Methods using high performance liquid chromatography with diode array detection (HPLC-DAD) and tandem mass spectrometry (HPLC-MS/MS) were developed and validated for the simultaneous determination of 5 chromones and 6 coumarins: prim-O-glucosylcimifugin (1), cimifugin (2), nodakenin (3), 4'-O-β-d-glucosyl-5-O-methylvisamminol (4), sec-O-glucosylhamaudol (5), psoralen (6), bergapten (7), imperatorin (8), phellopterin (9), 3'-O-angeloylhamaudol (10) and anomalin (11), in Radix Saposhnikoviae. The separation conditions for HPLC-DAD were optimized using an Ascentis Express C18 (4.6 mm×100 mm, 2.7 μm particle size) fused-core column. The mobile phase was composed of 10% aqueous acetonitrile (A) and 90% acetonitrile (B) and the elution was performed under a gradient mode at a flow rate of 1.0 mL/min. The detection wavelength was set at 300 nm. The HPLC-DAD method yielded a base line separation of the 11 components in 50% methanol extract of Radix Saposhnikoviae with no interfering peaks detected. The HPLC-DAD method was validated in terms of linearity, accuracy and precision (intra- and inter-day), limit of quantification (LOQ), recovery, and robustness. Specific determination of the 11 components was also accomplished by a triple quadrupole tandem mass spectrometer equipped with an electrospray ionization (ESI) source. This HPLC-MS/MS method was also validated by determining the linearity, limit of quantification, accuracy, and precision. Quantification of the 11 components in 51 commercial Radix Saposhnikoviae samples was successfully performed using the developed HPLC-DAD method. The identity, batch-to-batch consistency, and authenticity of Radix Saposhnikoviae were successfully monitored by the proposed HPLC-DAD and HPLC-MS/MS methods.  相似文献

Single-molecule mechanical manipulation has enabled quantitative understanding of not only the kinetics of both bond rupture and protein unfolding, but also the free energy landscape of chemical bond and/or protein folding. Despite recent studies reporting the role of loading device in bond rupture, a loading device effect on protein unfolding mechanics has not been well studied. In this work, we have studied the effect of loading-device stiffness on the kinetics of both bond rupture and protein unfolding mechanics using Brownian dynamics simulations. It is shown that bond rupture forces are dependent on not only loading rate but also the stiffness of loading device, and that protein unfolding mechanics is highly correlated with the stiffness of loading device. Our study sheds light on the importance of loading device effect on the mechanically induced bond ruptures and protein unfolding.  相似文献

The possibility of sampling the free and particle‐bound concentrations of organic compounds was studied using two different sampling techniques at the same time: needle trap device (NTD) and solid‐phase microextraction (SPME). In this study, a mosquito coil was used to produce gaseous (free) and particle‐bound compounds. Allethrin, the active ingredient in mosquito coils, was chosen as the target analyte. Under the same sampling conditions, the amount of allethrin extracted from the mosquito‐coil smoke was higher for the NTD compared to the SPME fiber, while the extracted amounts were almost the same for both devices when sampling gaseous samples of allethrin. These results can be explained by the fact that the SPME fiber can only extract free molecules (based on diffusion), whereas the NTD, an exhaustive sampling device, collects both free and particle‐bound allethrin. Breakthrough for NTD and carryover for both NTD and SPME were negligible under the given sampling and desorption conditions.  相似文献

Mechanical characterization of protein molecules has played a role on gaining insight into the biological functions of proteins, because some proteins perform the mechanical function. Here, we present the mesoscopic model of biological protein materials composed of protein crystals prescribed by Go potential for characterization of elastic behavior of protein materials. Specifically, we consider the representative volume element (RVE) containing the protein crystals represented by C(alpha) atoms, prescribed by Go potential, with application of constant normal strain to RVE. The stress-strain relationship computed from virial stress theory provides the nonlinear elastic behavior of protein materials and their mechanical properties such as Young's modulus, quantitatively and/or qualitatively comparable with mechanical properties of biological protein materials obtained from experiments and/or atomistic simulations. Further, we discuss the role of native topology on the mechanical properties of protein crystals. It is shown that parallel strands (hydrogen bonds in parallel) enhance the mechanical resilience of protein materials.  相似文献

Pulsed electromagnetic field (PEMF) has been shown to improve the rate of peripheral nerve regeneration. In the present study we investigated the expression of neuronal nitric oxide synthase (nNOS) and phospholipase C-gamma1 (PLC-gamma1) in regenerating rat laryngeal nerves during the exposure to PEMF after surgical transection and reanastomosis. Axons were found to regenerate into the distal stump nearly twice faster in PEMF-exposed animals than in the control. Consistently, motor function was better recovered in PEMF-treated rats. The expression of nNOS and PLC-gamma1 was highly enhanced in the regenerated nerves.  相似文献

A convergent synthesis of (+)-asteltoxin (1) has been achieved by the Horner-Emmons olefination of bis(tetrahydrofuran) aldehyde 53 and alpha-pyrone phosphonate 5. A key step features the stereoselective construction of a sterically congested quaternary center embedded in the densely functionalized bis(tetrahydrofuran) subunit by a Lewis acid-catalyzed, pinacol-type rearrangement of an epoxy silyl ether. This pivotal rearrangement methodology parallels the proposed biosynthetic pathway of 1 and is ripe for applications to the stereocontrolled synthesis of structurally complex natural products.  相似文献

A novel all-fiber optical delay line for an advanced optical coherence tomography (OCT) system is proposed. Using the distributed reflectance characteristic of a chirped fiber Bragg grating (CFBG), the optical delay can be obtained without leaving the fiber that forms the system. The proposed delay line consists of a pair of identical CFBGs. The gratings are cascaded in reverse order to cancel the inherent wavelength dependency of CFBGs. Tunable optical delay is obtained by applying small strain on one of the CFBGs through an attached piezo actuator. The small displacement induced by the piezo actuator is amplified by a factor of up to 100 due to the distributed reflectance characteristic of the CFBGs. This principle and characteristics of the proposed optical delay line are analyzed in detail.  相似文献
