In this study, the suitability of SPE coupled on-line to CE-electrospray-MS (SPE-CE-ESI-MS) was evaluated for the analysis of neuropeptides in human plasma. First, CE-ESI-MS was investigated and a sample pretreatment based on precipitation with ACN was used for cleanup of plasma samples. The main quality parameters were determined and were consistent with those previously obtained for the analysis of standard mixtures, e.g., the LODs were of around 1 microg/mL. SPE-CE-ESI-MS employing microcartridges containing a C18 stationary phase was explored in order to decrease the LODs. A double-step sample cleanup pretreatment consisting of precipitation with ACN and ultrafiltration through 3000 Da MW cut-off membranes was needed to prevent the microcartridge saturation. Repeatability, reproducibility, linearity, and LODs of the SPE-CE-ESI-MS method and the durability of the microcartridges were studied. A rapid and simple high-performance liquid chromatographic method for the analysis of 1,3-dihydroxy-2-methylxanthone (DHMXAN) in biodegradable poly(D,L-lactide-co-glycolide) (PLGA) nanosphere and nanocapsule formulations is developed and validated. The method does not require any complex sample extraction procedure. Chromatographic separation is made with a reversed-phase C18 column, using methanol-water (90:10, v/v) containing 1% (v/v) acetic acid as a mobile phase at a flow rate of 1 mL/min. Identification is made by UV detection at 237 nm. The isocratic system operates at ambient temperature and requires 7.5 min of chromatographic time. The developed method is statistically validated according to ICH guidelines and USP 29 for its specificity, linearity, accuracy, and precision. The assay method proposed in this study is specific for DHMXAN in the presence of nanosphere and nanocapsule excipients. Diode-array analyses confirm the purity of DHMXAN peak in stress conditions (> 99.0%). The method is shown to be linear (r > or = 0.999) over the concentration range of 0.25-3.0 microg/mL. Recovery ranges from 99.0% to 102.7% (RSD: 1.49%) and from 98.3% to 101.6% (RSD: 1.07%) for nanospheres and nanocapsules, respectively. Repeatability (intra-assay precision) and intermediate precision is acceptable with RSD values ranging from 0.6% to 1.9% and from 0.3% to 2.0%, respectively. Surface tension, conductivity, density, and ultrasound velocity measurements have been performed in order to determine in a systematic manner some of the aggregation properties of the phenothiazine drugs promazine and triflupromazine hydrochlorides. Both drugs are structurally related, differing in an extra CF3 group in the triflupromazine molecular structure. Surface tension data showed that the presence of an extra CF3 in the molecular structure of triflupromazine involves a higher hydrophobicity of this drug and a restriction in the number of conformations molecules can adopt due to the presence of this bulkier atomic group. This involves a larger surface area in order to accommodate triflupromazine molecules at the interface. From conductivity measurements at different temperatures, the thermodynamic quantities of the micellization process of these drugs indicate that the aggregation is a spontaneous process, mainly enthalpic, where the London-dispersion forces play an active role. Using density and ultrasound velocity measurements, apparent molar volume and adiabatic compressibility of aqueous solutions of the amphiphilic cationic drugs have been determined. Positive deviations from the Debye–Hückel limiting law of the apparent molar volume were obtained from both drugs over the whole temperature range, which provides evidence of possible pre-association at concentrations below the critical concentration. Apparent molar adiabatic compressibility of the aggregates formed by these drugs was typical of those corresponding for an aggregate formed by a stacking process. 相似文献

The S1-S0 limiting anisotropy of a widely used fluorophore, rhodamine 101, is determined with unprecedented accuracy. From time-resolved and steady-state fluorescence measurements in several solvents, it is shown that the limiting anisotropy of rhodamine 101 is for all practical purposes equal to the theoretical one-photon fundamental anisotropy value of 2/5, both in rigid and in fluid media. This fact, along with the favorable chemical and photophysical properties of rhodamine 101, point to its use as a standard for fluorescence polarization measurements. It is also shown that if the excitation pulse can be considered a delta impulse with respect to the time scale of the anisotropy decay (but not necessarily to the time scale of the intensity decay), then no deconvolution procedure is needed for anisotropy decay analysis. 相似文献

Fourier Transform mid-infrared and Raman spectroscopies were used to investigate the cation/polymer, cation/urea bridge, cation/anion and hydrogen bonding interactions in poly(oxyethylene) (POE)/siloxane di-ureasil networks prepared by the sol–gel route and doped with lithium bis(trifluoromethanesulfonyl)imide (LiTFSI). Materials with compositions 200 ?n ? 5 (where n expresses the molar ratio OCH2CH2/Li+) were studied. The Li+ ions coordinate to the urea carbonyl oxygen atoms over the whole range of salt concentration considered. Bonding to the ether oxygen atoms of the POE chains occurs at n ? 40, although a significant fraction of the POE chains remain non-coordinated. In these high salt content samples, the cations interact with the anions forming contact ion pairs. “Free” ions are probably the main charge carriers at the room temperature conductivity maximum of these ormolytes. 相似文献

The uptake of ascorbate by neuroblastoma cells using a ruthenium oxide hexacyanoferrate (RuOHCF)-modified carbon fiber disc (CFD) microelectrode (r = 14.5 microm) was investigated. By use of the proposed electrochemical sensor the amperometric determination of ascorbate was performed at 0.0 V in minimum essential medium (MEM, pH = 7.2) with a limit of detection of 25 micromol L(-1). Under the optimum experimental conditions, no interference from MEM constituents and reduced glutathione (used to prevent the oxidation of ascorbate during the experiments) was noticed. The stability of the RuOHCF-modified electrode response was studied by measuring the sensitivity over an extended period of time (120 h), a decrease of around 10% being noticed at the end of the experiment. The rate of ascorbate uptake by control human neuroblastoma SH-SY5Y cells, and cells transfected with wild-type Cu,Zn-superoxide dismutase (SOD WT) or with a mutant typical of familial amyotrophic lateral sclerosis (SOD G93A), was in agreement with the level of oxidative stress in these cells. The usefulness of the RuOHCF-modified microelectrode for in vivo monitoring of ascorbate inside neuroblastoma cells was also demonstrated. 相似文献

In 2010 we presented an update of the nuclear quadrupole moments (Q) for the Cd and Zn isotopes, based essentially on straightforward density functional (DF) calculations (H. Haas and J.G. Correia, Hyperfine Interact 198, 133–137 (2010)). It has been apparent for some years that the standard DF procedure obviously fails, however, to reproduce the known electric-field gradient (EFG) for various systems, typical cases being Cu2O, As and Sb, and the solid halogens. Recently a cure for this deficiency has been found in the hybrid DF technique. This method is now applied to solid Cd and Zn, and the resultant quadrupole moments are about 15 % smaller than in our earlier report. Also nuclear systematics, using the recently revised values of Q for the long-lived 11/2 isomers in111Cd to129Cd, together with earlier PAD data for107,109Cd, leads to the same conclusion. In addition, EFG calculations for the cadmium dimethyl molecule further support the new values: Q(111Cd, 5/2+) = .683(20) b, Q(67Zn, gs) = .132(5) b. This implies, that the value for the atomic EFG in the \(^{3}\textit {P}_{1}\) state of Zn must be revised, as it has been for Cd. 相似文献

The molecular structure of conformational isomorphs given by X-ray diffraction for racemic and enantiomeric atenolol were optimized at the HF/6-31G* level of theory and the infrared spectra of the structure were calculated. These spectra are used to characterize the differences between the various atenolol conformers. The spectra of the (R,S)- and S-atenolol solid forms were recorded and the bands corresponding to the functional groups identified with the aid of the calculated spectra, fitting analysis, temperature effect and H/D isotopic exchange. 3-Benzyl-furan-2(5H)-one (2a) and 3-(4-bromobenzyl)-furan-2(5H)-one (2b) were treated with TBDMSOTf and converted into the corresponding tert-butyldimethyl-silylfuran ethers. These furans were further condensed with several aromatic aldehydes affording compounds 5-14 with general 3-benzyl-5-arylidene-furan-2(5H)-one structures in 31% to 98% yields. Such compounds are analogues of the naturally occurring nostoclide lactones, reported to present moderate cytotoxic activity. Erythropoietin (EPO) is a hormone that regulates red blood cell production. Recombinant human EPO (rHuEPO) and NESP (novel
erythropoiesis stimulating protein) have been produced for therapeutic purposes and also to improve sports performance. The
primary sequences of rHuEPO and NESP differ by just five amino acids. Due to the high homology, no antibodies that are able
to discriminate between both molecules have been obtained until now. The aim of the present work was to design synthetic peptides
corresponding to the sequence that differs between EPO and NESP (87–90aa), that can then be used as immunogens to develop
specific rabbit polyclonal antibodies for selectively detecting EPO and NESP. Three peptides were synthesized: EPO (81–95),
NESP (81–95), and NESP (86–104), and these were coupled to KLH and OVA for immunization and screening purposes, respectively.
The sera obtained were tested by ELISA on synthetic peptide–OVA conjugates and purified by immunoaffinity chromatography against
the corresponding synthetic peptide. The specific purified antibodies were characterized by ELISA, SDS-PAGE, and isoelectric
focusing, followed by western blot. Antisera raised against EPO (81–95) recognized rHuEPO but not NESP. In contrast, anti-NESP
(84–106) sera gave a specific anti-NESP response only after immunoaffinity purification on a NESP (86–91) column. An efficient
strategy for generating specific antibodies against EPO and NESP can be achieved by selecting suitable synthetic peptides.
The antibodies obtained are able to differentiate between rHuEPO and NESP, and may be particularly useful for screening purposes
in both therapeutic and antidoping contexts. 相似文献
