Supramolecular protein complexes are the corner stone of biological processes; they are essential for many biological functions. Unraveling the interactions responsible for the (dis)assembly of these complexes is required to understand nature and to exploit such systems in future applications. Virus capsids are well-defined assemblies of hundreds of proteins and form the outer shell of non-enveloped viruses. Due to their potential as a drug carriers or nano-reactors and the need for virus inactivation strategies, assessing the intactness of virus capsids is of great interest. Current methods to evaluate the (dis)assembly of these protein assemblies are experimentally demanding in terms of instrumentation, expertise and time. Here we investigate a new strategy to monitor the disassembly of fluorescently labeled virus capsids. To monitor surfactant-induced capsid disassembly, we exploit the complex photophysical interplay between multiple fluorophores conjugated to capsid proteins. The disassembly of the capsid changes the photophysical interactions between the fluorophores, and this can be spectrally monitored. The presented data show that this low complexity method can be used to study and monitor the disassembly of supramolecular protein complexes like virus capsids. However, the range of labeling densities that is suitable for this assay is surprisingly narrow.  相似文献

A detailed investigation on the effect of solvent polarity, temperature, and microwave irradiation on periselectivity in cycloaddition reactions of pentafulvenes with 3-oxidopyrylium betaine is described. The base catalyzed generation of 3-oxidopyrylium betaine in CHCl3 resulted in the exclusive formation of [6+3] adducts. With increase in solvent polarity and temperature, mixtures of [6+3] and [3+2] adducts were formed, where as under microwave irradiation, [3+2] adducts were formed exclusively. The experimental results have been rationalized on the basis of theoretical calculations.  相似文献

The shape of liquid drops on solid surfaces deviates from the spherical as tension decreases and gravity effects start affecting the drop shape. This paper attempts to define this deviation and estimates the dimensionless Eotvos number limits above which the deviation becomes "significant." The use of these limiting values can facilitate estimation of contact angle in the following manner. It is well known that the equilibrium contact angle made by a liquid drop on a solid surface can be estimated from measurements of two drop parameters. These parameters can be any two chosen from the drop volume, height, and wetted radius. In case the effect of gravity on the drop shape is negligible, simple algebraic relations derived from the spherical section assumption exist, from which the contact angle can be estimated. In systems where the "spherical section" assumption is invalid, the Laplace equation for the drop shape has been solved numerically with any two of the above parameters as the constraints, to obtain the contact angle. In this paper, Eotvos numbers at which the deviation of the drop profile from the spherical is significant enough to result in contact angle deviation of 1 degrees are estimated. The limiting values of Eotvos number, expressed as a function of the original contact angle made by the spherical profile, are obtained by solving the Laplace equation for the drop shape with the drop volume and wetted radius constraints for decreasing values of Interfacial tension. These limiting values are also estimated for different drop sizes and for cases where the drop phase is heavier (sessile) and lighter (buoyant) than the surrounding fluid. The independence of the Eotvos number estimates from the sign of the density difference as well as the drop size is shown. These Eotvos number limits can be used to check if the spherical section assumption, with the resulting simple algebraic relations, can be used for contact angle estimation and other shape-related analysis for a system.  相似文献

Three bisphenols containing cardo perhydrocumyl cyclohexylidene group, namely; 1,1-bis(4-hydroxyphenyl)-4-perhydrocumylcyclohexane, 1,1-bis(4-hydroxy-3-methylphenyl)-4-perhydrocumylcyclohexane and 1,1-bis(4-hydroxy-3,5-dimethylphenyl)-4-perhydrocumylcyclohexane were synthesized starting from p-cumyl phenol. Each of these bisphenols was polycondensed with both terephthaloyl chloride and isophthaloyl chloride by phase transfer-catalyzed interfacial polymerization to obtain a series of new aromatic polyesters. Inherent viscosities and number average molecular weights of polyesters were in the range 0.51-0.64 dL/g and 17390-41430?g/mol, respectively which indicated the formation of reasonably high molecular weight polymers. The detailed NMR studies revealed that axial and equatorial identity of the phenyl rings of bisphenols was retained in polyesters resulting in constitutional isomerism. Polyesters containing perhydrocumyl cyclohexylidene groups showed excellent solubility in organic solvents viz, chloroform, dichloromethane, 1,1,2,2-tetrachloroethane and tetrahydrofuran. The self-standing films of polyesters could be cast from their chloroform solution. The 10% weight loss temperatures and glass transition temperatures of polyesters were in the range 453–485?°C and 201–267?°C, respectively demonstrating their excellent thermal characteristics. The gas permeability study of polyesters was carried out for He, H2 and N2 by variable-volume method. An improvement in permeability and decrease in selectivity was observed due to symmetric methyl substituents while reverse trend was observed in case of polyesters with asymmetric methyl substituents.  相似文献

In search of 2,7‐ethylene‐bridged naphthalenophanes with desymmetrized naphthalene cores as inherently chiral cyclophanes, nucleophilic substitution of 1‐bromo‐7‐(bromomethyl)‐2‐[(trimethylsilyl)methyl]naphthalene, a desymmetrized building block bearing an electrophilic group (CH2Br) and a masked nucleophilic functionality (CH2TMS) which can be activated by fluoride anion, was examined. As a result, in contrast to the case of parent naphthalenophanes wherein the smallest [2.2]naphthalenophane was obtained as the major product, only [2.2.2.2](2,7)‐1‐bromonaphthalenophane was obtained albeit in low yields, whereas the corresponding [2.2]‐ or [2.2.2]naphthalenophanes were not obtained. Though the [2.2.2.2]‐1‐bromonaphthalenophane can adopt four idealized geometries of different symmetry, among which three are inherently chiral, theoretical calculations predict that three conformers have almost equal energy and may equilibrate in solution. The X‐ray crystallographic study shows that it adopts a C2 conformation with anti,anti,anti orientation of the C?Br bonds at least as a major component in crystal.  相似文献

Available online Development of water soluble AIE-active “light-up” bioprobes for the detection of biomacromolecules has drawn huge research interests in recent past. In this study, a series of ethylene glycol modified water soluble tetrameric tetraphenylethylene amphiphiles with pyridinium polar heads (TPE-xEG-Py, x = 3, 4, 6 or 1a-c) have been synthesized by varying the ethylene glycol spacer. Their unique structure allows them to form vesicles and other nanoaggregates in aqueous solutions. These amphiphiles were successfully utilized for fluorimetric detection and quantitation of BSA and DNA based on the electrostatic interactions to trigger AIE-emission from the TPE moiety. The electrostatic interaction was also proved very effective in wash-free imaging of both Gram-negative (E. coli) and Gram-positive (S. aureus) bacteria with up to 92 folds increase in fluorescence response within bacterial concentration 0–12 × 108 CFU mL?1. The strategy is advantageous due to cost-effective and easy synthesis, high water solubility, and fast response.  相似文献

The syntheses, structures, and N2 adsorption properties of six new supramolecular metallacycles are reported. Flexible ditopic linkers, 1-4, with systematically varied lengths and conformational degrees of freedom were synthesized utilizing ester linkages. They were used in combination with (dppp)M(OTf)2, where M = Pt(II) and Pd(II), and cis-(Me3P)2Pt(OTf)2 to form flexible supramolecular metallacycles 5-10 in 88-98% isolated yields. Their structures were characterized via multinuclear NMR and X-ray crystallography. The metallacycles stack to form porous structures in the crystalline state. The pore dimensions depend on both the phosphorus ligands attached to the metals and the flexible linkers. Adsorption studies on the porous materials show that 5a, 6, 8, and 9 held 11.7, 16.5, 5.7, and 6.8 cm3/g STP of N2 at 77 K, respectively. A guest-exchange study with nitromethane and toluene reveals that the nanopore in 5 is flexible, a property which was transferred from the linker to the supramolecular structure in the solid state.  相似文献

A series of platinum(II) complexes with the formulas Pt(diimine)(pip(2)NCNH(2))(L)(2+) [pip(2)NCNH(2)(+) = 2,6-bis(piperidiniummethyl)phenyl cation; L = Cl, Br, I, NCS, OCN, and NO(2); diimine = 1,10-phenanthroline (phen), 5-nitro-1,10-phenanthroline (NO(2)phen), and 5,5'-ditrifluoromethyl-2,2'-bipyridine (dtfmbpy)] were prepared by the treatment of Pt(pip(2)NCN)Cl with a silver(I) salt followed by the addition of the diimine and halide/pseudohalide under acidic conditions. Crystallographic data as well as (1)H NMR spectra establish that the metal center is bonded to a bidentate phenanthroline and a monodentate halide/pseudohalide. The pip(2)NCNH(2)(+) ligand with protonated piperidyl groups is monodentate and bonded to the platinum through the phenyl ring. Structural and spectroscopic data indicate that the halide/pseudohalide group (L(-)) and the metal center in Pt(phen)(pip(2)NCNH(2))(L)(2+) behave as Br?nsted bases, forming intramolecular NH···L/NH···Pt interactions involving the piperidinium groups. A close examination of the 10 structures reported here reveals linear correlations between N-H···Pt/L angles and H···Pt/L distances. In most cases, the N-H bond is directed toward the Pt-L bond, thereby giving the appearance that the proton bridges the Pt and L groups. In contrast to observations for Pt(tpy)(pip(2)NCN)(+) (tpy = 2,2';6',2"-terpyridine), the electrochemical oxidation of deprotonated adducts, Pt(diimine)(L)(pip(2)NCN), is chemically and electrochemically irreversible.  相似文献

A simple, mild, and ecofriendly method has been developed for the synthesis of 3-vinylchromones from 4-oxo-4H-1-benzopyran-3-carboxaldehyde (3-formylchromone) by simple Knoevenagel condensation with various active methylene compounds (AMC) in aqueous micellar media in the presence of catalytic amounts of cetyl trimethylammonium bromide (CTAB) and 1,4-diazabicyclo[2.2.2]octane (DABCO). In the case of malonic acid as AMC, the reaction resulted in formation of only Doebner decarboxylated products under the standard reaction condition. It has been also observed that 3-formylchromone derivatives primarily undergo tandem Knoevenagel and Michael reactions in the presence of > 2 equiv. of ethyl acetoacetate to produce benzophenone derivatives, by opening of pyran ring, as the sole product in good yields.  相似文献
