln = Sm ( 4 a ), Lu ( 4 b )) are synthesized in an analogous manner. 1H‐, 13C‐NMR‐, and mass spectra of all new compounds as well as the X‐ray crystal structures of 3 b and 4 b are discussed.  相似文献

A highly sensitive and simple method, based on hydride generation and atomic fluorescence detection, has been developed for the determination of As, Bi, Sb, Se(IV) and Te(IV) in aqua regia extracts from atmospheric particulate matter samples. Atmospheric particulates matter was collected on glass fiber filters using a medium volume sampler (PM1 particulate matter). Two-level factorial designs have been used to optimise the hydride generation atomic fluorescence spectrometry (HG-AFS) procedure. The effects of several parameters affecting the hydride generation efficiency (hydrochloric acid, sodium tetrahydroborate and potassium iodide concentrations and flow rates) have been evaluated using a Plackett-Burman experimental design. In addition, parameters affecting the hydride measurement (delay, analysis and memory times) have been also investigated. The significant parameters obtained (sodium tetrahydroborate concentration, sodium tetrahydroborate flow rate and analysis time for As; hydrochloric acid concentration and sodium tetrahydroborate flow rate for Se(IV); and sodium tetrahydroborate concentration and sodium tetrahydroborate flow rate for Te(IV)) have been optimized by using 2n + star central composite design. Hydrochloric acid concentration and sodium tetrahydroborate flow rate were the significant parameters obtained for Sb and Bi determination, respectively. Using a univariate approach these parameters were optimized. The accuracy of methods have been verified by using several certified reference materials: SRM 1648 (urban particulate matter) and SRM 1649a (urban dust). Detection limits in the range of 6 × 10−3 to 0.2 ng m−3 have been achieved. The developed methods were applied to several atmospheric particulate matter samples corresponding to A Coruña city (NW Spain).  相似文献

The synthesis of palladium nanoparticles is performed by hydrogenation of a precursor, Pd2(dba)3 (1) or [Pd(C3H5)Cl]2 (2) in the presence of either a weakly coordinating ligand (hexadecylamine, HDA) or good ligands (polyphosphines). It is shown in the case of 1 that good ligands lead to stable spherical nanoparticles of small size (near 2 nm) whereas the protective effect of HDA depends on the amount of ligand added as a result of equilibria present at the surface of the particles as monitored by solution NMR spectroscopy. The decomposition of 2 being very fast, the particle growth cannot be controlled except in the case of the use of a large excess of HDA which leads to spongelike particles resulting from the agglomeration of initially obtained nanocrystallites.  相似文献

Seven 2‐amino‐4‐aryl‐1,3‐thiazoles ( 1a–g ) and their corresponding 2‐aminoacetyl ( 2a–g ) and 2‐aminoacetyl‐5‐bromo ( 3a–g ) derivatives were synthesized and tested in vitro against 11 reference strains, three Gram‐positive and four Gram‐negative bacteria, two yeasts, and two moulds. Toxicity of the compounds was also evaluated using the brine shrimp test. Compounds 1a, 1b, 1e–g , and 3b showed moderate antimicrobial activity at different concentrations. The results indicated that acetylation of the amino group and bromination at position 5 of the thiazole moiety cause lost of activity. Compounds 1a, 1e , and 1f showed toxicity to brine shrimp nauplii below 10 ppm. Most other compounds showed moderate toxicity, LD50 above 100 ppm. A sensitive and specific method for determining fenitrothion and its main metabolites, 3-methyl-4-nitrophenol and fenitrooxon, in poplar leaves using deuterated isotopes as the internal standard is described. The analytes and the labeled isotopes were extracted from leaves by solid-phase microextraction and subsequently analysed by gas chromatography coupled with mass spectrometry. The method had a chromatographic run time of 17.0 min and good linearity over the range 0.01-10 mg kg(-1). The detection limits ranged between 2.5 and 0.6 microg kg(-1). The isotopic dilution technique allowed improving significantly the repetitivity even using different fibers with the same coating (RSD<5.1%). The method was applied successfully to study the persistence of fenitrothion in forestal matrices in a poplar forest after cannon spray application of the insecticide.  相似文献
