In time-resolved and spatially resolved experiments, singlet molecular oxygen, O2(a1Deltag), was created in a single nerve cell upon irradiation of a sensitizer incorporated in the cell using a focused laser beam. The singlet oxygen thus produced was detected by its infrared phosphorescence. Data obtained indicate that in both the cytoplasm and the nucleus of the cell, this reactive species is approximately 1-2 orders of magnitude longer-lived than previously believed. The data demonstrate that deactivation of singlet oxygen in the cell is dominated by interactions with the solvent not cellular constituents such as proteins. These results provide a new perspective for mechanistic studies of the role of O2(a1Deltag) in photoinduced cell death and intracellular signaling.  相似文献

Computer aided synthesis planning (CASP) is part of a suite of artificial intelligence (AI) based tools that are able to propose synthesis routes to a wide range of compounds. However, at present they are too slow to be used to screen the synthetic feasibility of millions of generated or enumerated compounds before identification of potential bioactivity by virtual screening (VS) workflows. Herein we report a machine learning (ML) based method capable of classifying whether a synthetic route can be identified for a particular compound or not by the CASP tool AiZynthFinder. The resulting ML models return a retrosynthetic accessibility score (RAscore) of any molecule of interest, and computes at least 4500 times faster than retrosynthetic analysis performed by the underlying CASP tool. The RAscore should be useful for pre-screening millions of virtual molecules from enumerated databases or generative models for synthetic accessibility and produce higher quality databases for virtual screening of biological activity.

The retrosynthetic accessibility score (RAscore) is based on AI driven retrosynthetic planning, and is useful for rapid scoring of synthetic feasability and pre-screening of large datasets of virtual/generated molecules.  相似文献

The lowest excited electronic state of molecular oxygen, singlet molecular oxygen, O(2)(a (1)Delta(g)), is a reactive species involved in many chemical and biological processes. To better understand the roles played by singlet oxygen in biological systems, particularly at the sub-cellular level, optical tools have been developed to create and directly detect this transient state in time- and spatially-resolved experiments from single cells. Data obtained indicate that, contrary to common perception, this reactive species can be quite long-lived in a cell and, as such, can diffuse over appreciable distances including across the cell membrane into the extracellular environment. On one hand, these results demonstrate that the behavior of singlet oxygen in an intact cell can be significantly different from that inferred from model bulk studies. More generally, these results provide a new perspective for mechanistic studies of intra- and inter-cellular signaling and events that ultimately lead to photo-induced cell death.  相似文献

We present a study of the LHC sensitivity to the W boson mass based on simulation studies. We find that both experimental and phenomenological sources of systematic uncertainties can be strongly constrained with Z measurements: the lineshape, dσ Z /dm, is robustly predicted, and its analysis provides an accurate measurement of the detector resolution and absolute scale, while the differential cross-section analysis, d2 σ Z /dydp T , absorbs the strong interaction uncertainties. A sensitivity δ m W ∼7 MeV for each decay channel (We ν, Wμ ν), and for an integrated luminosity of 10 fb−1, appears as a reasonable goal.  相似文献

With this letter, we report how friction can be controlled by inducing physical bonds solely within a polyelectrolyte brush layer, while keeping repulsive interactions between the brush layer and the bare surface that slides above. Our results imply that the nature of the bare surface is of minor importance as long as the repulsive surface interaction is maintained.  相似文献

The well-known Roorda frame displays an asymmetric postbuckling behavior in the elastic case, and it is therefore sensitive to asymmetric geometric imperfections. By contrast it is shown here that an elastic-plastic frame displays practically symmetric imperfection sensitivity since the maximum load is attained by a basically different mechanism that only to an insignificant degree invokes that part of the geometric nonlinearity which induces the asymmetry.  相似文献

Hydrodistilled leaf oils of Pistacia chinensis Bunge from five locations in China were analyzed using GC/MS. A total of 58 compounds was identified in the oils, and a relatively high variation in their contents was found. The major compounds include β-phellandrene (0.54–53.86%), α-pinene (4.74–54.44%), β-pinene (0.49–42.90%), caryophyllene (5.64–20.01%), cis-ocimene (tr−43.93%), eudesmadiene (0–15.06%), and camphene (tr−20.57%). Cluster analysis classified the leaf oils into two chemotypes: one rich in α-pinene and β-pinene, and the other rich in β-phellandrene. The interaction between a colloidal polystyrene particle mounted on an AFM cantilever and a hydrophilic and a hydrophobic surface in aqueous solution is investigated. Despite the apparent simplicity of these two types of systems a variety of different types of interactions are observed. The system containing the polystyrene particle and a hydrophilic surface shows DLVO-like interactions characteristic of forces between charged surfaces. However, when the surface is hydrophobized the interaction changes dramatically and shows evidence of a bridging air bubble being formed between the particle and the surface. For both sets of systems, plateaus of constant force in the force curves are obtained when the particle is retracted from the surface after being in contact. These events are interpreted as a number of individual polystyrene molecules that are bridging the polystyrene particle and the surface. The plateaus of constant force are expected for pulling a hydrophobic polymer in a bad (hydrophilic) solvent. The plateau heights are found to be of uniform spacing and independent of the type of surface, which suggests a model by which collapsed polymers are extended into the aqueous medium. This model is supported by a full stretching curve showing also the backbone elasticity and a stretching curve obtained in pentanol, where the plateau changes to a nonlinear force response, which is typical for a polymer in a good or neutral solvent. We suggest that these polymer bridges are important in particular for the interaction between polystyrene and the hydrophilic surface, where they to some extent counteract the long-range electrostatic repulsion.  相似文献

Using two identical 110 femtosecond (fs) optical pulses separated by 310 fs, we launch two dissociative wave packets in I2. We measure the square of the wave function as a function of both the internuclear separation, /Psi(R)/(2), and of the internuclear velocity, /Psi(v(R))/(2), by ionizing the dissociating molecule with an intense 20 fs probe pulse. Strong interference is observed in both /Psi(R)/(2) and in /Psi(v(R))/(2). The interference, and therefore the shape of the wave function, is controlled through the phase difference between the two dissociation pulses in good agreement with calculations.  相似文献

Spectral compression by self-phase modulation of amplitude- and phase-shaped pulses is demonstrated as superior compared to pulses that have only been phase shaped. We synthesize linearly negatively chirped parabolic pulses, which we send through a nonlinear photonic crystal fiber, in which self-phase modulation compresses the spectrum of the pulses to within 20% of the Fourier transform limit.  相似文献
