The conformation of peptide and protein drugs in various microenvironments and the interaction with drug carriers such as liposomes are of considerable interest. In this study the influence of microenvironments such as pH, salt concentration, and surface charge on the secondary structure of a model protein, lysozyme, either in solution or entrapped in liposomes with various molar ratios of phosphatidylcholine (PC):cholesterol (Chol) was investigated. It was found that entrapment efficiency was more pronounced in negatively charged liposomes than in non-charged liposomes, which was independent of Chol content and pH of hydration medium. The occurrence of aggregation, decrease in zeta potential, and alteration of 31P NMR chemical shift of negatively charged lysozyme liposomes compared to blank liposomes suggested that the electrostatic interaction plays a major role in protein–lipid binding. Addition of sodium chloride could impair the neutralizing ability of positively charged lysozyme on negatively charged membrane via chloride counterion binding. Neither lysozyme in various buffer solutions with sodium chloride nor that entrapped in liposomes showed any significant change in their secondary structures. However, significant decrease in α-helical content of lysozyme in non-charged liposomes at higher pH and salt concentrations was discovered. 相似文献

Excited state potential energy hypersurfaces of 7H-furo[3,2-g][1]benzopyran-7-one (psoralen) have been explored employing (time-dependent) Kohn-Sham density functional theory. At selected points, we have determined electronic excitation energies and electric dipole (transition) moments utilizing a combined density functional/multireference configuration interaction method. Spin-orbit coupling has been taken into account employing an efficient, non-empirical spin-orbit mean-field Hamiltonian. Franck-Condon factors have been computed for vibrational modes with large displacements in the respective Dushinsky transformations. The simulated band spectra closely resemble experimental band shapes and thus validate the theoretically determined nuclear structures at the S(0), S(1), and T(1) minima. In the S(1) (pi(HOMO)-->pi*(LUMO)) state, the lactone bond of the pyrone ring is significantly elongated. From excited vibrational levels of the S(1) state a conical intersection between a (pi-->sigma*) excited state and the electronic ground state may be energetically accessible. Fast non-radiative decay via this relaxation pathway could explain the low fluorescence quantum yield of psoralen. The T(1) (pi(HOMO-1)-->pi*(LUMO)) exhibits a diradicaloid electronic structure with a broken C(5)-C(6) double bond in the pyrone ring. A variational multireference spin-orbit configuration interaction procedure yields a phosphorescence lifetime of 3 s, in excellent agreement with experimental estimates. 相似文献

ZrO2 Inorganic NanoPatterns have been prepared on conductive electrodes by the direct block copolymer templating method associated with sol–gel dip-coating followed by thermal treatment. They have then been successfully utilised to direct the co-electrodeposition of FePt nanoparticles into the 2D well-ordered arrays of nanoelectrodes with controlled dimension and periodicity. By this method, the FePt nanodomains are confined and stabilised into the well-ordered cavities of the ZrO2 membrane. While this system constitutes the first steps towards high-density magnetic storage media, the present fully bottom-up method can be easily scaled up and generalised to infinite nanocomposite combinations. 相似文献

Substrate‐competitive kinase inhibitors represent a promising class of kinase inhibitors, however, there is no methodology to selectively identify this type of inhibitor. Substrate activity screening was applied to tyrosine kinases. By using this methodology, the first small‐molecule substrates for any protein kinase were discovered, as well as the first substrate‐competitive inhibitors of c‐Src with activity in both biochemical and cellular assays. Characterization of the lead inhibitor demonstrates that substrate‐competitive kinase inhibitors possess unique properties, including cellular efficacy that matches biochemical potency and synergy with ATP‐competitive inhibitors. 相似文献

We describe the preparation of the first crystalline compounds based on arylboronate ligands PhB(OH)(3)(-) coordinated to metal cations: [Ca(PhB(OH)(3))(2)], [Sr(PhB(OH)(3))(2)]·H(2)O, and [Ba(PhB(OH)(3))(2)]. The calcium and strontium structures were solved using powder and single-crystal X-ray diffraction, respectively. In both cases, the structures are composed of chains of cations connected through phenylboronate ligands, which interact one with each other to form a 2D lamellar structure. The temperature and pH conditions necessary for the formation of phase-pure compounds were investigated: changes in temperature were found to mainly affect the morphology of the crystallites, whereas strong variations in pH were found to affect the formation of pure phases. All three compounds were characterized using a wide range of analytical techniques (TGA, IR, Raman, XRD, and high resolution (1)H, (11)B, and (13)C solid-state NMR), and the different coordination modes of phenylboronate ligands were analyzed. Two different kinds of hydroxyl groups were identified in the structures: those involved in hydrogen bonds, and those that are effectively "free" and not involved in hydrogen bonds of any significant strength. To position precisely the OH protons within the structures, an NMR-crystallography approach was used: the comparison of experimental and calculated NMR parameters (determined using the Gauge Including Projector Augmented Wave method, GIPAW) allowed the most accurate positions to be identified. In the case of the calcium compound, it was found that it is the (43)Ca NMR data that are critical to help identify the best model of the structure. 相似文献

We present three formulas for calculating intersystem crossing rates in the Condon approximation to the golden rule by means of a time-dependent approach: an expression using the full time correlation function which is exact for harmonic oscillators, a second-order cumulant expansion, and a short-time approximation of this expression. While the exact expression and the cumulant expansion require numerical integration of the time correlation function, the integration of the short-time expansion can be performed analytically. To ensure convergence in the presence of large oscillations of the correlation function, we use a Gaussian damping function. The strengths and weaknesses of these approaches as well as the dependence of the results on the choice of the technical parameters of the time integration are assessed on four test examples, i.e., the nonradiative S(1) ? T(1) transitions in thymine, phenalenone, flavone, and porphyrin. The obtained rate constants are compared with previous results of a time-independent approach. Very good agreement between the literature values and the integrals over the full time correlation functions are observed. Furthermore, the comparison suggests that the cumulant expansion approximates the exact expression very well while allowing the interval of the time integration to be significantly shorter. In cases with sufficiently high vibrational density of states also the short-time approximation yields rates in good agreement with the results of the exact formula. A great advantage of the time-dependent approach over the time-independent approach is its excellent computational efficiency making it the method of choice in cases of large energy gaps, large numbers of normal modes, and high densities of final vibrational states. 相似文献
