An efficient synthetic approach for the preparation of macrocyclic peptidomimetics for inhibition of HCV NS3 is presented. The macrocyclic core is built using ring-closing metathesis (RCM) of a tripeptidic diene. The presented approach allows the introduction of heteroatoms in strategic places along the macrocyclic ring. [reaction: see text] Two formal total syntheses of the (-)-salicylihalamides, based on chiral pool approaches, are reported. D-glucose and L-rhamnose were used to prepare advanced intermediates 23 and 54, which can be converted in three or four steps, respectively, to the target compounds. The synthesis of 23 from a known D-glucose-derivative was accomplished in 12 steps and 17% overall yield, and the synthesis of 54 from a known L-rhamnose-derivative was done in nine steps and 6% overall yield. A key step in the synthesis was a ring-closing metathesis reaction to prepare the macrocyclic ring system. It was demonstrated that the phenolic protecting group was critical for inducing the preferential formation of the desired E isomer. It was further shown that the protecting group at the C13 hydroxyl group had no significant influence on the E:Z ratio during the ring-closing metathesis reaction. 相似文献

The synthesis of the new potentially hexadentate ligands N,N'-bis(2,2'-bipyridin-6-ylmethyl)butane-1,4-diamine (bmbu), N,N'-bis(2,2'-bipyridin-6-ylmethyl)pentane-1,5-diamine (bmpt) and N,N'-bis(2,2'-bipyridin-6-ylmethyl)octane-1,8-diamine (bmot) from the condensation of 2,2'-bipyridine-6-carbaldehyde with the appropriate diamine (butane-1,4-diamine, pentane-1,5-diamine and octane-1,8-diamine, respectively) and subsequent reduction, is reported. Bmet, bmpp and bmbu all form mononuclear complexes with first-row transition metal ions (Co(3+), Fe(2+), Ni(2+), Mn(2+)), and X-ray structures of [Mn(bmet)](ClO(4))(2), [Ni(bmet)](ClO(4))(2), [Fe(bmet)](ClO(4))(2), [Mn(bmpp)](ClO(4))(2)·2MeCN and [Co(bmpp)](ClO(4))(3)·H(2)O are reported. As the aliphatic methylene chain increases in length, formation of dinuclear, and in some cases trinuclear, complexes becomes more pronounced, as evidenced by mass spectral analysis of solutions containing Ni(2+) and bmpt, and Ni(2+), Fe(2+) and Mn(2+) with bmot. The increasing preference for multinuclear complexes with increasing chain length is ascribed to the difficulty of incorporating a medium-sized (8 to 13-membered) chelate ring in a mononuclear complex. 相似文献

Described is a bioorthogonal reaction that proceeds with unusually fast reaction rates without need for catalysis: the cycloaddition of s-tetrazine and trans-cyclooctene derivatives. The reactions tolerate a broad range of functionality and proceed in high yield in organic solvents, water, cell media, or cell lysate. The ligand, 2-((2-hydroxy-5-methyl-3-((pyridin-2-ylmethylamino)methyl)benzyl)(2-hydroxybenzyl)amino)acetic acid (H(3)HPBA), which contains a donor atom set that mimics that of the active site of purple acid phosphatase is described. Reaction of H(3)HPBA with iron(III) or iron(II) salts results in formation of the tetranuclear complex, [Fe(4)(HPBA)(2)(OAc)(2)(mu-O)(mu-OH)(OH(2))(2)]ClO(4) x 5H(2)O. X-Ray structural analysis reveals the cation consists of four iron(III) ions, two HPBA(3-) ligands, two bridging acetate ligands, a bridging oxide ion and a bridging hydroxide ion. Each binucleating HPBA(3-) ligand coordinates two structurally distinct hexacoordinate iron(III) ions. The two metal ions coordinated to a HPBA(3-) ligand are linked to the two iron(III) metal ions of a second, similar binuclear unit by intramolecular oxide and hydroxide bridging moieties to form a tetramer. The complex has been further characterised by elemental analysis, mass spectrometry, UV-vis and MCD spectroscopy, X-ray crystallography, magnetic susceptibility measurements and variable-temperature M?ssbauer spectroscopy. 相似文献

The syntheses and X-ray structures of [Co(Me-tpa)O(2)COZnCl(3)], [Co(pmea)O(2)COZnCl(3)].H(2)O [Co(trpyn)O(2)COZn(OH(2))(4)OCO(2)Co(trpyn)](ZnCl(4))(2).H(2)O, [Co(trpyn)(O(2)COH)]ZnCl(4).3H(2)O and [Co(trpyn)(O(2)CO)]ClO(4) are reported (Me-tpa = [(6-methyl-2-pyridyl)methyl]bis(2-pyridylmethyl)amine, pmea = bis(2-pyridylmethyl)-2-(2-pyridylethyl)amine, trpyn = tris(2-(1-pyrazolyl)ethyl)amine). The chelated bicarbonate complex [Co(trpyn)(O(2)COH)]ZnCl(4).3H(2)O is isolated as a crystalline solid from an acidic solution of the parent carbonate [Co(trpyn)(O(2)CO)]ClO(4), and X-ray structural analysis shows that lengthening of the C[double bond, length as m-dash]O(exo) bond and shortening of the C-O(endo) bond accompanies protonation. The bimetallic complex [Co(Me-tpa)O(2)COZnCl(3)] results from the unexpected coordination of ZnCl(3)(-) to the exo O atom of a chelated carbonate ligand. This complex is obtained from both acidic and neutral solutions in which [Zn(2+)] = 1.0 M, while the structurally similar complex [Co(pmea)O(2)COZnCl(3)].H(2)O is isolated from an analogous neutral solution. The trimetallic complex [Co(trpyn)O(2)COZn(OH(2))(4)OCO(2)Co(trpyn)](ZnCl(4))(2).H(2)O crystallises on prolonged standing of [Co(trpyn)(O(2)CO)]ClO(4) in a neutral solution having [Zn(2+)] = 1.0 M. The Zn-O bond lengths in all three complexes are indicative of bonds of significant strength. DFT calculations show that the nature of the bonding interaction between the Co(iii) ion and the endo O atoms of the carbonate ligand remain essentially unaffected by coordination of Zn(2+) to the exo O atom. They also show that such coordination of Zn(2+) decreases the C-O(exo) bond order. 相似文献

Thin films of tungsten carbonitride have been formed on glass by low-pressure chemical vapour deposition (LP)CVD at 550 degrees C from four closely related precursors: [W(mu-N(t)Bu)(N(t)Bu)Cl(2)(H(2)N(t)Bu)](2), [W(N(t)Bu)(2)Cl(2)(TMEDA)] (TMEDA = N,N,N',N'-tetramethylethylenediamine), [W(N(t)Bu)(2)Cl(2)(py)(2)] (py = pyridine) and [W(N(t)Bu)(2)Cl(N{SiMe(3)}(2))]. The grey mirror-like films were grown with a nitrogen or ammonia bleed gas. In all cases the chlorine content of the deposited films was less than 1 at% and the oxygen content of the films was lower for those grown using ammonia. Surprisingly, the use of ammonia did not significantly change the carbon content of the resulting films. Despite the coordination environment around the metal being essentially the same and the materials having a comparable volatility, some differences in film quality were observed. The films were uniform, adhesive, abrasion resistant, conformal and hard, being resistant to scratching with a steel scalpel. X-Ray powder diffraction patterns of all the films showed the formation of beta-WN(x)C(y). As a comparison the aerosol-assisted chemical vapour deposition (AA)CVD of [W(mu-N(t)Bu)(N(t)Bu)Cl(2)(H(2)N(t)Bu)](2) was investigated and amorphous tungsten carbonitride films were deposited. 相似文献
