The fast parallel restoration (FPR) scheme is proposed to achieve the fast setup of restoration label switched path (LSP) in the distributed optical networks. The scheme is derived by dividing the whole restoration LSP into several segments of sub-LSP and triggering each sub-LSP along the new route to finish the signaling procedure concurrently, and subsequently merging all sub-LSPs into a whole LSP. The theoretical analysis and simulation results show that the FPR scheme outperforms the other two typical restoration schemes in terms of connection setup time.  相似文献

In this work, a novel method was established to isolate and purify Human plasminogen Kringle 5 (HPK5) as a histidine‐tagged fusion protein expressed in Escherichia coli BL21 (DE3). This method consisted of sample extraction using a Ni‐chelated Sepharose Fast‐Flow affinity column, ammonium sulfate salting‐out and Sephadex G‐75 size‐exclusion column in turn. The purity analysis by SDS–PAGE, high‐performance size‐exclusion and reversed‐phase chromatographies showed that the obtained recombinant fusion HPK5 was homogeneous and its purity was higher than 96%; the activity analysis by chorioallantoic membrane model of chicken embryos revealed that the purified recombinant HPK5 exhibited an obvious anti‐angiogenic activity under the effective range of 5.0–25.0 µg/mL. Through this procedure, about 19 mg purified recombinant fusion HPK5 can be obtained from 1 L of original fermentation solution. Approximate 32% of the total recombinant fusion HPK5 can be captured and the total yield was approximately 11%. Copyright © 2013 John Wiley & Sons, Ltd.  相似文献

This study examined the interaction of indirubin with bovine serum albumin (BSA) at three temperatures (286, 297, 308 K) at pH 7.40. In the presence of indirubin, the drug-BSA binding mode, binding constant and the protein structure changes in aqueous solution were determined by fluorescence quenching methods including Fourier transform infrared (FT-IR) spectroscopy and UV-Vis spectroscopy. The FT-IR change indicates that indirubin binds to BSA. The change in protein secondary structure accompanying ligand binding has been proved by fluorescence spectra data. The thermodynamic parameters, the enthalpy change (DeltaH), and the entropy change (DeltaS) calculated by the van't Hoff equation possess small negative (-2.744 kJ.mol(-1)) and positive values (112.756 J.mol(-1).K(-1)), respectively, which indicated that hydrophobic interactions play the main role in the binding of indirubin to BSA. Furthermore, the displacement experiment shows that indirubin can bind to the subdomain IIA and the distance between the tryptophan residues in BSA and indirubin bound to site I was estimated to be 2.24 nm according to F?ster's equation on the basis of fluorescence energy transfer.  相似文献

Catalysis of electron transfer by a Cu-substituted wheel-type oxomolybdate cluster–anion, , (1), is demonstrated. Data provided include aqueous-solution chemistry (stability) studies of 1 and , (2), derivatives of the “plenary” {Mo154} anion, , (3). Combined use of cyclic voltammetry and UV–vis spectroscopy shows that, while both 1 and 2 appear to be stable in solution at pH 0.33 (0.5 M H2SO4), 1 is more stable than 2 at pH 3 (in 0.2 M Na2SO4). Cyclic voltammetric analysis in the presence of O2 shows that 1 is an electrocatalyst for electron transfer to O2. Bulk electrolysis of 1 in the presence of O2 (ca. 1 mM) is used to assess catalyst stability under turnover conditions, and to demonstrate that the final product of electrocatalytic reduction is water, rather than H2O2. Finally, control experiments using 1, 2, and CuSO4 (no oxomolybdate-cluster present), show that catalytic activity is due to specific interaction(s) between Cu ions and the Mo142 type oxomolybdate structure of 1.  相似文献

The strong fluorescence Tb/acetyl acetone (acac)/Poly (Acrylamide) (PAM) composite nanoparticles have been prepared under ultrasonic radiation. The nanoparticles were water-soluble, stable and have extremely narrow emission bands and high internal quantum efficiencies. Based on the fluorescence quenching of Tb/acac/PAM by Cr (VI), a method for the selective determination of Cr (VI), without separation of Cr (III) in water, was developed. The reaction condition between Cr (VI) and Tb/acac/PAM were investigated in detail. Under optimal experimental conditions, the linear calibration curve was obtained over the concentration range of 5-600 ng mL(-1) with a correlation coefficient of 0.9939. The corresponding detection limit is 0.8 ng mL(-1) and the relative standard deviation is 1.5% for 0.05 microg mL(-1) (n=7). The proposed method has been applied to the selective quantification of Cr (VI) in synthetic samples and waste-water samples with the satisfactory results. The assay is characterized by short reaction time, very few interference, stable fluorescence signals, simple instrument and simplicity.  相似文献

In this paper, we developed a facile way to synthesize highly ordered optically active MCM-48 at room temperature, by using mixtures of hemicyanine dye N-alkyl-2-[p-(N,N-diethylamino)-o-(alkyloxy)]pyridinium bromide (denoted as o-CnPOCm, Scheme 1) and cetyltrimethylammonium bromide (CTAB) as the structure-directing agents. The mesoporous materials were systematically characterized by powder X-ray diffraction, transmission electron microscopy, nitrogen sorption, and thermogravimetry. The resultant MCM-48 exhibits unusually high thermal stability. For example, in the case of o-C(2)POC(14), it can retain its cubic structure even under calcinations at 900 degrees C for 5 h, although the pore size is shifted to the micropore region because of shrinkage of the framework. The typical surface area and pore volume are 980 m(2)/g and 0.44 cm(3)/g, respectively, for the powder calcined under such a high temperature. This is the first report of room-temperature synthesis of MCM-48 with such good thermal stability using cationic-cationic mixed surfactant as the structure-directing agent. The fluorescence lifetimes of the as-synthesized mesostructured MCM-48 were also measured, and the result showed that the incorporated dye molecules have a 1 order of magnitude longer lifetime than that of free species in solution, showing that the hemicyanine dye molecules are well dispersed within the CTAB surfactant matrix. Furthermore, we compared eight other dye congeners (Scheme 1) to fully investigate the mesophase resulting from the dye-CTAB system. The results show that, upon addition of the dye surfactant to the starting mixtures, the mesostructured silica undergoes an intrinsic phase-transition process; however, specific dye geometry is required to obtain MCM-48 at room temperature. Those functionalities as well as the designed synthesis of this novel mesostructured MCM-48 material promise a bright future in multifunctional optical and electric nano- and microdevices (e.g., waveguides, laser, light-emitting diodes, etc.) and also shed light on the self-assembly behavior in complex colloidal system.  相似文献

A method has been developed for the qualitative and quantitative analysis of pharmacologically active astragalosides isolated from several species of the genus Astragalus by high performance liquid chromatography coupled with electrospray ionization time-of-flight mass spectrometry. Seven astragalosides in Radix Astragali and their commercial pharmaceutical preparations were analyzed using the developed method. The extracted ion current chromatograms were obtained from the total ion current chromatogram using the m/z of [M+Na]+ ions produced by target compounds for peak determination. The limits of detection and limits of quantification were in the range of 0.10-0.22 ng and 0.22-0.52 ng in full scan mode, respectively. All calibration curves showed good linear regression (r2 > or = 0.9965) within the test range. The overall intra- and inter-day precision was less than 2.86% for peak area and the accuracy was higher than 92.9% on using ginsenoside I as internal standard. The assay was successfully utilized to analyze the major biologically active astragalosides in six samples of Astragalus membranaceus (Fisch.) Bge var. mongholicus (Bge.) Hsiao. and eight commercial preparations. The overall results demonstrate that this method is simple, selective, and suitable for the quality control of Chinese medicine and their preparation in the low nanogram range.  相似文献

Bipyrimidines have been chosen as (N∧N)(N∧N) bridging ligands for connecting metal centers. IrIII-LnIII (Ln = Nd, Yb, Er) bimetallic complexes [Ir(dfppy)2(μ-bpm)Ln(TTA)3]Cl were synthesized by using Ir(dfppy)2(bpm)Cl as the ligand coordinating to lanthanide complexes Ln(TTA)3·2H2O. The stability constants between Ir(dfppy)2(bpm)Cl and lanthanide ions were measured by fluorescence titration. The obvious quenching of visible emission from IrIII complex in the IrIII-LnIII (Ln = Nd, Yb, Er) bimetallic complexes indicates that energy transfer occurred from IrIII center to lanthanides. NIR emissions from NdIII, YbIII, and ErIII were obtained under the excitation of visible light by selective excitation of the IrIII-based chromophore. It was proven that Ir(dfppy)2(bpm)Cl as the ligand could effectively sensitize NIR emission from NdIII, YbIII, and ErIII.  相似文献
