Vapor-phase adsorption and separation of the C8 alkylaromatic components p-xylene, m-xylene, o-xylene, and ethylbenzene on the metal-organic framework MIL-47 have been studied. Low coverage Henry adsorption constants and adsorption enthalpies were determined using the pulse chromatographic technique at temperatures between 230 and 290 degrees C. The four C8 alkylaromatic components have comparable Henry constants and adsorption enthalpies. Adsorption isotherms of the pure components were determined using the gravimetric technique at 70, 110, and 150 degrees C. The adsorption capacity and steepness of the isotherms differs among the components and are strongly temperature dependent. Breakthrough experiments with several binary mixtures were performed at 70-150 degrees C and varying total hydrocarbon pressure from 0.0004 to 0.05 bar. Separation of the different isomers could be achieved. In general, it was found that the adsorption selectivity increases with increasing partial pressure or degree of pore filling. The separation at a high degree of pore filling in the vapor phase is a result of differences in packing modes of the C8 alkylaromatic components in the pores of MIL-47.  相似文献

A library of stereo- and regiochemically diverse aminoglycoside derivatives was screened at 1 microM using surface plasmon resonance (SPR) against RNA hairpin models of the bacterial A-site, and the HIV viral TAR and RRE sequences. In order to double the stereochemical diversity of the library, the compounds were screened against both enantiomers of each of these sequences. Remarkably, this initial screen suggested that the same four aminoglycoside derivatives bound most tightly to all three of the RNAs, suggesting that these compounds were good RNA binders which, nonetheless, discriminated poorly between the RNA sequences. The interactions between selected isomeric aminoglycoside derivatives and the RNA hairpins were then studied in more detail using an SPR assay. Three isomeric tight-binding aminoglycoside derivatives, which had been identified from the initial screen, were found to bind more tightly to the RNA hairpins (with K(D) values in the range 0.23 to 4.7 microM) than a fourth isomeric derivative (which had K(D) values in the range 6.0 to 30 microM). The magnitude of the tightest RNA-aminoglycoside interactions stemmed, in large part, from remarkably slow dissociation of the aminoglycosides from the RNA targets. The three tight-binding aminoglycoside derivatives were found, however, to discriminate rather poorly between alternative RNA sequences with, at best, around a twenty-fold difference in affinity for alternative RNA hairpin sequences. Within the aminoglycoside derivative library studied, high affinity for an RNA target was not accompanied by good discrimination between alternative RNA sequences.  相似文献

Biomolecule-metal or semiconductor nanoparticle (NP) hybrid systems combine the recognition and catalytic properties of biomolecules with the unique electronic and optical properties of NPs. This enables the application of the hybrid systems in developing new electronic and optical biosensors, to synthesize nanowires and nanocircuits, and to fabricate new devices. Metal NPs are employed as nano-connectors that activate redox enzymes, and they act as electrical or optical labels for biorecognition events. Similarly, semiconductor NPs act as optical probes for biorecognition processes. Double-stranded DNA or protein chains that are modified with metallic nanoclusters act as templates for the synthesis of metallic nanowires. The nanowires are used as building blocks to assemble nano-devices such as a transistor or a nanotransporter.  相似文献

The solid-state1H MAS (magic-angle spinning),2H static,15N CP (cross polarization)-MAS and15N-1H dipolar CSA (chemical shielding anisotropy) NMR (nuclear magnetic resonance) spectra of two different modifications of Cα-deuteratedl5N-polyglycine, namely PG I and PG II (-CO-CD2-l5NH-)n are measured. The data from these spectra are compared to previous NMR, infrared, Raman and inelastic neutron scattering work. The deuteration of Cα eliminates the largest intramolecular1H-1H dipolar coupling. The effect of the remaining (N)H-(N)H interaction (~5 kHz) is not negligible compared to the15N-1H coupling (about 10 kHz). Its effect on the dipolar CSA spectra, described as a two-spin system, is analyzed analytically and numerically and it is shown that those parts of the powder spectrum, which correspond to orientations with a strong dipolar15N-1H interaction, can be described as an effective two-spin system, permitting the measurement of the strength of the15N-1H dipolar interaction and the orientation of the dipolar vector with respect to the15N CSA frame. While in the PG II system the15N CSA tensor is collinear with the amide plane, in the PG I system the CSA tensor is tilted ca. 16° with respect to the (δ11δ22) CSA plane.  相似文献

Many schemes for calculating reaction rates and free energy barriers require an accurate reaction coordinate, but it is difficult to quantify reaction coordinate accuracy for complex processes like protein folding and nucleation. The histogram test, based on estimated committor probabilities, is often used as a qualitative indicator for good reaction coordinates. This paper derives the mean and variance of the intrinsic committor distribution in terms of the mean and variance of the histogram of committor estimates. These convenient formulas enable the first quantitative calculations of reaction coordinate error for complex systems. An example shows that the approximate transition state surface from Peters' and Trout's reaction coordinate for nucleation in the Ising model gives a mean committor probability of 0.495 and a standard deviation of 0.042.  相似文献

Chronic skin exposure to ultraviolet light stimulates the production of cytokines known to be involved in the initiation of skin cancer. Recent studies in mouse models suggested a role for macrophage migration inhibitory factor (MIF) in the UVB‐induced pathogenesis of nonmelanoma skin cancer (NMSC). Our studies aimed at defining the pathophysiological function of MIF in cutaneous inflammatory reactions and in the development and progression of NMSC. Immunohistochemical analysis revealed a moderate expression of MIF in normal human skin samples but an enhanced expression of this cytokine in lesional skin of patients with actinic keratosis or cutaneous SCC. Enzyme‐linked immunosorbent assay studies showed a time‐dependent increase in MIF secretion after a moderate single‐dose UVB irradiation in NHEKs and SCC tumor cells. MIF is known to interact with CXCR2, CXCR4 and CD74. These receptors are not constitutively expressed in keratinocytes and HaCaT cells and their expression is not induced by UVB irradiation either. However, stimulation with IFNγ upregulated CD74 surface expression in these cells. Affymetrix® Gene Chip analysis revealed that only keratinocytes prestimulated with IFNγ are responsive to MIF. These findings indicate that MIF may be an important factor in the pathogenesis of NMSC tumorigenesis and progression in an inflammatory environment.  相似文献

Transition path sampling (TPS) algorithms have been implemented with deterministic dynamics, with thermostatted dynamics, with Brownian dynamics, and with simple spin flip dynamics. Missing from the TPS repertoire is an implementation with kinetic Monte Carlo (kMC), i.e., with the underlying dynamics coming from a discrete master equation. We present a new hybrid kMC-TPS algorithm and prove that it satisfies detailed balance in the transition path ensemble. The new algorithm is illustrated for a simplified Markov State Model of trp-cage folding. The transition path ensemble from kMC-TPS is consistent with that obtained from brute force kMC simulations. The committor probabilities and local fluxes for the simple model are consistent with those obtained from exact methods for simple master equations. The new kMC-TPS method should be useful for analysis of rare transitions in complex master equations where the individual states cannot be enumerated and therefore where exact solutions cannot be obtained.  相似文献

A fully-detailed LC-MS qualitative profiling of red grape skin, extracted with a mixture of ethanol and water (70:30 v:v) has permitted the identification of 65 compounds which can be classified into the following chemical classes: organic and phenolic acids (14 compounds), stilbenoids (1 compound), flavanols (21 compounds), flavonols (15 compounds) and anthocyanins (14 compounds). The extraction yield obtained with water at different temperatures (100 °C, 70 °C, room temperature) was then evaluated and the overall polyphenol content indicates that EtOH:H2O solvent is the most efficient and selective for polyphenol extraction. However, by analyzing the recovery yield of each single polyphenol, we found that water extraction under heating conditions is effective (extraction yield similar or even better in respect to the binary solvent) for some polyphenolic classes, such as hydrophilic procyanidins, phenolic acids, flavonol glucosides and stilbenoids. However, according to their lipophilic character, a poor yield was found for the most lipophilic components, such as flavonol aglycones, and in general for anthocyanins. The radical scavenging activity was in accordance with the polyphenol content, and hence, much higher for the extract obtained with the binary solvent in respect to water extraction. All the tested extracts were found to have an anti-inflammatory activity in the R3/1 cell line with NF-kb reporter challenged with 0.01 µg/mL of IL-1α, in a 1 to 250 µg/mL concentration range. An intriguing result was that the EtOH:H2O extract was found to be superimposable with that obtained using water at 100 °C despite the lower polyphenol content. Taken together, the results show the bioactive potentialities of grape skin extracts and the possibility to exploit this rich industrial waste. Water extraction carried out by heating is an easy, low-cost and environmentally friendly extraction method for some polyphenol classes and may have great potential for extracts with anti-inflammatory activities.  相似文献

Cadmium (Cd) is a heavy metal that occurs in all areas of the environment, including the food chain. In the body, it causes oxidative stress by producing free radicals that are harmful to the cells. Grape seed extract (GSE) contains a wide range of biologically active components that help to neutralize the adverse effects of free radicals. In this study, the effects of GSE prepared form semi-resistant grapevine cultivar Cerason, which is rich in phenolics, on biochemical markers of brown rats exposed to the effects of cadmium were monitored. GSE increased the plasma antioxidant activity and, in the kidneys and the liver, Cd content was significantly lowered by GSE co-administration. Accordingly, the increase in creatinine content and alanine aminotransferase activity and the decrease of catalase and superoxide dismutase activities caused by cadmium were slowed down by GSE co-administration. The results of this work reveal that grape seed extract offers a protective effect against the intake of heavy metals into the organism.  相似文献
