Equimolar mixtures of dodecyltrimethylammonium chloride (DTAC) and sodium octyl sulfonate (SOSo) show a vesicle phase at >99 wt % water and a single, fluid lamellar phase for water fractions below 80 wt %. This combination is consistent with the bilayer bending elasticity kappa approximately k(B)T and zero bilayer spontaneous curvature. Caillé line shape analysis of the small-angle X-ray scattering from the lamellar phase shows that the effective kappa depends on the lamellar d spacing consistent with a logarithmic renormalization of kappa, with kappa(o) = (0.8 +/- 0.1)k(B)T. The vesicle size distribution determined by cryogenic transmission electron microscopy is well fit by models with zero spontaneous curvature to give (kappa + (kappa/2)) = (1.7 +/- 0.1)k(B)T, resulting in kappa = (1.8 +/- 0.2)k(B)T. The positive value of kappa and the lack of spontaneous curvature act to eliminate the spherulite defects found in the lamellar gel phases found in other catanionic mixtures. Current theories of spontaneous bilayer curvature require an excess of one or more components on opposite sides of the bilayer; the absence of such an excess at equimolar surfactant ratios explains the zero spontaneous curvature.  相似文献

Flexible spacer chains are utilized to enhance the hybridization of terminally anchored oligonucleotide probes of DNA microarrays. A polymer physics approach identifies an underlying mechanism and yields guidelines for the optimal spacer length in terms of the effect on the equilibrium state. For low grafting densities, the dominant effect arises because of the decimation in the number of accessible chain configurations due to the impenetrable surface. Opposing trends are found for long targets and for short targets. At higher grafting densities, different brush regimes introduce an extra hybridization penalty. A novel brush regime is obtained for long neutral spacers and short targets at intermediate ionic strength where the chain stretching is due to the electrostatic interactions between the probes.  相似文献

Amphiphilic macromolecules containing a polystyrene-adherent peptide domain and a cell-repellent poly(ethylene glycol) domain were designed, synthesized, and evaluated as a cytophobic surface coating. Such cytophobic, or cell-repellent, coatings are of interest for varied medical and biotechnological applications. The composition of the polystyrene binding peptide domain was identified using an M13 phage display library. ELISA and atomic force spectroscopy were used to evaluate the binding affinity of the amphiphile peptide domain to polystyrene. When coated onto polystyrene, the amphiphile reduced cell adhesion of two distinct mammalian cell lines and pathogenic Staphylococcus aureus strains.  相似文献

A convenient synthesis of (t)Bu(3)SiSH and (t)Bu(3)SiSNa(THF)(x)() led to the exploration of "(t)Bu(3)SiSMX" aggregation. The dimer, [((t)Bu(3)SiS)Fe](2)(mu-SSi(t)Bu(3))(2) (1(2)), was formed from [{(Me(3)Si)(2)N}Fe](2)(mu-N(SiMe(3))(2))(2) and the thiol, and its dissolution in THF generated ((t)Bu(3)SiS)(2)Fe(THF)(2) (1-(THF)(2)). Metathetical procedures with the thiolate yielded aggregate precursors [X(2)Fe](mu-SSi(t)Bu(3))(2)[FeX(THF)]Na(THF)(4) (3-X, X = Cl, Br) and cis-[(THF)IFe](2)(mu-SSi(t)Bu(3))(2) (4). Thermal desolvations of 3-Cl, 3-Br and 4 afforded molecular wheels [Fe(mu-X)(mu-SSi(t)Bu(3))](12)(C(6)H(6))(n) (5-FeX, X = Cl, Br) and the ellipse [Fe(mu-I)(mu-SSi(t)Bu(3))](14)(C(6)H(6))(n) (6-FeI). Related metathesis and desolvation sequences led to wheels [Co(mu-Cl)(mu-SSi(t)Bu(3))](12)(C(6)H(6))(n) (5-CoCl) and [Ni(mu-Br)(mu-SSi(t)Bu(3))](12)(C(6)H(6))(n) (5-NiBr). The nickel wheel disproportionated to give, in part, [((t)Bu(3)SiS)Ni](2)(mu-SSi(t)Bu(3))(2) (7), which was also synthesized via salt metathesis. X-ray structural studies of 1(2) revealed a roughly planar Fe(2)S(4) core, while 1-(THF)(2), 3-Br, and 4 possessed simple distorted tetrahedral and edge-shared tetrahedral structures. X-ray structural studies revealed 5-MX (MX = FeCl, FeBr, CoCl, NiBr) to be wheels based on edge-shared tetrahedra, but while the pseudo-D(6)(d) wheels of 5-FeCl, 5-CoCl, and 5-FeBr pack in a body-centered arrangement, those of pseudo-C(6)(v)() 5-NiBr exhibit hexagonal packing and two distinct trans-annular d(Br...Br). Variable-temperature magnetic susceptibility measurements were conducted on 5-FeCl, 5-CoCl, 5-FeBr, and 6-FeI, and the latter three are best construed as weakly antiferromagnetic, while 5-FeCl exhibited modest ferromagnetic coupling. Features suggesting molecular magnetism are most likely affiliated with phase changes at low temperatures.  相似文献

The ribosomal peptidyl transferase center is expected to be regiospecific with regard to its tRNA substrates, yet the ester linkages between the tRNA and the amino acid or peptide are susceptible to isomerization between the O2' and O3' hydroxyls of the terminal A76 ribose sugar. To establish which isomer of the P site tRNA ester is utilized by the ribosome, we prepared two nonisomerizable transition state inhibitors with either an A76 O2' or O3' linkage. Strong preferential binding to the O3' regioisomer indicates that the peptidyl transferase proceeds through a transition state with an O3'-linked peptide in the P-site.  相似文献

The structural content of the denatured state has yet to be fully characterized. In recent years, large residual dipolar couplings (RDCs) from denatured proteins have been observed under alignment conditions produced by bicelles and strained polyacrylamide gels. In this report, we describe efforts to extend our picture of the residual structure in denatured nuclease by measuring RDCs with multiple alignment tensors. Backbone amide 15N-1H RDCs were collected from 4 M urea for a total of eight RDC data sets. The RDCs were analyzed by singular value decomposition (SVD) to determine the number of independent alignment tensors present in the data. On the basis of the resultant singular values and propagated error estimates, it is clear that there are at least three independent alignment tensors. These three independent RDC datasets can be reconstituted as orthogonal linear combinations, (OLC)-RDC datasets, of the eight actually recorded. The first, second, and third OLC-RDC datasets are highly robust to the removal of any single experimental RDC dataset, establishing the presence of three independent alignment tensors, sampled well above the level of experimental uncertainty. The observation that the RDC data span three or more dimensions of the five-dimensional parameter space demonstrates that the ensemble average structure of denatured nuclease must be asymmetric with respect to these three orthogonal principal axes, which is not inconsistent with earlier work demonstrating that it has a nativelike topology.  相似文献

Rainwater is a very low concentrated matrix and, for dissolved organic matter (DOM) characterization, an efficient extraction procedure is essential. Isolation procedures based on the adsorption onto XAD-8 and C-18 sorbents have been used in the literature for rainwater DOM isolation, but a comparison between these procedures is lacking. In this work, UV-visible and molecular fluorescence spectroscopies highlighted differences between rainwater DOM isolated by DAX-8 (replacement for XAD-8) and by C-18. It was possible to recover higher rainwater DOM percentage by the C-18 based procedure than by the DAX-8 one. Rainwater protein-like compounds were better concentrated by the C-18 procedure than by the DAX-8 one, while humic-like compounds were similarly concentrated by both procedures. Furthermore, rainwater DOM extracted by the C-18 procedure was more representative of the global matrix, while DAX-8 preferentially extracted humic-like compounds.  相似文献

The resolution attainable with stimulated emission depletion (STED) microscopy greatly depends on the quality of the STED laser focus. So far, visual inspection of a measured STED focus has been the only convenient means of gauging the source of aberrations. Here we describe a method, requiring no instrument modifications, for obtaining an equivalent to the complex pupil function at the back aperture of the objective and show that it provides quantitative information about aberration sources (including aberrations induced by the objective or sample). We show the accuracy of this field representation to be sufficient for reconstructing the STED focus in three dimensions and determining corrective steps.  相似文献
