An extracellular β-glucanase secreted by Kluyveromyces marxianus was identified for the first time. The optimal conditions for the production of this enzyme were evaluated by response surface methodology. The optimal conditions to produce β-glucanase were a glucose concentration of 4 % (w/v), a pH of 5.5, and an incubation temperature of 35 °C. Response surface methodology was also used to determine the pH and temperature required for the optimal enzymatic activity. The highest enzyme activity was obtained at a pH of 5.5 and a temperature of 55 °C. Furthermore, the enzyme was partially purified and sequenced, and its specificity for different substrates was evaluated. The results suggest that the enzyme is an endo-β-1,3(4)-glucanase. After optimizing the conditions for β-glucanase production, the culture supernatant was found to be effective in digesting the cell wall of the yeast Saccharomyces cerevisiae, showing the great potential of β-glucanase in the biotechnological production of soluble β-glucan. 相似文献

Wheat allergens are responsible for symptoms in 60–70% of bakers with work-related allergy, and knowledge, at the molecular level, of this disorder is progressively accumulating. The aim of the present study is to investigate the panel of wheat IgE positivity in allergic Italian bakers, evaluating a possible contribution of novel wheat allergens included in the water/salt soluble fraction. The water/salt-soluble wheat flour proteins from the Italian wheat cultivar Bolero were separated by using 1-DE and 2-DE gel electrophoresis. IgE-binding proteins were detected using the pooled sera of 26 wheat allergic bakers by immunoblotting and directly recognized in Coomassie stained gel. After a preparative electrophoretic step, two enriched fractions were furtherly separated in 2-DE allowing for detection, by Coomassie, of three different proteins in the range of 21–27 kDa that were recognized by the pooled baker’s IgE. Recovered spots were analyzed by nanoHPLC Chip tandem mass spectrometry (MS/MS). The immunodetected spots in 2D were subjected to mass spectrometry (MS) analysis identifying two new allergenic proteins: a glucose/ribitol dehydrogenase and a 16.9 kDa class I heat shock protein 1. The article focuses on the optimization of the extraction process of biologically active compounds (BAC) from grape marc—a by-product of the wine industry. The influence of temperature, specifically 30 °C, 45 °C and 65 °C, and ethanol concentration in solutions, specifically 0–96% (v/v) on the extraction yield of polyphenols, flavonoids, tannins and anthocyanins, were investigated. The composition of individual polyphenols, anthocyanins and organic acids, antioxidant activity (DPPH and ABTS) and CIELab chromatic characteristics of the grape marc extracts (GME), were characterized. The microbiostatic and microbicidal effects in direct contact of GME with pathogenic microorganisms, Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, were determined in vitro. The influence of extraction parameters on the total polyphenol content (TPC), total flavonoid content (TFC), tannin content (TC), total anthocyanin content (TAC) and their interdependencies were studied using information analysis. A mathematical model was developed on cubic spline functions. The analysis of individual compounds showed the presence of a wide range of flavonoids (procyanidin B2, procyanidin B1, hyperoside and quercetin), flavones (catechin), hydroxybenzoic acid derivatives (gallic, protocatechuic, p-hydroxybenzoic acids, m-hydroxybenzoic acid, syringic acid), hydroxycinic acid derivatives and ferulic acid methyl ester. The malvidol-3-glucoside was the main anthocyanin identified in the extract. A high amount of tartaric acid was also found. GME showed significant antimicrobial activity against Gram-positive bacteria and lower activity against Gram-negative bacteria. 相似文献

Marine drugs are abundant in number, comprise of a diverse range of structures with corresponding mechanisms of action, and hold promise for the discovery of new and better treatment approaches for the management of several chronic diseases. There are huge reserves of natural marine biological compounds, as 70 percent of the Earth is covered with oceans, indicating a diversity of chemical entities on the planet. The marine ecosystems are a rich source of bioactive products and have been explored for lead drug molecules that have proven to be novel therapeutic targets. Over the last 70 years, many structurally diverse drug products and their secondary metabolites have been isolated from marine sources. The drugs obtained from marine sources have displayed an exceptional potential in the management of a wide array of diseases, ranging from acute to chronic conditions. A beneficial role of marine drugs in human health has been recently proposed. The current review highlights various marine drugs and their compounds and role in the management of chronic diseases such as cancer, diabetes, neurodegenerative diseases, and cardiovascular disorders, which has led to the development of new drug treatment approaches. 相似文献

Obesity and diabetes are the most demanding health problems today, and their prevalence, as well as comorbidities, is on the rise all over the world. As time goes on, both are becoming big issues that have a big impact on people’s lives. Diabetes is a metabolic and endocrine illness set apart by hyperglycemia and glucose narrow-mindedness because of insulin opposition. Heftiness is a typical, complex, and developing overall wellbeing worry that has for quite some time been connected to significant medical issues in individuals, all things considered. Because of the wide variety and low adverse effects, herbal products are an important hotspot for drug development. Synthetic compounds are not structurally diverse and lack drug-likeness properties. Thus, it is basic to keep on exploring herbal products as possible wellsprings of novel drugs. We conducted this review of the literature by searching Scopus, Science Direct, Elsevier, PubMed, and Web of Science databases. From 1990 until October 2021, research reports, review articles, and original research articles in English are presented. It provides top to bottom data and an examination of plant-inferred compounds that might be utilized against heftiness or potentially hostile to diabetes treatments. Our expanded comprehension of the systems of activity of phytogenic compounds, as an extra examination, could prompt the advancement of remedial methodologies for metabolic diseases. Extended multiplicative signal correction (EMSC) is a widely used preprocessing technique in infrared spectroscopy. EMSC is a model-based method favored for its flexibility and versatility. The model can be extended by adding constituent spectra to explicitly model-known analytes or interferents. This paper addresses the use of constituent spectra and demonstrates common pitfalls. It clarifies the difference between analyte and interferent spectra, and the importance of orthogonality between model spectra. Different normalization approaches are discussed, and the importance of weighting in the EMSC is demonstrated. The paper illustrates how constituent analyte spectra can be estimated, and how they can be used to extract additional information from spectral features. It is shown that the EMSC parameters can be used in both regression tasks and segmentation tasks. 相似文献

Single- and multi-shot ablation thresholds of gold films in the thickness range of 31-1400 nm were determined employing a Ti:sapphire laser delivering pulses of 28 fs duration, 793 nm center wavelength at 1 kHz repetition rate. The gold layers were deposited on BK7 glass by an electron beam evaporation process and characterized by atomic force microscopy and ellipsometry. A linear dependence of the ablation threshold fluence Fth on the layer thickness d was found for d ≤ 180 nm. If a film thickness of about 180 nm was reached, the damage threshold remained constant at its bulk value. For different numbers of pulses per spot (N-on-1), bulk damage thresholds of ∼0.7 J cm−2 (1-on-1), 0.5 J cm−2 (10-on-1), 0.4 J cm−2 (100-on-1), 0.25 J cm−2 (1000-on-1), and 0.2 J cm−2 (10000-on-1) were obtained experimentally indicating an incubation behavior. A characteristic layer thickness of Lc ≈ 180 nm can be defined which is a measure for the heat penetration depth within the electron gas before electron-phonon relaxation occurs. Lc is by more than an order of magnitude larger than the optical absorption length of α−1 ≈ 12 nm at 793 nm wavelength. 相似文献

The vertical profile of Saharan dust in the atmosphere is generally characterized by a large aerosol concentration in the mid troposphere, differently from the climatological distribution of other types of particles, that show a peak at the surface and a rapid decrease with height. Saharan dust is also characterized by particles of relatively large size of irregular shape, and variable values of the single scattering albedo (the ratio between radiation scattering and extinction). The dust's peculiar vertical distribution is expected to produce an effect on the calculation of the direct aerosol radiative forcing at the surface and at the top of the atmosphere. This effect is investigated by comparing estimates of aerosol direct visible radiative forcing at the surface and at the top of the atmosphere for dust vertical profiles measured in the Mediterranean, and for the climatological profile. The radiative forcing is estimated by means of an accurate radiative transfer model, and for the ocean surface. The sensitivity of the results on the solar zenith angle, aerosol optical depth, and aerosol absorption is also investigated. The aerosol radiative forcing at the surface shows a very small dependency on the aerosol vertical profile. At the top of the atmosphere, the radiative forcing is weakly dependent on the vertical profile (up to 10% variation on the daily average forcing) for low absorbing particles; conversely, it shows a strong dependency (the daily radiative forcing may vary up to 100%) for absorbing particles. The top of the atmosphere visible radiative forcing efficiency produced by dust having single scattering albedo <0.7 is higher by 4 W m−2 when the observed vertical profile instead of the standard profile is used in the calculations (i.e. it produces a lower cooling). For values of the single scattering albedo around 0.67, the sign of the forcing depends on the vertical profile. The influence of the vertical distribution on the radiative forcing is largest at small values of the solar zenith angle, and at short wavelengths. 相似文献

The detection of gravitational waves is a long-awaited event in modern physics and, to achieve this challenging goal, detectors
with high sensitivity are being used or are under development. In order to extract gravitational signals emitted by coalescing
binary systems of compact objects (neutron stars and/or black holes), from noisy data obtained by interferometric detectors,
the matched filter technique is generally used. Its computational kernel is a box-constrained global optimization problem
with many local solutions and a highly nonlinear and expensive objective function, whose derivatives are not available. To
tackle this problem, we designed a real-coded genetic algorithm that exploits characteristic features of the problem itself;
special attention was devoted to the choice of the initial population and of the recombination operator. Computational experiments
showed that our algorithm is able to compute a reasonably accurate solution of the optimization problem, requiring a much
smaller number of function evaluations than the grid search, which is generally used to solve this problem. Furthermore, the
genetic algorithm largely outperforms other global optimization algorithms on significant instances of the problem. 相似文献

The paper summarizes results achieved in the Institute of Radio Engineering and Electronics in the field of investigation
and fabrication of novel fiber-optic structures for evanescent-wave chemical sensing. Several approaches for increasing the
evanescent-wave sensitivity of multimode silica optical fibers to gaseous and liquid chemicals are shown. These approaches
make use of the decrease of the core diameter in sectorial and capillary sectorial fibers, modification of trajectories of
optical rays in the cores of inverted-graded-index fibers, tailoring of the refractive index of porous layers applied on silica
cores, an output mode filter preventing the detection of rays propagating at low axial angles or fibers bent in single or
multiple turns.
