The retention of histidine-containing peptides in immobilized metal-affinity chromatography is studied using several hundred modeled peptides. Retention is driven primarily by the number of histidine residues; however, the amino acid composition in the immediate vicinity plays a significant role. Specifically, the arginine and tryptophan content has to be taken into consideration. During the course of this study, an alternative tag that can be used similarly to a polyhistidine tag is discovered.  相似文献

Flexible and economic sensor devices are the focus of increasing interest for their potential and wide applications in medicine, food analysis, pollution, water quality, etc. In these areas, the possibility of using stable, reproducible, and pocket devices can simplify the acquisition of data. Among recent prototypes, sensors based on laser-induced graphene (LIGE) on Kapton represent a feasible choice. In particular, LIGE devices are also exploited as electrodes for sensing in liquids. Despite a characterization with electrochemical (EC) methods in the literature, a closer comparison with traditional graphite electrodes is still missing. In this study, we combine atomic force microscopy with an EC cell (EC-AFM) to study, in situ, electrode oxidation reactions when LIGE or other graphite samples are used as anodes inside an acid electrolyte. This investigation shows the quality and performance of the LIGE electrode with respect to other samples. Finally, an ex situ Raman spectroscopy analysis allows a detailed chemical analysis of the employed electrodes.  相似文献

A partial least squares (PLS-1) calibration model based on kinetic—spectrophotometric measurement, for the simultaneous determination of Cu(II), Ni(II) and Co(II) ions is described. The method was based on the difference in the rate of the reaction between Co(II), Ni(II) and Cu(II) ions with 1-(2-pyridylazo)2-naphthol in a pH 5.8 buffer solution and in micellar media at 25°C. The absorption kinetic profiles of the solutions were monitored by measuring the absorbance at 570 nm at 2 s intervals during the time range of 0–10 min after initiation of the reaction. The experimental calibration matrix for the partial least squares (PLS-1) model was designed with 30 samples. The cross-validation method was used for selecting the number of factors. The results showed that simultaneous determination could be performed in the range 0.1-2 μg mL−1 for each cation. The proposed method was successfully applied to the simultaneous determination of Cu(II), Ni(II) and Co(II) ions in water and in synthetic alloy samples.   相似文献

A simple and non-separative analytical method for selective determination of amylose in Iranian rice has been developed. It was based on the reduction of silver ions by amylose and production of Ag nanoparticles, which exhibit surface plasmon resonance (SPR) spectra in the ultraviolet/visible region. The formation of Ag nanoparticles in the presence of amylose was monitored by transmission electron microscopy (TEM) and dynamic light scattering (DLS). The experimental conditions were optimized to obtain the highest yield for nanoparticle formation. Partial least square (PLS) regression as an efficient multivariate spectral calibration method was employed to make a connection between the SPR spectra of the generated Ag nanoparticles and the amylose content (AC) of the rice starch. The number of PLS latent variables was optimized by leave-one-out cross-validation utilizing prediction residual error sum of square (PRESS). The proposed model exhibited a high ability for prediction of amylose concentration in both standard starch samples and real rice samples prepared from different regions of Iran. The relative errors of prediction were almost lower than ±5% for different real samples and the detection limit was 3.23 weight percent of amylose in rice. In comparison to the reference method (Juliano method), the proposed method is simpler and does not need tedious sample preprocessing steps.  相似文献

Central composite design (CCD) and response surface methodology (RSM) were developed as experimental strategies for modeling and optimization of the influence of some variables on the performance of a new PVC membrane triiodide ion-selective electrode. This triiodide sensor is based on triiodide-clozapine ion-pair complexation. PVC, plasticizers, ion-pair amounts and pH were investigated as four variables to build a model to achieve the best Nernstian slope (59.9 mV) as response. The electrode is prepared by incorporating the ion-exchanger in PVC matrix plasticized with 2-nitrophenyl octal ether, which is directly coated on the surface of a graphite electrode. The influence of foreign ions on the electrode performance was also investigated. The optimized membranes demonstrate Nernstian response for triiodide ions over a wide linear range from 5.0 x 10(-6) to 1.0 x 10(-2)mol L(-1) with a limit of detection 2.0 x 10(-6) mol L(-1) at 25 degrees C. The electrodes could be used over a wide pH range 4-8, and have the advantages of easy to prepare, good selectivity and fast response time, long lifetime (over 3 months) and small interferences from hydrogen ion. The proposed electrode was successfully used as indicator electrode in potentiometric titration of triiodide ions and ascorbic acid.  相似文献

A two-dimensional optical storage (TwoDOS) format with binary modulation is being developed in which channel bits are arranged on a two-dimensional hexagonal lattice [W. M. J. Coene, in Optical Data Storage, Vol. 88 of OSA Trends in Optics and Photonics Series (Optical Society of America, Washington, D.C., 2003), pp. 90-92]. The aim is to increase the capacity by a factor of 2 and the data rate by a factor of 10 over third-generation Blu-ray Disc technology. Following a route similar to that used in one-dimensional conventional optical storage [Jpn. J. Appl. Phys. 42, 1074 (2003)] could lead to a further increase in capacity by the addition of another dimension to writing data, such as the use of multiple levels instead of the two levels (pit and land) used in the binary TwoDOS disk format. We present a nonlinear signal-processing model for signal waveform generation as a function of the M-ary channel symbols, as well as simulated signal readouts for multilevel TwoDOS.  相似文献

We present a facile and efficient method for modifying the surface of silica-coated Fe3O4 magnetic nanoparticles (MNPs) with bis(pyrazolyl) triazine ruthenium(II) complex [ MNPs@BPT–Ru (II) ] . Field emission-scanning electron microscopy, thermogravimetric/derivative thermogravimetry analysis, X-ray powder diffraction, Fourier-transform infrared spectroscopy, vibrating sample magnetometry, and energy-dispersive X-ray spectrometry analyses were employed for characterizing the structure of these nanoparticles. MNPs@BPT–Ru(II) nanoparticles proved to be a magnetic, reusable, and heterogeneous catalyst for the hydrogen transfer reduction of ketone derivatives. In addition, highly pure products were obtained with excellent yields in relatively short times in the presence of this catalyst. A comparison of this catalyst with those previously used for the hydrogen transfer reactions proved the uniqueness of MNPs@BPT–Ru(II) nanoparticle which is due to its inherent magnetic properties and large surface area. The presented method also had other advantages such as simple reaction conditions, eco-friendliness, high recovery ability, easy work-up, and low cost.  相似文献

The interaction of Love waves with square array of pillars deposited on a cavity defined in a 2D holey phononic crystal is numerically investigated using Finite Element Method. First, the existence of SH surface modes is demonstrated separately for phononic crystals that consist of square arrayed holes, or rectangular arrayed Ni pillars, respectively in, or on, a SiO2 film deposited on a ST-cut quartz substrate. The coupling between SH modes and torsional mode in pillars induces a transmission dip that occurs at a frequency located in the range of the band-gap of the holey phononic crystal. Second, a cavity is constructed by removing lines of holes in the holey phononic crystal and results in a transmission peak that matches the dip. The optimal geometrical parameters enable us to create a coupling of the cavity mode and the localized pillar mode by introducing lines of pillars into the cavity, which significantly improved the efficiency of the cavity without increasing the crystal size. The obtained results will pave the way to implement advanced designs of high-performance electroacoustic sensors based on coupling modes in phononic crystals.  相似文献

We associate to each infinite primitive Lie pseudogroup a Hopf algebra of ‘transverse symmetries,’ by refining a procedure due to Connes and the first author in the case of the general pseudogroup. The affiliated Hopf algebra can be viewed as a ‘quantum group’ counterpart of the infinite-dimensional primitive Lie algebra of the pseudogroup. It is first constructed via its action on the étale groupoid associated to the pseudogroup, and then realized as a bicrossed product of a universal enveloping algebra by a Hopf algebra of regular functions on a formal group. The bicrossed product structure allows to express its Hopf cyclic cohomology in terms of a bicocyclic bicomplex analogous to the Chevalley-Eilenberg complex. As an application, we compute the relative Hopf cyclic cohomology modulo the linear isotropy for the Hopf algebra of the general pseudogroup, and find explicit cocycle representatives for the universal Chern classes in Hopf cyclic cohomology. As another application, we determine all Hopf cyclic cohomology groups for the Hopf algebra associated to the pseudogroup of local diffeomorphisms of the line.  相似文献
