We use a unique multifocal multiphoton microscope to directly characterize the pulse in the focal plane of a high-NA objective using second-harmonic generation frequency-resolved optical gating (FROG). Because of the nature of the optical setup, femtosecond laser pulses of orthogonal polarization states are generated in the focal plane, each acquiring a different spectral dispersion. By applying an additional constraint on the phase extraction algorithm, we simultaneously extract both the gate and probe pulses from a single spectrogram with a FROG error of 0.016. 相似文献

By replacing the dispersive element in a zero-dispersion pulse shaper with a grism, we have constructed a quartic-phase-limited pulse shaper. We demonstrate compensation of 4500 fs2 without the use of a dynamic element in the pulse shaping line, which is approximately the amount of dispersion induced by a typical multiphoton microscope. We also demonstrate that detuning the pulse shaper to compensate for quadratic phase induces negligible spatial chirp, thereby maintaining a high-quality focal spot for a microscopy setup. 相似文献

We demonstrate the use of a simple tool to simultaneously visualize and characterize chromatic and spherical aberrations that are present in multiphoton microscopy.
