From cultures of thermophilic soil fungus Humicola grisea var thermoidea, a δ-lactam derivative (3-(2-(4-hydroxyphenyl)-2-oxoethyl)-5,6-dihydropyridin-2(1H)-one) that displayed anti-allergic activity was isolated, which was predicted by in silico computational chemistry approaches. The in?vitro anti-allergic activity was investigated by β-hexosaminidase release assay in rat basophilic leukaemia RBL-2H3 cells. The δ-lactam derivative exhibited similar anti-allergic activity (IC(50)?=?18.7?±?6.7?μM) in comparison with ketotifen fumarate (IC(50)?=?15.0?±?1.3?μM) and stronger anti-allergic activity than azelastine (IC(50)?=?32.0?μM). Also, the MTT cytotoxicity assay with RBL-2H3 cells showed that δ-lactam does not display cytotoxicity at concentrations lower than 50?μM. This study suggests that the δ-lactam derivative has the potential to be used as a lead compound in the development of anti-allergic drugs for clinical use in humans.  相似文献

An invertibility theory for classes of convolution type operators on a union of bounded intervals whose kernels have Fourier transforms which are related to solutions of corona problems is established and the corresponding formulas for the inverse operators are given. A generalization of the portuguese transformation for matrix functions is obtained and is used to establish the invertibility theory for one of the above mentioned classes of operators. The same transformation allows, also, to establish the equivalence between convolution type operators on an union of disjoint intervals and convolution type operators on a bounded interval.  相似文献

Hydroethanolic extracts of C. langsdorffii leaves have therapeutic potential. This work reports a validated chromatographic method for the quantification of polar compounds in the hydroethanolic extract of C. langsdorffii leaves. A reliable HPLC method was developed using two monolithic columns linked in series (100 x 4.6 mm - C18), with nonlinear gradient elution, and UV detection set at 257 nm. A procedure for the extraction of flavonols was also developed, which involved the use of 70% aqueous ethanol and the addition of benzophenone as the internal standard. The developed method led to a good detection response as the values for linearity were between 10.3 and 1000 microg/mL, and those for recovery between 84.2 and 111.1%. The detection limit ranged from 0.02 to 1.70 microg/mL and the quantitation limit from 0.07 to 5.1 microg/mL, with a maximum RSD of 5.24%. Five compounds, rutin, quercetin-3-O-alpha-L-rhamnopyranoside, kaempferol-3-O-alpha-L-rhamnopyranoside, quercetin and kaempferol, were quantified. This method could, therefore, be used for the quality control of hydroethanolic extracts of Copaifera leaves and their cosmetic and pharmaceutical products.  相似文献

The influence of several experimental parameters on the ultrasonic extraction of Hibiscus tiliaceus L. flowers were investigated: extraction time, solvent polarity, sample amount, solvent volume and sample particle size. It was concluded that the most influential variables were extraction time and solvent polarity. The optimized procedure employed 5 g of ground flowers, 150 mL of methanol and 140 min of extraction. The extracts were fractionated using preparative silica columns and the resulting fractions were analyzed by GC/MS. Some saturated hydrocarbons, fatty acids, fatty acid methyl esters, phytosterols, and vitamin E were identified in the plant extracts.  相似文献

A simple and fast voltammetric method based on a new electrode composed of carbon paste electrode/bifunctional hybrid ion imprinted polymer (CPE/IIP) was developed for the quantification of Cd2+ in water samples. The voltammetric measurements by Differential Pulse Voltammetry were performed by using CPE containing 11.0 mg of IIP under phosphate buffer solution at concentration 0.1 mol L?1 and pH 6.5. The electrochemical method was carried out by Cd2+ preconcentration at ?1.2 V during 210 s, followed by anodic stripping. The performance of IIP towards Cd2+ determination was evaluated by comparison to non-imprinted polymer, whose detectability of IIP was much higher (45%). The sensitivity of the sensor was found to be 0.0105 µA/µg L?1. The limits of detection and limits of quantification were found to be 4.95 μg L?1 and 16.4 μg L?1, respectively. The developed method was successfully applied to Cd2+ determination in mineral, tap and lake water samples, whose results are in agreement with thermospray flame furnace atomic absorption spectrometry (TS-FF-AAS) used as reference analytical technique. According to achieved results, the developed method can be used for routine analysis of quality control of water samples from different sources.  相似文献

Finite interval convolution operators with periodic kernel-functions are studied from the point of view of Fredholm properties and invertibility. These operators are associated with Wiener-Hopf operators with matrix-valued symbols defined on a space of functions whose domain is a contour consisting of two parallel straight-lines. For the Fredholm study a Wiener-Hopf operator is considered on a space of functions defined on a contour composed of two closed curves having a common multiple point.
