The introduction of Asn-linked glycans to nascent polypeptides occurs in the lumen of the endoplasmic reticulum of eukaryotic cells. After the removal of specific sugar residues, glycoproteins acquire signals in the glycoprotein quality control (GPQC) system and enter the folding cycle composed of lectin-chaperones calnexin (CNX) and calreticulin (CRT), glucosidase II (G-II), and UDP-Glc:glycoprotein glucosyltransferase (UGGT). G-II initiates glycoproteins’ entry and exit from the cycle, and UGGT serves as the “folding sensor”. This account summarizes our effort to analyze the properties of enzymes and lectins that play important roles in GPQC, especially those involved in the CNX/CRT cycle. To commence our study, general methods for the synthesis of high-mannose-type glycans and glycoproteins were established. Based on these, various substrates to analyze components of the GPQC were created, and properties of CRT, G-II, and UGGT have been clarified.  相似文献

A high-speed counter-current chromatography (HSCCC) technique in a preparative scale has been applied to separate and purify cordycepin from the extract of Cordyceps militaris(L.) Link by a one-step separation. A high efficiency of HSCCC separation was achieved on a two-phase solvent system of n-hexane-n-butanol-methanol-water (23:80:30:155, v/v/v/v) by eluting the lower mobile phase at a flow rate of 2 ml/min under a revolution speed of 850 rpm. HSCCC separation of 216.2 mg crude sample (contained cordycepin at 44.7% purity after 732 cation-exchange resin clean-up) yielded 64.8 mg cordycepin with purity of 98.9% and 91.7% recovery. Identification of the target compound was performed by UV, IR, MS, (1)H NMR and (13)C NMR.  相似文献

Recently, graphene sheet is one of interesting systems to realize novel electronic properties. Especially, interaction between graphene and adsorbed oxygen molecule is very important to control electronic condition. In this paper, we employed some aromatic hydrocarbons as simple systems of graphene sheet and ab initio MO calculations were carried out to investigate inter-molecular interaction. It is found that not triplet but singlet O2 molecule have potential of chemisorption onto graphene surface. From the calculated potential energy surface (PES) for distance between benzene and O2 molecules, meta-stable structure is found at about 1.5 Å with potential barrier. In the optimized structure of its meta-stable state, structural strain can be relaxed through bending of planer benzene ring. Its energy is estimated at 70.10 kcal/mol for benzene. We also estimated the strain effects for naphthalene and pyrene molecules as larger case of graphene and they were 80.85 and 72.45 kcal/mol, respectively.  相似文献

Background

The ubiquitin ligase COP1, COnstitutively Photomorphogenic 1, functions in many biological responses in mammalian cells, but its downstream pathway remains unclear.

Results

Here, we identified FIP200, a key regulator of mammalian autophagy, as a novel COP1-interacting protein by yeast two-hybrid screening. The interaction was confirmed by a GST-pulldown assay. Split-GFP analysis revealed that interaction between COP1 and FIP200 predominantly occurred in the cytoplasm and was enhanced in cells treated with UV irradiation. Different forms of FIP200 protein were expressed in cultured mammalian cells, and ectopic expression of COP1 reduced one of such forms.

Conclusions

These data suggest that COP1 modulates FIP200-associated activities, which may contribute to a variety of cellular functions that COP1 is involved in.  相似文献

The reaction of 3,4-dichlorotetragermetene derivative 2 with Na(2)[Fe(CO)(4)] in THF produced a (tetragermacyclobutadiene)tricarbonyliron complex, [{η(4)-((t)Bu(2)MeSi)(4)Ge(4)}]Fe(CO)(3)4, which has a slightly folded Ge(4) ring perhaptocoordinated to the Fe center. Structural and spectral characteristics of 4 show a remarkable π-donating ability of the tetragermacyclobutadiene ligand toward the transition metal, surpassing that of tetrasilacyclobutadiene and cyclobutadiene ligands. Reduction of 2 with KC(8) resulted in exclusive formation of the dipotassium salt of the tetragermacyclobutadiene dianion derivative 3(2-)·[K(+)(thf)(2)](2), representing a rare example of a 6π-electron compound that, on the basis of its structural and magnetic properties, was recognized as a nonaromatic species. Reaction of 3(2-)·[K(+)(thf)(2)](2) with CpCoI(2)(PPh(3)) produced a (cyclopentadienyl)(tetragermacyclobutadiene)cobalt complex, [{η(4)-((t)Bu(2)MeSi)(4)Ge(4)}]CoCp 7, as the first example of a sandwich compound featuring an all-germanium-containing cyclic polyene ligand.  相似文献

Kinetically protected 2‐silyl‐1,3‐diphosphapropenes that bear both sp2‐ and sp3‐type phosphorus atoms were employed in the preparation of gold complexes. The structural properties of the 1,3‐diphosphapropene digold(I) complexes were characterized by spectroscopic and crystallographic analyses, which revealed unique aurophilic interactions and conformational properties of the ligand. The 2‐silyl‐1,3‐diphosphapropene‐bis(chlorogold) complexes catalyzed cycloisomerization reactions of 1,6‐enyne derivatives even in the absence of silver co‐catalyst, and were able to be recovered after the reaction. The catalytic activity of the digold complexes primarily depended on the sp2‐type phosphorus atom and the silyl group, and could be tuned by the sp3‐phosphino group. Additionally, results on the catalytic activity of the digold complex in the presence and absence of silver salts showed considerable differences.  相似文献

We have developed a novel synthetic method that enables us to easily synthesize metal‐capsulated carbon nanotubes (CNTs) in a laboratory by using a combined technology of electrospinning‐metallization and microwave heating. These techniques greatly shorten the time for the synthesis of the CNTs in comparison with the conventional methods. TEM analysis confirmed a successful formation of the CNTs, and the resulting CNTs were multi‐walled and found to be about 25–100 nm in diameters. The products prepared by heating at 600 and 900°C exhibited less‐developed and strongly curved CNTs, whereas the products prepared by heating at 700 and 800°C relatively well‐developed long CNTs. The permeability of carbon dioxide (CO2) through imidazolium-based ionic liquid membranes was measured by a sweep gas method. Six species of ionic liquids were studied in this work as follows: [emim][BF4], [bmim][BF4], [bmim][PF6], [bmim][Tf2N], [bmim][OTf], and [bmim][dca]. The ionic liquids were supported with a polyvinylidene fluoride porous membrane. The measurements were performed at T = (303.15 to 343.15) K. The partial pressure difference between feed and permeate sides was 0.121 MPa. The permeability of the CO2 increases with temperature for the all ionic liquid species. Base on solution diffusion theory, it can be explained that the diffusion coefficient of CO2 in an ionic liquid affects the temperature dependence more strongly than the solubility coefficient. The greatest permeability was obtained with the [bmim][Tf2N] membrane. The membrane of [bmim][PF6] presents the lowest permeability.The separation coefficient between CO2 and N2 through the ionic liquid membranes was also investigated at the volume fraction of CO2 at feed side 0.10. The separation coefficient decreases with the increase of temperature for the all ionic liquid species. The membrane of [emim][BF4] and [bmim][BF4] gives the highest separation coefficient at constant temperature. The lowest separation coefficient was obtained from [bmim][Tf2N] membrane which presents the highest permeability of CO2.  相似文献

The extraction behavior of uranium(VI), plutonium(IV) and some fission products like zirconium(IV), ruthenium(III) and europium(III) from 3.5M nitric acid with -irradiated organic phase pre-equilibratedn-dodecane solutions of dihexyl derivatives of hexanamide (DHHA), octanamide (DHOA) and decanamide (DHDA) has been investigated as a function of absorbed dose upto 184·104 Gy. The results indicate that the extraction of uranium(VI) decreases gradually with dose upto 72·104 Gy and becomes almost constant thereafter, while, the extraction of plutonium(IV) decreases upto a dose of 20·104 Gy and then increases rapidly up to a dose of 82·104 Gy indicating synergistic effects of radiolytic products formed at higher doses. Extraction of zirconium(IV) increases gradually upto a dose of 72·104 Gy. Europium(III) does not get extracted with any of these amides in the entire dose range (0–184·104 Gy) studied, however, ruthenium shows insignificant increase in extraction with dose. The decrease inD values noticed in the case of plutonium and zirconium after the dose of 72·104 Gy which was attributed to the third phase formation and emulsification. Infrared studies confirm the final products of radiolysis as the respective amines and carboxylic acids. The degraded amide contents have been estimated by quantitative IR spectrophotometric technique. Extraction data obtained for uranium(VI) and plutonium(IV) with TBP/n-dodecane system have also been compared under similar experimental conditions.  相似文献

Dithia‐tetrahomodiaza‐calix[4]arenes were synthesized by the cyclization reactions of bis(3‐(chloro‐methyl)‐2‐hydroxyphenyl)sulfide with cystine peptides in moderate yields. Conformational analysis of the macrocycles by using nmr spectroscopy reveled that the cyclophanes adopt a cone‐like form as a preferable conformation and the cystine bridge moiety is incorporated in the cavity. The calixarene analogs can extract transion metals such as Zn2+ and Cu2+ ions from an aqueous phase into chloroform.  相似文献
