Versatile peroxidases (VP) constitute a new class of high redox potential fungal enzymes that are able to degrade lignin and large substrate molecules. These enzymes catalyze the oxidation of substrates at an exposed tryptophan radical formed by a long-range electron transfer mechanism to heme following the activation by H2O2. In a previous paper, it was demonstrated using electron paramagnetic resonance (EPR) and electron-nuclear double resonance experiments on wild-type VP that Trp164 was the radical site and that it was in a hydrogen-bonded neutral form. In this paper, the W164Y variant is analyzed and it is shown that also the variant is able to form the so-called Compound I (VPI) in the form of protein radical, although in different yields with respect to the wild type. The X-band EPR experiments in combination with density functional theory/polarizable continuum model calculations show that the W164Y mutant is able to form a neutral radical on Tyr164 residue, after activation by H2O2, but in contrast to Trp164, tyrosine is not expected to be hydrogen bonded.  相似文献

Academic research and the financial industry have recently shown great interest in Machine Learning algorithms capable of solving complex learning tasks, although in the field of firms' default prediction the lack of interpretability has prevented an extensive adoption of the black-box type of models. In order to overcome this drawback and maintain the high performances of black-boxes, this paper has chosen a model-agnostic approach. Accumulated Local Effects and Shapley values are used to shape the predictors' impact on the likelihood of default and rank them according to their contribution to the model outcome. Prediction is achieved by two Machine Learning algorithms (eXtreme Gradient Boosting and FeedForward Neural Networks) compared with three standard discriminant models. Results show that our analysis of the Italian Small and Medium Enterprises manufacturing industry benefits from the overall highest classification power by the eXtreme Gradient Boosting algorithm still maintaining a rich interpretation framework to support decisions.  相似文献

The aim of the present study was to evaluate the effects of supplementation with a fixed combination of citicoline 500 mg, homotaurine 50 mg, and vitamin E 12 mg (CIT/HOMO/VITE) on contrast sensitivity and visual-related quality of life in patients with primary open-angle glaucoma (POAG) in mild stage. This was a multicenter, observational, cross-over, short-term, pilot study on POAG patients with stable controlled intraocular pressure (IOP). Patients were randomly assigned to Group 1 (current topical therapy for 4 months and then current topical therapy plus CIT/HOMO/VITE for 4 months) or Group 2 (CIT/HOMO/VITE in addition to current topical therapy for 4 months and then topical therapy alone for 4 months). Best-corrected visual acuity, IOP, visual field, and the Spaeth/Richman contrast sensitivity (SPARCS) test score were recorded at baseline and after 4 and 8 months. The Glaucoma Quality of Life-15 (GQL-15) questionnaire was administered at each check time. Forty-four patients were assigned to Group 1 and 65 to Group 2. Over the follow-up period, there were no significant changes in IOP or visual field findings, whereas SPARCS and GQL-15 findings significantly varied from baseline, both being improved in subjects treated with CIT/HOMO/VITE fixed combination. These results demonstrate that a daily intake of a fixed combination of citicoline, homotaurine, and vitamin E in addition to the topical medical treatment significantly increased the total score of the contrast sensitivity test and the quality of life in patients with POAG.  相似文献

The conformation of chiral 2,2′-dinitro-3,3′-bithienyl-4,4′-dicarboxylic acid (Ia), its dimethylester (Ib), and analogous selenium derivatives Ic and Id was investigated by means of induced cholesteric mesophases. A parallel investigation on the diester (Ib) and on other model 3,3′-bithienyls was carried out with dipole moment measurements. The conclusions reached with the two independent methods for derivative Ib are in excellent agreement and indicate, if a single conformation is assumed, a preferred cisoid conformation with a dihedral angle somewhat smaller than 90°. Alternatively, if the existence of both cisoid and transoid conformations having supplementary dihedral angles is assumed, the data indicate a higher population for the cisoid form.  相似文献

There is chemical and kinetic evidence that the mechanism of action of mandelate racemase occurs via two acid/base catalysts, one to abstract a proton from the -carbon and the second to deliver it back to the opposite face of the chiral center. Since the mechanistic details are not known, a few hypotheses have been put forward. Therefore we examine the viability of both concerted and sequential mechanisms on proton abstraction from the -carbon of mandelate, either without taking into account environmental effects or with the addition to the model system of several functional groups of the residues belonging to the active site, in order to restrain the partners to particular positions without actually imposing arbitrary constraints. The importance of the environment in screening the most highly charged groups in the substrate and in facilitating the -carbon-bound proton abstraction is evident. From the perusal of the geometries of the system along the reaction energy profile, it seems that there is no tendency toward a concerted mechanism: the proton delivery from mandelate to Lys occurs and subsequently the extra proton on protonated histidine is in turn delivered to mandelate.  相似文献

Reduction of 1,2-diaryl-substituted ethenes with Na metal in dry THF allowed easy access to a variety of 1,2-diaryl-1,2-disodiumethanes. These diorganometallic intermediates were elaborated into the corresponding 1,2-diarylethanes (aqueous work up), or cycloalkylated with 1,3-dichloropropanes. The last reaction led to a highly diastereoselective synthesis of trans-1,2-diaryl-substituted cyclopentanes.  相似文献

We present an extension of our semiempirical floating occupation MO-CI approach for the determination of ground and excited state potential energy surfaces of interest in photochemistry. The QM/MM variant of the method, which allows for electrostatic and van der Waals interactions between the QM and MM subsystems, is supplemented with a treatment of covalent interactions based on Antes and Thiels connection atom approach. We concentrate on the correct treatment of electrostatic interactions concerning the connection atom, on the specific requirements for the representation of excited states, and on the transferability of the optimal parameters. We show the viability of the method with four examples of connection atoms: S in a thioether bridge, acylic C, aliphatic C, and N in a peptide. The results obtained with the QM/MM treatment compare well with all-QM results of the same level.  相似文献

The crystal and molecular structure of Ti(n5-C5H4CH3)2S5has been determined by X-ray diffraction studies. The substance crystallizes in the monoclinic crystal system [a = 6.8642(5), b = 16.507(1), c = 13.074(1) Å, β = 82.407(3)°, space group P21/n, Z = 4]. The geometry about the titanium atom is a distorted tetrahedron, with a (centroid)-Ti-(centroid) angle of 131.29° and a S? Ti? S angle of 93.38°. The six-membered ring TiS5 has a cyclohexane-like chair configuration. The structural results are compared to those for similar type titanium complexes.  相似文献
