Pulmonary hypertension (PH) in sickle cell anemia (SCA) is characterized by decreased nitric oxide bioavailability that might, in part, be related to oxidative stress. Oxidative post-translational modifications of plasma proteins may serve as hallmarks of disease severity and could result in altered protein function and structure. We hypothesized that serum albumin in patients with PH of SCA undergoes oxidative post-translational modification and that this modification may reflect important mediators of disease pathogenesis that are common to both idiopathic pulmonary arterial hypertension (IPAH) and PH of SCA. To explore this hypothesis, we studied albumin purified from the plasma of patients in four subject groups: SCA and PH, SCA steady-state without PH, IPAH, and normal volunteers. Purified albumin was analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) and liquid chromatography/tandem mass spectrometry (LC/MS/MS). Using MALDI-TOFMS, we identified that an ion corresponding to a malondialdehyde (MDA)-modified albumin peptide was differentially present in patients with IPAH and PH of SCA. These results were confirmed by dot-blotting and Western analysis. We localized the site of MDA modification to albumin residue K159 using LC/MS/MS. Thus, we have identified an MDA modification of serum albumin that appears to be a common link between PH of SCA and IPAH. This finding supports the notion that oxidative stress modulates the pathogenesis of PH of SCA and suggests that this and other post-translational modifications may be important biomarkers of disease. 相似文献

Novel copper(II)-nitroxide complexes exhibiting a spin-transition-like behavior have been prepared and characterized. They include meso, chiral, and racemic 2-(3-pyridyl)-nitronyl nitroxides differently substituted in positions 4 and/or 5 by ethyl groups and pyrimidyl nitroxides. Depending on the stoichiometry of the reaction, tetranuclear and binuclear complexes were obtained whose structures are cyclic. The tetranuclear species, which include two intracyclic and two exocyclic metal sites, are similar to the previously reported complex of the tetramethylated analogue, while the binuclear complexes involve only endocyclic metal ions and have uncoordinated N-oxyl groups. The tetranuclear complexes exist as two isomers depending on the temperature of crystallization: at room temperature, N-oxyl ligand coordination is axial-axial, while it is axial-equatorial at low temperature. Unexpectedly, this isomerism concerns N-oxyl bonding to the exocyclic metal centers for the derivatives of 4,5-diethyl-substituted ligands while it involves the endocyclic metal site in the complex of the monoethylated ligand, which converts reversibly from a high-spin state to a low-spin state, as observed for the complex of the tetramethylated ligand. Binuclear complexes are diamagnetic at room temperature but convert to a paramagnetic state on warming (90-110 degrees C); the transition is irreversible and sharp. 相似文献

Endothermic ion-molecule reactions in a tandem mass spectrometer have been used for a number of years for determining thermodynamic quantities, such as heats of formation and proton affinities, for gaseous ions. Recently, the reactive, endothermic collision has been exploited as an analytical technique for the structural analysis of peptides and other biomolecules. The technique is based upon the endothermic transfer of protons associated with amide bonds to ammonia. This reaction proceeds via a long-lived collision complex. When additional beam energy is supplied, other dissociation channels are opened up, leading to the production of sequence ions for the mass-selected, protonated analyte that are normally observed in high energy collision-induced dissociation spectra. The advantage, however, is that such spectra can be produced at very low beam energies. Elemental sulphur, a common constituent of marine sediments, has been shown to give dialkyl sulphides with the Grignard reagents commonly used to derivatize alkyltin species before their determination by gas chromatography with flame photometric detection (GC–FPD). Further, it has been demonstrated that even with the red filter for 610 nm (normally used for organotin compounds) fitted to the detector, sulphur compounds do give rise to an emission signal, which may be mistaken for tin emission from a pentylated or propylated alkyltin compound, as the respective retention times are in some cases quite close. © 1997 by John Wiley & Sons, Ltd. 相似文献

Many studies have been conducted on the impact of animal feed on isoflavones and their metabolite concentrations in bovine milk, but few studies have focused on the development and validation of analytical protocols for quantifying these compounds in biological matrices other than milk and plants. The purpose of this study was to develop a method that would enable four isoflavones and equol in cows’ feces and digestive fluids to be quantified simultaneously. The method is based on aglycones released by methanolic ultrasound-assisted extraction, followed by enzymatic hydrolysis and high-performance liquid chromatography tandem–mass spectrometry analysis. The sample preparation was optimized using the Box–Behnken design. The selected extraction conditions were 80°C, 10?min, and 50% methanol for digestive fluids and 70°C, 35?min, and 60% methanol for feces. For hydrolysis, the selected conditions were 37°C, 1?h, and a pH of 6 for both matrices. The analytical method showed a good linear regression model ranging from 5 to 125?ng?mL?1. Both inter- and intraday accuracy (≤8.5 and ≤12.3%) and precision (≤11.1 and ≤15.2%) were suitable. No matrix effects were found. There was good repeatability and extract stability for at least 4 days of storage at???20 and 6°C. All recoveries were in the acceptable range of 70–120% for both matrices, except for biochanin A in feces, where the value was approximately 43%. This sensitive and reliable method will be useful for monitoring the passage of isoflavones and equol in the digestive system of ruminants. 相似文献
