The interaction of hybrid lipid/gramicidin A (gA) monolayers with dextran sulfate (DS) and the effect of this interaction on ion transfer at a liquid-liquid interface is reported. The interfacial and physicochemical properties are studied with Langmuir-Blodgett (LB) and electrochemical techniques. The results obtained from compression isotherms demonstrate that the interactions between the different species in the hybrid monolayer vary according to the chemical nature of the lipid (hydrocarbon region and charge of the head group). Interfacial capacitance measured with AC voltammetry indicates that the DS chains form a rather flat and compact layer when adsorbed to either zwitterionic or negatively charged phospholipid monolayers, and that calcium, even at low concentrations, interacts with the monolayers. These results are successfully described by a model based on the solution of the Poisson-Boltzmann equation in the interfacial region. Ion transfer and interactions with the lipid/gA/DS-modified monolayers were also studied with electrochemical techniques. Admittance data show that although the studied ions are not using gA channels for the transfer through the lipid membranes, the incorporation of gA in the lipid domain and the adsorption of DS at the interface have a significant effect on ion transfer across the monolayers. This effect can be explained as a consequence of the modified surface charge and of the compactness of the lipid domain due to its interaction with gA and to calcium and DS adsorption at the interface. The ion-transfer rate, therefore, depends on the composition of the monolayer and the chemical nature of the ion.  相似文献

The fact that a titanium screw can be implanted into the mastoid portion of the human skull, at the same time establishing a permanent, reaction-free skin penetration, has made it possible to attach a new bone conduction hearing aid directly to the skull. To understand and improve this new method of bone stimulation, the mechanical point impedance of the titanium screw-skull system was measured. The conventional point impedance of the skin-covered mastoid portion of the temporal bone was also measured and the difference in magnitude between the two impedances was calculated. An impedance head (Brüel & Kjaer 8001) and an FFT analyzer (Hewlett-Packard 5423) were used for mechanical point impedance measurements. Seven patients have been investigated. The magnitude of the impedance for the screw-skull system was found to be generally between 10 and 30 dB higher than that for the conventional skin-covered mastoid bone. One conclusion is that the conventional point impedance of the skin-covered mastoid portion of the human skull is essentially due to the properties of the skin and subcutaneous soft tissue. Another conclusion is that a much lower stimulation velocity is needed, with skin penetration, to produce a given hearing sensation.  相似文献

A method is proposed for direct optical comparison of the three-dimensional shapes of objects by light-in-flight speckle holography. The basic idea of this technique is to use an ultrashort laser pulse with a short coherence length to produce interference patterns that present a single contouring of the object. A new approach for the control and interruption of enzymatic reactions via selective enzyme immobilization has been developed. The technique was exemplified by the use of three model enzymes with the corresponding macromolecular substrates: α-amylase/starch, trypsin/ insoluble collagen, and alkaline phosphatase/plasmid DNA. Prior to incubation with its substrate, each enzyme was provided withde novo thiol-groups by a two-step reaction involvingN-succinimidyl-3-(2-pyridyldithio) propionate (SPDP) and DTT. The chemical modification was achieved such that at least 80% of the native enzyme activity was preserved in all cases. In order to interrupt rapidly the reactions in which the enzymes were used, the modified enzyme was immobilized by reaction via its thiol groups on a thiolsulfinate-agarose derivative. The gel-bound enzyme could then be easily removed from unreacted substrate and product by filtration or centrifugation. Comparative studies showed that the immobilized enzymes had much lower activities in the reactions studied than the corresponding soluble ones. The potential for enzyme reuse was also demonstrated with the a-amylase derivatives, which were quantitatively released and eluted in fully active form from the agarose. We have shown that it is possible to achieve practically complete enzyme immobilization in short times and thus to control the progress of the reactions. Because of its simplicity and high efficiency, this approach may represent an interesting alternative for biotechnological processes involving macromolecular or solid substrates.  相似文献

A novel MS-based analytical method for simultaneous analysis of the antiviral drugs acyclovir, its metabolite 9-carboxymethoxymethylguanine, ganciclovir, and penciclovir in human serum is described. These antiviral drugs are active against herpes virus infections. Acyclovir and penciclovir are regarded as safe and effective medicines with mild side effects such as headache and gastrointestinal discomfort, and ganciclovir is regarded as more toxic and is known to cause, for example, bone marrow suppression. Acyclovir’s main metabolite 9-carboxymethoxymethylguanine is a presumptive neurotoxin and should be monitored in patients with impaired renal function or in cases with neurotoxic symptoms. A sample was prepared using protein precipitation with 1% formic acid in methanol containing isotopically labeled internal standard. Chromatographic separation on a biphenyl column and mass spectrometric detection were performed in multiple reaction monitoring (MRM) mode on a Xevo TQ-S micro with ESI in positive ion mode, within 3 min. Inter-day assay accuracies for the quality controls varied between 95 and 104% and intra-day assay between 93 and 105%. Inter-day and intra-day assay imprecision for the quality controls ranged between 1.4 and 4.2% and 1.7 and 6.5% respectively. The lower limit of quantification for all four substances was 0.156 μmol/L. It is an accurate and reproducible method for therapeutic drug monitoring.  相似文献

Recently, Carlsson and Osipov reported on observations of the rotation of smectic layers under certain experimental conditions, and a dynamic theory describing such rotations was presented [Carlsson, T., and Osipov, M. A., 1999, Phys. Rev. E, 60, 5619]. In the present work, the rotational motion of the smectic layers of a SmA* liquid crystal, over which an oblique magnetic field has been applied, is studied theoretically by the aid of this theory. The role of the interaction between the smectic layers and the substrates is discussed and it is shown that if this interaction is small enough, the smectic layers orient themselves in such a way that the layer normal is almost parallel to the magnetic field. The relevant material parameters governing the dynamical response of the induced tilt and the layer normal are identified, and the time dependence of the response of these two dynamical variables is calculated. From this calculation, expressions for the response times of the system are given and a numerical value of the rotational viscosity of the smectic layers is estimated.  相似文献
