Five polymeric architectures with a systematic increase in architectural complexity were synthesized by “click” reactions from a toolbox of functional linear polymers and small molecule linkers. The amphiphilic architectures ranged from a simple 3‐miktoarm star block copolymer to the more complex third generation dendrimer‐like block copolymer, consisting of polystyrene (PSTY) and polyacrylic acid (PAA). Micellization of these architectures in water at a pH of 7 under identical ionic strength gave spherical micelles ranging in size from 9 to 30 nm. Subsequent calculations of the PSTY core density, average surface area per PAA arm on the corona‐core interface, and the relative stretching of the PAA arms provided insights into the effect of architecture on the self‐assembly processes. A particular trend was observed that with increased architectural complexity the hydrodynamic diameter, radius of the core in the dry state and the aggregation number also increased with the exception of the third generation dendrimer. On the basis of these observations, we postulate that thermodynamic factors controlling self‐assembly were the entropic penalty of forming PSTY loops in the core counterbalanced by the reduction in repulsive forces through chain stretching. This results in a greater number of aggregating unimers and consequently larger micelle sizes. The junction points within the architecture also play an important role in controlling the self‐assembly process. The G3 dendrimer showed results contradictory to the aforementioned trend. We believe that the self‐assembly process of this architecture was dominated by the increased attractive forces due to stretching of the PSTY core chains to form a more compact core. © 2009 Wiley Periodicals, Inc. The investigation of multi‐site ligand–protein binding and multi‐step mechanisms is highly demanding. In this work, advanced NMR methodologies such as 2D 1H–15N line‐shape analysis, which allows a reliable investigation of ligand binding occurring on micro‐ to millisecond timescales, have been extended to model a two‐step binding mechanism. The molecular recognition and complex uptake mechanism of two bile salt molecules by lipid carriers is an interesting example that shows that protein dynamics has the potential to modulate the macromolecule–ligand encounter. Kinetic analysis supports a conformational selection model as the initial recognition process in which the dynamics observed in the apo form is essential for ligand uptake, leading to conformations with improved access to the binding cavity. Subsequent multi‐step events could be modelled, for several residues, with a two‐step binding mechanism. The protein in the ligand‐bound state still exhibits a conformational rearrangement that occurs on a very slow timescale, as observed for other proteins of the family. A global mechanism suggesting how bile acids access the macromolecular cavity is thus proposed.  相似文献

A novel nanocomposite film for sustained release of anionic ophthalmic drugs through a double-control process has been examined in this study. The film, made as a drug-loaded contact lens, consists principally of a polymer hydrogel of 2-hydroxyethyl methacrylate (HEMA), in whose matrix MgAl-layered double hydroxide (MgAl-LDH) nanoparticles intercalated with the anionic drug are well dispersed. Such nanocomposite films (hydrogel-LDH-drug) contained 0.6–0.8 mg of MgAl-LDH and 0.08–0.09 mg of the ophthalmic drug (ketorolac) in 1.0 g of hydrogel. MgAl-drug-LDH nanoparticles were prepared with the hydrodynamic particle size of 40–200 nm. TEM images show that these nanoparticles are evenly dispersed in the hydrogel matrix. In vitro release tests of hydrogel-LDH-drug in pH 7.4 PBS solution at 32 °C indicate a sustained release profile of the loaded drug for 1 week. The drug release undergoes a rapid initial burst and then a monotonically decreasing rate up to 168 h. The initial burst release is determined by the film thickness and the polymerization conditions, but the following release rate is very similar, with the effective diffusion coefficient being nearly constant (3.0 × 10−12 m2/s). The drug release from the films is mechanistically attributed to anionic exchange and the subsequent diffusion in the hydrogel matrix.  相似文献

Quantile regression is applied in two retail credit risk assessment exercises exemplifying the power of the technique to account for the diverse distributions that arise in the financial service industry. The first application is to predict loss given default for secured loans, in particular retail mortgages. This is an asymmetric process since where the security (such as a property) value exceeds the loan balance the banks cannot retain the profit, whereas when the security does not cover the value of the defaulting loan then the bank realises a loss. In the light of this asymmetry it becomes apparent that estimating the low tail of the house value is much more relevant for estimating likely losses than estimates of the average value where in most cases no loss is realised. In our application quantile regression is used to estimate the distribution of property values realised on repossession that is then used to calculate loss given default estimates. An illustration is given for a mortgage portfolio from a European mortgage lender. A second application is to revenue modelling. While credit issuing organisations have access to large databases, they also build models to assess the likely effects of new strategies for which, by definition, there is no existing data. Certain strategies are aimed at increasing the revenue stream or decreasing the risk in specific market segments. Using a simple artificial revenue model, quantile regression is applied to elucidate the details of subsets of accounts, such as the least profitable, as predicted from their covariates. The application uses standard linear and kernel smoothed quantile regression.  相似文献

Partial LAD regression uses the L 1 norm associated with least absolute deviations (LAD) regression while retaining the same algorithmic structure of univariate partial least squares (PLS) regression. We use the bootstrap in order to assess the partial LAD regression model performance and to make comparisons to PLS regression. By exploring a new mode of nickel‐catalyzed cross‐coupling, a method to directly transform both aromatic and aliphatic aldehydes into either esters or amides has been developed. The success of this oxidative coupling depends on the appropriate choice of catalyst and organic oxidant, including the use of either α,α,α‐trifluoroacetophenone or excess aldehyde. Mechanistic data that supports a catalytic cycle involving oxidative addition into the aldehyde C? H bond is also presented.  相似文献

We study the external and internal Zappa–Szép product of topological groupoids. We show that under natural continuity assumptions the Zappa–Szép product groupoid is étale if and only if the individual groupoids are étale. In our main result we show that the \(C^*\)-algebra of a locally compact Hausdorff étale Zappa–Szép product groupoid is a \(C^*\)-blend, in the sense of Exel, of the individual groupoid \(C^*\)-algebras. We finish with some examples, including groupoids built from \(*\)-commuting endomorphisms, and skew product groupoids.  相似文献
