Liquid chromatographic-mass spectrometric analysis for screening of patients with cystinuria, and identification of cystine stone.

Analyses of amino acids in the urine of a normal human and of patients with heterozygous and homozygous cystinuria have been carried out, using liquid chromatography-mass spectrometry with an atmospheric pressure ionization interface system. A kidney cystine stone was also analysed by this system. Very intense quasi-molecular ions ([M + H]+) of standard cystine, arginine, lysine and ornithine were observed on mass chromatograms as base peaks. Mass chromatograms of the urine samples from a normal human and from patients with heterozygous and homozygous cystinuria were easily distinguishable. The retention times in the mass chromatogram and mass spectrum of kidney stone cystine was almost the same as that of authentic cystine.     

POLYETHYLENE GLYCOL-MODIFIED HEMATOPORPHYRIN AS PHOTOSENSITIZER

Abstract— Hematoporphyrin, having two carboxylic groups, was coupled with α-(3-aminopropyl)-ω-methoxypoly(oxyethylene), PEG-NH₂, through acid-amide bond formed with carbodtimide. PEG-modified hematoporphyrin was readily soluble not only in neutral aqueous solution but also in organic solvents. Its absorption spectrum showed a sharp band at 376 nm in neutral aqueous solution and at 403 nm in benzene. Modified hematoporphyrin acted as a photosensitizer; imidazole and indole were photooxidized in organic solvents such as benzene or chloroform, and uric acid was also photooxidized in neutral aqueous solution.     

Radiation-induced emission from golden hamster embryo cells

Emission from high-energy-electron-irradiated golden hamster embryo (GHE) cells has been studied over the temperature range 12–300 K both by a one-shot-single-photon-counting method and by photocurrent measurements with an oscilloscope. Emission from the irradiated phosphate buffered saline (PBS) also has been studied. The emission spectra from PBS at 12 and 77 K show a maximum around 330 and 380 nm, respectively, which are the same spectra as those from irradiated pure H₂O. The emission from irradiated GHE consists of the new band at 480 nm in addition to the emission from H₂O. The 480 nm emission is observed at the temperature range of 12–300 K, though the emission at 300 K is much lower than that at low temperature. The 480 nm emission is ascribed to the transition from excited organic substances in GHE cells. The intensity of 480 nm emission at 300 K increases linearly with increasing irradiation-dose in the range of 11–600 Gy.     

Study on the structures of 2-substituted iminothiazolidine derivatives

The reaction of 2-bromoethylamine 1 with methylisothiocyanate 2 under mild condition gave 2-methyl-amino-2-thiazoline 3 as the major product together with two kinds of byproducts, 3-(N-methylthiocar-bamoyl)-2-methyliminothiazolidine 4 and N,N′-dimethyl-N-(2-thiazolin-2-yl)thiourea 5. Thermal isomer-ization of 5 to 4 was observed. The structures of the byproducts were confirmed by X-ray crystallography.     

Multiplex amplified product-length polymorphism analysis of 36 mitochondrial single-nucleotide polymorphisms for haplogrouping of East Asian populations

We present a reliable, rapid, and economical multiplex amplified product-length polymorphism (APLP) method for analyzing the haplogroup-diagnostic mitochondrial single-nucleotide polymorphisms (mtSNPs) in East Asian populations. By examining only 36 haplogroup-specific mtSNPs in the coding region by using four 9-multiplex polymerase chain reaction (PCR) and subsequent electrophoresis, we could safely assign 1815 individuals from 8 populations of Japanese, Korean, Chinese, and Germans to 45 relevant haplogroups. This multiplex APLP analysis of coding-region mtSNPs for haplogrouping is especially useful not only for molecular phylogenetic studies but also for large-scale association studies due to its rapid and economical nature. This is the first panel of mtSNPs in the coding region to be used for haplogrouping of East Asian populations.     

Rate of spontaneous deactivation of styryl and α-methylstyryl carbanions

The deactivation of α-methylstyryl and styryl carbanions followed by spectral change and titration was studied kinetically. First-order rate constants were obtained for the deactivation of oligo-α-methylstyryl lithium, disodium, and dipotassium in tetrahydrofuran and of polystyryl potassium in varying degrees of polymerization in benzene that contained a small amount of tetrahydrofuran and n-hexane. It was observed that the addition of cryptate [222] exerts a significant effect on the system of oligo-α-methylstyryl disodium in tetrahydrofuran. The effect of dilution, degree of polymerization of the polymer, and counterions on the rate of deactivation of carbanions was discussed.     

Application of samarium diiodide (SmI2)-induced reduction of gamma-acetoxy-alpha,beta-enoates with alpha-specific kinetic electrophilic trapping for the synthesis of amino acid derivatives

Gamma-acetoxy-alpha,beta-enoates were easily reduced by samarium diiodide (SmI2) in THF to generate samarium dienolates which were kinetically trapped with ease at their alpha-positions by electrophiles (proton, aldehydes or ketones) to yield (E)-alkene dipeptide isosteres or gamma-amino acid derivatives in high chemical yields.     

Oxidase/peroxidase bilayer-modified electrodes as sensors for lactate,pyruvate, cholesterol and uric acid

Enzyme heterobilayer-modified electrodes were fabricated by successively covalently binding to the surface of a tin(IV) oxide plate horseradish peroxidase (HRP), then an oxidase (lactate, pyruvate or cholesterol oxidase or uricase), which liberates hydrogen peroxide by reaction with the respective substrate. The cooperative action of oxidase-HRP leads to an efficient amperometric sensor system with the minimum amount of enzyme immobilized on an electrode.     

Chemo-enzymatic Synthesis of Novel Oligo-N-acetyllactosamine Derivatives having a β(1-4)–β(1-6) Repeating Unit by Using Transition State Analogue Substrate

Chitinase-catalyzed hydrolytic and transglycosylating behavior of 1,2-oxazoline derivative of N-acetyllactosamine (LacNAc-oxa) 1 has been investigated. An extremely rapid hydrolysis (ring-opening of the oxazoline moiety) could be observed, suggesting that 1 behaves as a transition state analogue substrate for chitinase A1 (Bacillus circulans WL-12). This disaccharide monomer 1 was found to polymerize under basic conditions, giving rise to novel oligosaccharides having a β(1-4)–β(1-6) repeating unit in the main chain. The degree of polymerization of the resulting oligosaccharides was up to 5. This is the first example of enzymatic glycosylation reaction forming a β(1-6) bond catalyzed by chitinase.
