Debaryomyces hansenii is a polyol overproducing yeast that can have a potential use for upgrading lignocellulosic hydrolysates. Therefore, the establishment of its tolerance to metabolic inhibitors found in hydrolysates is of major interest. We studied the effects of selected aliphatic acids, phenolic compounds, and furfural. Acetic acid favored biomass production for concentrations <6.0 g/L. Formic acid was more toxic than acetic acid and induced xylitol accumulation (maximum yield of 0.21 g/g of xylose). All tested phenolics strongly decreased the specific growth rate. Increased toxicity was found for hydroquinone, syringaldehyde, and 4-methylcatechol and was correlated to the compound’s hydrophobicity. Increasing the amount of furfural led to longer lag phases and had a detrimental effect on specific growth rate and biomass productivity. 相似文献

Conditions have been established for the detection by chemical ionization mass spectrometry (CIMS) of compounds related to L -dopa (3-hydroxy-L -tyrosine) and L -tryptophan without derivatization. The least fragmentation of the parent ion occurs in positive ion CIMS when methylamine is used as reagent gas. The compounds can also be detected by negative ion CIMS using carbon tetrachloride as reagent gas. While the total ion current in the latter technique is lower than that obtained with positive ion CIMS, the background noise in the mass spectra of samples obtained from natural sources is greatly reduced. CIMS has been used to show the presence of aromatic amino acids in acid extracts of samples of two different classes of tumour without sample derivatization. 相似文献

The Brazilian agriculture activities have consumed about 288,000 tons of pesticides per year conditioned in about 107,000,000 packing with weight of approximately 23,000 tons. The discharge of empty plastic packing of pesticides can be an environmental concern causing problems to human health, animals, and plants if done without inspection and monitoring. The objective of this work is to study the ionizing radiation effect in the main pesticides used in Brazil for plastic packing decontamination. Among the commercial pesticides, chlorpyrifos has significant importance because of its wide distribution and extensive use and persistence. The radiation-induced degradation of chlorpyrifos in liquid samples and in polyethylene pack was studied by gamma radiolysis. Packing of high-density polyethylene (HDPE) three layer coextruded, named COEX, contaminated with chlorpyrifos, were irradiated using both a multipurpose Co-60 gamma irradiator and a gamma source with 5000 Ci total activity Gamma cell type. The chemical analysis of the chlorpyrifos was made using a gas chromatography associated to the Mass Spectrometry—GCMS from Shimadzu Model QP 5000. Gamma radiation was efficient for removing chlorpyrifos from the plastic packing, in all studied cases. 相似文献

Glucose utilization by lysine-producing fluoroacetate-sensitive mutants ofCorynebacterium glutamicum
A fluoroacetate-sensitive mutant was isolated fromCorynebacterium glutamicum, ATCC 21513, following mutagenesis with NTG. Batch fermentations show that in terms of growth kinetics, glucose utilization,
and lysine formation, there are significant differences between the mutant and the parent. The mutant’s specific growth rate
(0.22/h) is lower than that for the parent (0.34/h). Also, the yield expressed as lysine/glucose consumed does not alter as
a function of the glucose concentration for the mutant, and is about 0.22, whereas for the parent, this coefficient decreases
with increasing glucose concentration. The maximum specific rate of lysine production for the mutant is 1.3 g/L/h that is

Five species of halophytes were sampled in the salt marshes of the Tagus estuary, dried, ground and digested. They were further extracted with ethanol and the extracts passed through weak and strong cationic ion-exchange resins, purified through TLC and submitted to pyrolysis mass spectrometry and HPLC–ICP/MS. Arsenic content and hydride-forming arsenic species were verified, in each step, by GF–AA and HG–QFAA. A high content of arsenic was found in the samples of halophytes studied, both di- and tri-methylated arsenic compounds being present. A considerable fraction of this arsenic content seems to be refractory to hydride generation. Moreover, the arsenic fraction found seems to have the same ion-exchange behaviour as the refractory fractions formerly studied in estuarine water. A partial characterization of these structures by pyrolysis–GC–MS suggests the presence of arsenobetaine and arsenocholine compounds. Furthermore, HPLC–ICP/MS data seem to confirm the presence of these compounds. In addition, the latter hyphenated technique strongly suggests the presence of a number of other organoarsenicals including tetramethylarsonium (TMAs), trimethylarsine oxide (TMAO), cacodylate (DMA) and possibly an arsenosugar-type compound. © 1997 by John Wiley & Sons, Ltd. 相似文献

The aim of this work was to evaluate the influence of the purification process on the stability of Jatropha curcas biodiesel. The biodiesel was obtained using a variety of purification processes: three wet methods with different drying processes (vacuum oven, conventional oven and anhydrous sodium sulfate) and one dry method (purification with adsorbent magnesium silicate). Biodiesel was characterized through the analysis of carbon residue, acidity index, infrared and gas chromatography. The composition J. curcas oil indicated 56.3 % of unsaturated fatty acids and 43.7 % of saturated fatty acids. Jatropha oil presented high quantity of saturated acids, which are less susceptible at oxidation. The biodiesel sample that was chemically purified (PUsq) presented better purity, indicating be the process more efficient in remove the residues of synthesis. Thermogravimetric curves of purified biodiesel by wet method, PUsq, with chemical drying using anhydrous sodium sulfate, and PUsv, with vacuum drying, showed the highest initial decomposition temperatures, indicating higher thermal stability. The carbon residue and infrared analyses suggested that contamination by catalyst residue is a determining factor in reduction of the oxidative stability of biodiesel. The oxidative stability was evaluated using Rancimat and pressure differential scanning calorimetry. Biodiesel samples showing better oxidative stability were purified using PUsq and PUsv, which obtained stability of 6 h using the Rancimat technique, the minimum limit set by Brazilian legislation, without the addition of antioxidant, suggesting that these methods least influenced the stability of biodiesel. 相似文献
