The mechanisms for the anionic and cationic interactions with myofibrillar proteins in aqueous solutions were investigated by nuclear magnetic resonance over a wide range of salt concentration. Markedly nonlinear dependeces of the 17O and 23Na NMR transverse relaxation rates on salt concentration were analyzed with a thermodynamic linkage model of salt-dependent solubility and hydration (ligand-induced association model), according to Wyman's theory of linked functions. Nonlinear regression analysis of both 17O and 23Na NMR data suggested cooperative, reversible binding of hydrated ions to myofibrillar proteins. Both ions and water were found to exchange fast, on the NMR timescale, between the binding sites of the myofibrillar proteins and the aqueous solution. At sodium chloride concentrations higher than about 0.1 grams salt/gram water, ion activities have marked effects upon the NMR relaxation rates of both ions and water. A salt activity model allowed quantitative fitting of the NMR data at high salt concentrations. The effect of neglecting the ion activity in solutions of myofibrillar proteins was also estimated and compared with the ligand-induced, cooperative association model for myofibrillar proteins. The comparison between the 17O and 23Na results strongly suggests that water is exchanged as the hydrated ion species between the myofibrillar protein binding sites and the bulk, aqueous solution.  相似文献

Highly stereospecific polymerization of monosubstituted acetylenes was carried out using the Rh complex, [Rh(norbornadiene)Cl]2 catalysts. The resulting polyacetylenes were characterized in detail by 1HNMR, ESR, laser Raman, diffuse reflective UV, and wide angle X-ray diffraction methods. The data showed that the Rh complex were the preferred catalyst to selectively yield the corresponding cis-transoid polymers even at room temperature when alcohol, triethylamine or water was used as the polymerization solvent. Additionally, the resulting cis polyacetylenes were found to have a helical form whose polymer is amorphous or composed of pseudohexagonal structures called π-conjugated columnar as self-assembly or super structure. Further compression of the amorphous cis polymers resulted in cis to trans isomerization at room temperature under vacuum, breaking rotationally the cis C=C bonds giving π-radicals called solitons as the origin of a polymer magnet. On the other hand, the π-conjugated columnar was also found to show an extremely longer wavelength absorption compared with that of the amorphous one, although the absorption maximum was shifted to a shorter wavelength when the columnar was destroyed by the compression. Therefore, the formation of the π-conjugated columnar can be considered as a new and quite useful control method concerning color of such conjugated polymers, i.e., a new concept concerning the color of conjugated polymers.  相似文献

Efficient 1,2‐carboboration reactions to the C=N bond of carbodiimides with 9‐borafluorenes, which give rise to cyclic borane‐amidine conjugates with a seven‐membered BNC5 ring, are reported. The resulting cyclic borane‐amidine conjugates can be hydrolyzed into an acyclic bifunctional biaryl compound carrying both boronic acid and amidine groups, rendering the utility of the two‐step protocol for the synthesis of multi‐functionalized molecular systems with a potential as a supramolecular building block. Furthermore, the conjugated structure of the cyclic boron‐amidine compounds can be changed upon alkylation of the boron atom that increases the coordination number of boron. The combination of Lewis acid (borane) and conjugated base (amidine) provides rich structural diversity of heteroatom‐containing π‐conjugated systems.  相似文献

Poly(phenylsilsesquioxane) containing tributylstannyl groups (TBPSQ) was a useful intermediate for the synthesis of organofunctionalized polysilsesquioxane ( 2 ). The reaction of TBPSQ and monochlorosilane ( 1 ), which possessed an organofunctional group, proceeded under mild and neutral conditions to give 2 with the elimination of tributylstannylchloride. The combined use of the different chlorosilanes 1 enabled the ready preparation of multifunctionalized polysilsesquioxanes 2 . To elucidate intrinsic recognition of carboxamides by zinc(II) in carbonic anhydrase (CA) (as inhibitors) and carboxypeptidase A (CPA) (as substrates), a new series of Zn(2+)-carboxamide-appended cyclen complexes have been synthesized and characterized (cyclen = 1,4,7,10-tetraazacyclododecane). Two types of Zn(2+)-carboxamide interactions have been found. In the first case represented by a zinc(II) complex of carbamoylmethyl-1,4,7,10-tetraazacyclododecane (L(1)), the amide oxygen binds to zinc(II) at slightly acidic pH (to form ZnL(1)), and the deprotonated amide N(-) binds to zinc(II) at alkaline pH (to form ZnH(-1)L(1)) with pK(a) = 8.59 at 25 degrees C and I = 0.1 (NaNO(3)), as determined by potentiometric pH titrations, infrared spectral changes, and (13)C and (1)H NMR titrations. The X-ray crystal structure of ZnH(-1)L(3) (where L(3) = N-(4-nitrophenyl)carbamoylmethyl cyclen, pK(a) = 7.01 for ZnL(3) <==> ZnH(-1)L(3)) proved that the zinc(II) binds to the amidate N(-) (Zn-N(-) distance of 1.974(3) A) along with the four nitrogen atoms of cyclen (average Zn-N distance 2.136 A). Crystal data: monoclinic, space group P2(1)/n (No. 14) with a = 10.838(1) A, b = 17.210(2) A, c = 12.113(2) A, b = 107.38(1) degrees, V = 2156.2(5) A(3), Z = 4, R = 0.042, and R(w) = 0.038. These model studies provide the first chemical support that carboxamides are CA(-) inhibitors by occupying the active Zn(2+) site both in acidic and alkaline pH to prevent the occurrence of the catalytically active Zn(2+)-OH(-) species. In the second case represented by a zinc(II) complex of 1-(N-acetyl)aminoethylcyclen, ZnL(6), the pendant amide oxygen had little interaction with zinc(II) at acidic pH. At alkaline pH, the monodeprotonation yielded a zinc(II)-bound hydroxide species ZnL(6)(OH(-)) (pK(a) = 7.64) with the amide pendant remaining intact. The ZnL(6)(OH(-)) species showed the same nucleophilic activity as Zn(2+)-cyclen-OH(-). The second case may mimic the Zn(2+)-OH(-) mechanism of CPA, where the nucleophilic Zn(2+)-OH(-) species does not act as a base to deprotonate a proximate amide.  相似文献

Indium-catalyzed addition of 1,3-dicarbonyl compounds to 1-iodo-1-alkynes takes place exclusively in a syn-fashion to produce E-iodoalkenes. The iodine atom serves both as an activating group and as a group that controls the regioselectivity of the addition. The E-alkenyl iodide product can be further derivatized using either a one-pot or a two-pot procedure into trisubstituted olefins in high overall yield with retention of the stereochemistry.  相似文献

Ethanol is determined by a sensor system using purified, immobilized mernbrane-bound alcohol dehydrogenase frorn Gluconobacter suboxydans, attached to a platinum disk electrode (3 mm diameter), and covered with a dialysis membrane. Hexacyanoferrate (III) is used as the redox acceptor. To correct for the influence of interfering substances, this alcohol sensor is compensated by a control electrode which has no immobilized enzyme. The potential of these platinum electrodes was set at + 350 mV vs. Ag/AgCl. Linearity was observed in the range 0.1–5 mM ethanol, the response time was less than 5 min, the maximum sensitivity was obtained at 45°C and the optimum pH was in the range 4.5–5.5. The sensitivity decreased to 80% of the initial value after 1 month at 30°C. When the alcohol sensor system was applied to the determination of ethanol in alcoholic beverages, a good correlation was obtained between the results and those obtained by gas chromatography.  相似文献
