The collapse profiles of foams formed from model aqueous and non-aqueous foaming solutions have been recorded using a novel device, the Optical Transmission Foam Meter. An equation comprising two exponential terms has been used to describe the profiles and an attempt has been made to relate the equation parameters to some of the physical properties of the gas and liquid phases.  相似文献

We have investigated the use of a "split-sinc" RF pulse to selectively store magnetization from a selected region of a sample, for later recall in imaging or spectroscopy experiments. The pulse sequence is based on an original suggestion by Post et al. (West German Patent No. P3209263.6, 13 March 1982), later implemented by Aue et al. (J. Magn. Reson. 56, 350 (1984)). We have carried out detailed numerical calculations using the Bloch equations and show that this particular sequence is robust in the face of strong transverse relaxation, and we demonstrate its application to imaging of polymer samples in shearing and extensional flow cells.  相似文献

Boron-11 nuclear magnetic resonance imaging and spectroscopy have been used to characterise the nature and distribution of boron compounds after preservative treatment of radiata pine wood with trimethylborate (TMB). One day after treatment, 11B magnetic resonance imaging microscopy showed significant differences in the morphological distribution of boron species, with there apparently being no treatment penetration of the latewood. 11B MAS NMR spectroscopy of freshly preservative-treated radiata pine wood, which had been separated into latewood and earlywood, showed the only boron species present in the latewood to be boric acid while in the earlywood both TMB and boric acid were initially present. Due to quadrupolar broadening and a short T2 value, the boric acid signal relaxed too quickly to be observable and this caused the apparent lack of 11B in the image when only boric acid was present. TMB undergoes hydrolysis to form boric acid and in radiata pine latewood, this hydrolysis reaction is rapid.  相似文献

Most cheeses can be considered as solid emulsions of milk fat in a matrix of water and proteins. Regions of each of the phases can be liquid during processing and maturation. Identifying these regions and monitoring changes in them is important as a prelude to controlling the structure of the final cheese. We concentrate on the behavior of water in the vicinity of proteins as a function of cheese aging. Our method utilizes nuclear magnetic relaxation dispersion (NMRD) associated with the frequency dependence of water spin-lattice relaxation rates using the field cycling NMR technique. This method provides insight into the dynamical behavior of water molecules on a very large time scale. Moreover, we can distinguish between molecular motion in bulk and motion in the vicinity of a source of relaxation, such as proteins. A fit of our dispersion data using a theory developed by J.-P. Korb and R.G. Bryant (J. Chem. Phys. 115 (2001) 23) allowed us to determine the degree of hydration of proteins as a function of aging. In particular, we find that protein hydration increases with ripening.  相似文献

Present in all organisms, DNA ligases catalyse the formation of a phosphodiester bond between a 3′ hydroxyl and a 5′ phosphate, a reaction that is essential for maintaining genome integrity during replication and repair. Eubacterial DNA ligases use NAD+ as a cofactor and possess low sequence and structural homology relative to eukaryotic DNA ligases which use ATP as a cofactor. These key differences enable specific targeting of bacterial DNA ligases as an antibacterial strategy. In this study, four small molecule accessible sites within functionally important regions of Escherichia coli ligase (EC-LigA) were identified using in silico methods. Molecular docking was then used to screen for small molecules predicted to bind to these sites. Eight candidate inhibitors were then screened for inhibitory activity in an in vitro ligase assay. Five of these (geneticin, chlorhexidine, glutathione (reduced), imidazolidinyl urea and 2-(aminomethyl)imidazole) showed dose-dependent inhibition of EC-LigA with half maximal inhibitory concentrations (IC50) in the micromolar to millimolar range (11–2600 µM). Two (geneticin and chlorhexidine) were predicted to bind to a region of EC-LigA that has not been directly investigated previously, raising the possibility that there may be amino acids within this region that are important for EC-LigA activity or that the function of essential residues proximal to this region are impacted by inhibitor interactions with this region. We anticipate that the identified small molecule binding sites and inhibitors could be pursued as part of an antibacterial strategy targeting bacterial DNA ligases.  相似文献

The messa di voce, in its pure form a crescendo and decrescendo on one note, has been revered for centuries in classical singing, but the pedagogical assumptions of linearity and symmetry have received little critical assessment, especially across a wide fundamental frequency (F0) range. Five trained female classical singers performed a total of 318 messe di voce across their musical F0 range to identify its acoustical characteristics and the influence of F0. Sound pressure level (SPL) range was generally greater during crescendo at higher F0's and during decrescendo at lower FO's. Change in SPL during the messa di voce was predominantly nonlinear, and the shape of the SPL traces differed greatly between crescendo and decrescendo. Nonlinearity in SPL change was not related to SPL range but did show a F0 influence in decrescendo. Change in spectral balance (0-2 vs. 2-4 kHz) with respect to SPL change showed markedly more symmetry than linearity, so that changes in the mode of phonation during the messa di voce were dependent upon SPL regardless of whether the singer was in crescendo or decrescendo. Perceptual and physiological implications are discussed.  相似文献

Surface characterization and foaming studies were carried out with nine industrially manufactured, colloidal silica dispersions with particles sizes from 5-40 nm. All the silica sols produced transient foams with short decay times and the dynamic foam generation (foamability) was found to vary according to the sol type with the greatest foamability occurring for the hydrophobically modified sol and the deionized hydrophilic sol. However, it was found that improved foamability of all the sols could be achieved by changing the pH to within the region of the pH(pzc) which corresponds to the region of lowest hydrophilicity. An increase in pH (and build-up of negative charge) enhances the surface hydrophilicity and caused a decrease in foamability. In addition, for selected hydrophilic sols, it was shown that the foamability (a) increased with decrease in particle size (within the 6-40 nm range) and (b) increased with particle concentration (within the range of 1-15 wt%). Overall, it was concluded that the foamability was primary controlled by hydrophobicity (and hence by pH) and also by the particle concentration, the particle size and the degree of agglomeration.  相似文献

The azaspiracids are a family of lipophilic polyether marine biotoxins that have caused a number of human intoxication incidents in Europe since 1995 after consumption of contaminated shellfish (Mytilus edulis). Levels of azaspiracids in shellfish for human consumption are monitored in accordance with EU guidelines: only shellfish with less than 160 μg kg−1 are deemed safe. The limited availability of commercially available standards for azaspiracids is a serious problem, because validated LC–MS methods are required for routine analysis of these toxins in shellfish tissues. The procedure described herein has been used for the separation and the isolation of four azaspiracid (AZA) toxins from shellfish, for use as LC–MS–MS reference materials. Five separation steps have been used to isolate azaspiracids 1, 2, 3, and 6. The purity of the toxins obtained has been confirmed by multiple mass spectrometric methods using authentic azaspiracid standards. The same techniques have been used for quantification of the toxins extracted. The isolation procedure involves several chromatographic purification techniques: solid-phase extraction (diol sorbent, 90% mass reduction, and 95 ± 1% toxin recovery); Sephadex size-exclusion chromatography (87% mass reduction and up to 95 ± 2% toxin recovery), Toyopearl HW size-exclusion chromatography (90% mass reduction and up to 92.5 ± 2.5% toxin recovery), and semi-preparative LC (78 ± 3% toxin recovery). The procedure effectively separates the toxins from the sample matrix and furnishes azaspiracid toxins (AZA1, AZA2, AZA3 and AZA6) of sufficient purity with an average yield of 65% (n = 5). Triple-quadrupole mass spectrometry was used for qualitative and quantitative monitoring of the isolation efficiency after each stage of the process. High-resolution mass spectrometric evaluation of the toxic isolated material in both positive and negative modes suggests high purity.  相似文献

Aerosol OT/water exhibits a lamellar phase over a wide range of concentrations. We show, by magnetic resonance (NMR) and scanning electron microscopy (SEM), that the morphology of the lamellar phase varies significantly across that range and that the rate of equilibration depends strongly on concentration (25, 33, and 50 wt %) with, paradoxically, the faster equilibration at higher surfactant concentrations. We find that the 25 wt % sample exhibits a defect-rich local structure, characteristic of a superposed L(3) character. Further into the lamellar region, at 33 wt %, this defect-rich structure persists heterogeneously, while, at 50 wt %, the lamellar phase domains are highly ordered. The NMR methods used here included (2)H spectroscopy and the two-dimensional NMR method, diffusion-diffusion exchange spectroscopy (DEXSY). The latter was used to obtain quantitative information on the domain sizes and defects within the polydomain lamellar mesophase. Comparison of the NMR with the SEM results suggests that, at 25 wt % AOT, bilayer defects play an important role in influencing the (2)H NMR and DEXSY NMR results.  相似文献

We demonstrate here a method whereby molecular diffusion coefficients may be measured in the presence of the deformational flow field of a rheo-NMR cell. The method, which uses a repetitive CPMG train of rf pulses interspersed with magnetic field gradient pulses, allows the anisotropic diffusion spectrum to be directly probed. We focus on the cylindrical Couette cell, for which the radial, tangential, and axial directions correspond to the hydrodynamic velocity gradient, velocity, and vorticity directions. While ideal Couette flow does not perturb the vorticity direction, it does perturb diffusion measurements for the velocity gradient direction, and to a lesser extent, the velocity direction. We show that with closely spaced gradient pulses operating in a flow-compensating mode, there exists a diffusion limit below which one cannot measure, that scales as T(2)gamma(4), where gamma is the shear rate and T the gradient pulse repetition period. For a typical rheo-NMR cell, and for the more challenging velocity gradient direction, diffusion rates above 10(-12) m(2) s(-1) can be accurately measured (to 1% error) at shear rates up to 3s(-1). We demonstrate the use of the method in measuring the diffusion spectrum of a lyotropic lamellar phase under shear.  相似文献
