Bacteriophage (phage) proteins have been analyzed previously with matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS). However, analysis of phage major capsid proteins (MCPs) has been limited by the ability to reproducibly generate ions from MCP monomers. While the acidic conditions of MALDI‐TOF MS sample preparation have been shown to aid in disassembly of some phage capsids, many require further treatment to successfully liberate MCP monomers. The findings presented here suggest that β‐mercaptoethanol reduction of the disulfide bonds linking phage MCPs prior to mass spectrometric analysis results in significantly increased MALDI‐TOF MS sensitivity and reproducibility of Yersinia pestis‐specific phage protein profiles. An AlPO4 zeotype has been prepared using the aromatic diamine 1,10-phenanthroline and some of its methylated analogues as templates. In each case the two template N atoms bind to a specific framework Al site to expand its coordination to the unusual octahedral AlO4N2 environment. Furthermore, using this framework-bound template, Fe atoms can be included selectively at this site in the framework by direct synthesis, as confirmed by annular dark field scanning transmission electron microscopy and Rietveld refinement. Calcination removes the organic molecules to give large pore framework solids, with BET surface areas up to 540 m2 g−1 and two perpendicular sets of channels that intersect to give pore space connected by 12-ring openings along all crystallographic directions.  相似文献

FeII, CoII and NiII complexes of two tetraazamacrocycles (1,4,8,11‐tetrakis(carbamoylmethyl)‐1,4,8,11‐tetraazacyclotetradecane ( L1 ) and 1,4,7,10‐tetrakis(carbamoylmethyl)‐1,4,7,10‐tetraazacyclododecane ( L2 ) show promise as paraCEST agents for registration of temperature (paraCEST=paramagnetic chemical exchange saturation transfer). The FeII, CoII and NiII complexes of L1 show up to four CEST peaks shifted ≤112 ppm, whereas analogous complexes of L2 show only a single CEST peak at ≤69 ppm. Comparison of the temperature coefficients (CT) of the CEST peaks of [Co( L2 )]2+, [Fe( L2 )]2+, [Ni( L1 )]2+ and [Co( L1 )]2+ showed that a CEST peak of [Co( L1 )]2+ gave the largest CT (?0.66 ppm oC?1 at 4.7 T). NMR spectral and CEST properties of these complexes correspond to coordination complex symmetry as shown by structural data. The [Ni( L1 )]2+ and [Co( L1 )]2+ complexes have a six‐coordinate metal ion bound to the 1‐, 4‐amide oxygen atoms and four nitrogen atoms of the tetraazamacrocycle. The [Fe( L2 )]2+ complex has an unusual eight‐coordinate FeII bound to four amide oxygen atoms and four macrocyclic nitrogen atoms. For [Co( L2 )]2+, one structure has seven‐coordinate CoII with three bound amide pendents and a second structure has a six‐coordinate CoII with two bound amide pendents.  相似文献

A study of the kinetics and mechanism of the reaction between the dinuclear Pt complex [(trans-PtCl(NH(3))(2))(2)(mu-NH(2)(CH(2))(6)NH(2))](2+) (1) and the 14-mer duplex 5'-d(ATACATG(7)G(8)TACATA)-3'.5'-d(TATG(25)TACCATG(18)TAT)-3' is reported. [(1)H,(15)N]-HSQC NMR was used to follow the reaction at 298 K, pH 5.4. The product is primarily the 5'-5' 1,4-interstrand cross-link between G(8) and G(18) bases and exists in two conformational forms. No evidence for the possible 1,2-intrastrand G(7)G(8) adduct was seen, confirming the preferential formation of interstrand cross-links by these dinuclear complexes. An initial electrostatic association of (15)N-1 with the duplex is indicated by changes in its (1)H/(15)N chemical shifts, followed by aquation of 1 to form the monoaqua monochloro species 2, with a rate constant of 4.00+/-0.03x10(-5) s(-1). Monofunctional binding to the duplex occurs primarily at G(8), the 3' base of the nucleophilic GG grouping, with a rate constant of 1.5+/-0.7 M(-1) s(-1). Changes in the (1)H/(15)N shifts indicate there is an electrostatic interaction between the unbound (PtN(3)Cl) group of the monofunctional adduct and the duplex. No peaks for a transient aquated monofunctional species are seen and closure of 3 to form the 1,4-G(8)G(18) interstrand cross-link (5) was treated as direct, with a rate constant of 4.47+/-0.06x10(-5) s(-1). The G(8)G(18) cross-link was confirmed from analysis of the NOESY NMR spectrum of the final product. Structural perturbations for the 1,4-interstrand cross-link extend over approximately four base-pairs and are similar to those found for a 1,4-interstrand cross-link with a shorter 8-mer -GTAC- sequence. A major distortion was evident for the 5'T (T(17)) adjacent to the platinated G(18), consistent with the findings from the use of chemical probes to investigate the conformation of 1,4-interstrand cross-links.  相似文献

We have developed a novel system for coupling reverse-phase (RP) and hydrophilic interaction liquid chromatography (HILIC) online in a micro-flow scheme. In this approach, the inherent solvent incompatibility between RP and HILIC is overcome through the use of constant-pressure online solvent mixing, which allows our system to perform efficient separations of both hydrophilic and hydrophobic compounds for mass spectrometry-based proteomics applications. When analyzing the tryptic digests of bovine serum albumin, ribonuclease B, and horseradish peroxidase, we observed near-identical coverage of peptides and glycopeptides when using online RP-HILIC—with only a single sample injection event—as we did from two separate RP and HILIC analyses. The coupled system was also capable of concurrently characterizing the peptide and glycan portions of deglycosylated glycoproteins from one injection event, as confirmed, for example, through our detection of 23 novel glycans from turkey ovalbumin. Finally, we validated the applicability of using RP-HILIC for the analysis of highly complex biological samples (mouse chondrocyte lysate, deglycosylated human serum). The enhanced coverage and efficiency of online RP-HILIC makes it a viable technique for the comprehensive separation of components displaying dramatically different hydrophobicities, such as peptides, glycopeptides, and glycans.  相似文献

The twin monomer 2,2′‐spirobi[4H‐1,3,2‐benzodioxasiline] ( 1 ) can be polymerized to nanostructured SiO2/phenolic‐resin composite material by thermally induced twin polymerization. Thermally induced twin polymerization represents a way to produce nanocomposites simply by thermal induction of twin monomers. Besides 1 , the thermal reaction of several related salicylic (2‐oxybenzylic) silicon molecules has been investigated. The thermal cleavage of the molecules is studied by using several trapping reagents (e.g., vinyl compounds). A significant occurrence of quinone methide adducts indicates that the thermal mechanism proceeds not only by a ring opening at the oxymethylene position, but also with the ortho‐quinone methide as a central or alternative intermediate. This is supported by product analyses of thermally initialized reactions of 1 and its substituted analogues as well as by quantum chemical calculations.  相似文献

Heteroleptic silver(I) nitrate complexes containing the bis[2-(diphenylphosphano)phenyl]ether (DPEphos) ligand and the heterocyclic thioamides pyridine-2(1H)-thione (py2SH), pyrimidine-2(1H)-thione (pymtH), 4,6-dimethylpyrimidine-2(1H)-thione (dmpymtH), 1,4,5,6-tetrahydropyrimidine-2-thione (thpymtH) or 1,3-imidazolidine-2-thione (imtH2) have been synthesized and characterized by IR and UV-Vis spectroscopy, elemental analyses and melting point determinations. The complexes can be obtained by the addition of the thioamide ligand to an AgNO3-diphosphane adduct in dichloromethane/ethanol solution. The molecular structure of [Ag(DPEphos)(py2SH)2]NO3 complex has been established by single-crystal X-ray diffraction. The structure features a tetrahedral silver(I) center with two phosphorus atoms from the chelating diphos ligand, and the exocyclic sulfur atom of two heterocyclic thioamide units. Intense blue-green emission is observed in the region 470-483 nm for all the complexes in the solid state and in solution at ambient temperature.  相似文献

Human macrophages incubated for prolonged periods with mildly oxidized LDL (oxLDL) or cholesteryl ester-rich lipid dispersions (DISP) accumulate free and esterified cholesterol within large, swollen lysosomes similar to those in foam cells of atherosclerosis. The cholesteryl ester (CE) accumulation is, in part, the result of inhibition of lysosomal hydrolysis due to increased lysosomal pH mediated by excessive lysosomal free cholesterol (FC). To determine if the inhibition of hydrolysis was long lived and further define the extent of the lysosomal defect, we incubated THP-1 macrophages with oxLDL or DISP to produce lysosome sterol engorgement and then chased with acetylated LDL (acLDL). Unlike oxLDL or DISP, CE from acLDL normally is hydrolyzed rapidly. Three days of incubation with oxLDL or DISP produced an excess of CE in lipid-engorged lysosomes, indicative of inhibition. After prolonged oxLDL or DISP pretreatment, subsequent hydrolysis of acLDL CE was inhibited. Coincident with the inhibition, the lipid-engorged lysosomes failed to maintain an acidic pH during both the initial pretreatment and subsequent acLDL incubation. This indicates that the alterations in lysosomes were general, long lived, and affected subsequent lipoprotein metabolism. This same phenomenon, occurring within atherosclerotic foam cells, could significantly affect lesion progression.  相似文献
