Starting from 6-aryl-4-oxo-2-thioxo-1,2,3,4-tetrahydropyrimidine-5-carbonitrile (4a-d), a series of mono- and dialkyl derivatives 5a-j and 6a, b was synthesized. Hydrazinolysis of 4a, b, d and 5d afforded the hydrazino derivatives 7a-c which were cyclised to give the triazolopyrimidinones 8a-c and the pyrimidotriazinones 9a-c through the reaction with formic acid and chloroacetyl chloride, respectively. Most of the newly synthesized compounds were evaluated for their in-vitro antitumor activity. Compounds 6a and b displayed promising anticancer activity against leukaemia, non-small cell lung, melanoma, and renal cancer. On the other hand, all compounds prepared were screened for their in-vitro antibacterial and antifungal activities. Compounds 5h and j showed significant activity against Staphylococcus aureus, while compounds 5e, 7c and 8c displayed moderate inhibitory activity against Candida albicans.  相似文献

Verbena rigida L., Verbena tenera Spreng. and Verbena venosa L. were investigated for their flavonoid content. Analysis was carried out by high-performance liquid chromatography coupled to diode array UV detection (LC–UV), using different techniques, also using post-column addition of shift reagents, afforded precise structural information about the position of the free hydroxyl groups in the flavonoid nucleus. LC–MS using atmospheric pressure chemical ionization (APCI) in the positive mode provided the molecular weight, the number of hydroxyl groups, the number of sugars and an idea about the substitution pattern of the flavonoid. On-line UV and MS data demonstrated the presence of orientin, vitexin, isovitexin, luteolin, luteolin 7-O-glucoside, apigenin 7-O-glucoside in addition to luteolin, chryseriol and apigenin aglycones in the three Verbena species with different concentrations. Quantitative determination of flavonoid content revealed the presence of 69.84 mg/g dry sample, 88.26 mg/g dry sample and 85.82 mg/g dry sample total flavonoid compounds in V. rigida L., V. tenera Spreng. and V. venosa L., respectively. The method developed for identification is useful for further chromatographic fingerprinting of plant flavonoids.  相似文献

Herein, we report the synthesis, characterization, and preliminary pharmacological activity of a new series of substituted pyrazolopyridazine derivatives. Compound 1 was reacted with ethoxymethylene malononitrile 2 in refluxing ethanol to give the corresponding compound 3 , which was treated with hydrazine hydrate or formamide to give pyrazolo[3,4‐c]pyrazole 4 and pyrazolo pyrimidine 5 derivatives, respectively. Also, compound 3 was reacted with NH4SCN or carbon disulphide or ethyl acetoacetate to yield the corresponding pyrazolo derivatives 6 , 7 , 8 , respectively. Additionally, compound 3 was reacted with triethyl orthoformat in acetic anhydride to give 9 , which was treated with hydrazine hydrate to give hydrazino derivative 10 . The latter compound transformed into the pyrazolo[4,3‐e][1,2,4]triazolo[1,5‐c]‐pyrimidine 11 via refluxing with acetic anhydride. Finally, compound 9 was reacted with benzoic acid hydrazide or mercapto acetic acid to give compounds 12 and 13 , respectively. The latter compound was treated with refluxing ethanolic sodium ethoxide solution to afford the pyrazolothiazolopyrimidine 14 . Some of the compounds exhibited better activities as anti‐inflammatory and antimicrobial agents than the reference controls. The detailed synthesis, spectroscopic data, anti‐inflammatory, and antimicrobial activities of the synthesized compounds was reported.  相似文献

A bacteriocin-producing strain (9,000 AU/ml) was isolated from the rhizosphere of Algerian healthy plants Ononis angustissima Lam. and identified as Bacillus clausii strain GM17. The bacteriocin, called Bac-GM17, was purified from the culture supernatant after heat treatment, ammonium sulfate precipitation, Sephadex G-50 chromatography and Mono Q fast-performance liquid chromatography (FPLC). Based on matrix-assisted laser desorption ionization-time of flight mass spectrometry analysis, the purified Bac-GM17 is a monomer protein with a molecular mass of 5,158.11 Da. The N-terminal sequencing allowed for the straightforward identification of its first 20 residues, which were of pure bacteriocin. It also revealed that this bacteriocin contained a unique sequence, namely DWTCSKWSCLVCDDCSVELT, which suggests the identification of a novel compound. Bac-GM17 was extremely heat stable (20 min at 120 °C) and was stable within the pH range (3–9). It was found to be resistant to the proteolytic action of trypsin, pepsin, papain, pronase E, and proteinase K. It was also noted to display a bactericidal mode of action against Agrobacterium tumefaciens C58 and a fungistatic mode of action against Candida tropicalis R2 CIP203.  相似文献

The synthesis and investigation of fluorescence and antimicrobial properties of a new series of 1,2,3-triazoles were described. Acetylenes 4a–c were resulted via alkylation of 2-oxonicotinonitriles 3a–c with propargyl bromide in base medium. [2?+?3] cycloaddition of acetylenes 4a–c with ethyl 2-azidoacetate, p-acetylphenylazide, and p-tolylsulfonylazide in the presence of Cu(I) afforded 1,2,3-triazoles 5a–c, 7a–c, and 9, respectively (via click reaction). The triazoles 5a–c were subjected to saponification process to give the acids 6a–c. The fluorescence and antimicrobial properties of triazoles 5a–c, 7a–c, and 9 were investigated and significant results were obtained.  相似文献

Forty-eight derivatives of phorbol (9) and isophorbol (14) were evaluated for their inhibition of human immunodeficiency virus (HIV)-1 induced cytopathic effects (CPE) on MT-4 cells, as well as their activation of protein kinase C (PKC), as indices of anti-HIV-1 and tumor promoting activities, respectively. Of these compounds, the most potent inhibition of CPE was observed in 12-O-tetradecanoylphorbol 13-acetate (8) and 12-O-acetylphorbol 13-decanoate (6). The former also showed the strongest PKC activation activity, while the latter showed no activity at 10 ng/ml. Both activities were generally observed in those phorbol derivatives with an A/B trans configuration, but not in the isophorbol derivatives with an A/B cis configuration. Acetylation of 20-OH in the phorbol derivatives significantly reduced the inhibition of CPE, as shown in 12-O-, 20-O-diacetylphorbol 13-decanoate (6a) (IC100=15.6 microg/ml) vs. compound 6 (IC100=0.0076 microg/ml), and 12-O-tetradecanoylphorbol 13,20-diacetate (8a) (IC100=15.6 microg/ml) vs. 12-O-tetradecanoylphorbol 13-acetate (8) (IC100=0.00048 microg/ml), except in the case of 12-O-decanoylphorbol 13-(2-methylbutyrate) (4) and phorbol 12,13-diacetate (9c). The reduction of a carbonyl group at C-3 abruptly reduced the inhibition of CPE, as observed in 3beta-hydroxyphorbol 12,13,20-triacetate (9f) (IC100=500 microg/ml) vs. phorbol 12,13,20-triacetate (9d) (IC100=62.5 microg/ml). Although 8 was equipotent in the inhibition of CPE, and activation of PKC, both activities were abruptly decreased by the acetylation of 20-OH and methylation of 4-OH [as in 8a and 4-O-methyl-12-O-tetradecanoylphorbol 13,20-diacetate (8b), respectively]. On the other hand, its positional isomer (12-O-acetylphorbol 13-tetradecanoate (8c) showed neither activities. The removal of a long acyl group in 8 led to a substantial loss of both activities, as shown in phorbol 13-acetate (9b). Of the 12-O-acetyl-13-O-acylphorbol derivatives, the highest inhibition of CPE was observed in 6, which has a dodecanoyl residue at C-13. Both an increase and decrease in the number of fatty acid carbon chains resulted in significant reduction of the inhibition of CPE.  相似文献

Summary. We describe the effect of the solvent during the intra-cyclization of the arylhydrazonomalononitrile to obtain new and useful substituted cinnolines, namely 4-amino-3-cyanocinnolines and (4-amino-cinnolin-3-yl)arylmethanones. 4-Amino-3-cyano-5,7-dimethyl-cinnoline was treated with n-hexylamine, cyclohexylamine, and isopropylamine to yield the corresponding cinnoline-3-carboxamidine derivatives. Furthermore, 4-amino-3-cyano-5,7-dimethylcinnoline was reacted with diaminoethane to give 4-amino-5,7-dimethyl-3-(4,5-dihydroimidazol-2-yl)cinnoline. Treatment of 4-amino-3-cyano-5,7-dimethylcinnoline with hydrazine hydrate provided 3-amino-7,9-dimethyl-1H-pyrazolo[4,3-c]cinnoline. Moreover, (4-aminocinnolin-3-yl)phenylmethanones were treated with hydrazine hydrate to give 3-phenyl-1H-pyrazolo[4,3-c]cinnolines. Finally, reactions of (4-aminocinnolin-3-yl)phenylmethanone with 3,5-dimethylaniline, phenyl hydrazine, hydroxylamine, and phenyl isothiocyanate are discussed. Chemical and spectroscopic evidence for the structures of the new compounds is presented.  相似文献

Vitellogenin (VTG) is a protein produced by the liver of oviparous animals. It is being used as a biomarker for exposure to endocrine disruptors in many species. Rainbow trout Vtg has recently been sequenced by the conventional cDNA nucleotide approach. We focused on protein characterization of the intact protein and its derived tryptic and cyanogen bromide peptides by matrix-assisted laser desorption/ionisation and electrospray ionisation mass spectrometry. The molecular mass of the intact protein was found to be 183127 Da. A large number of unidentified peptide ions encourage further structural analysis to propose possible sequence variants and post-translational modifications.  相似文献

Complex equilibria of DCPHD with proton and Ni2+, Sm3+, and Yb3+ ions has been measured in various mixed aqueous solvents, viz: isopropanol-water, acetone-water, ethanol-water, and methanol-water. Based on potentiometric equilibrium measurements of hydrogen ion concentration at 30°C, ionic strenght 0.10 M (KNO3) and in the above various mixed aqueous solvents, the values of the protonation constants of DCPHD and the stability constants of DCPHD with Ni2+, Sm3+, and Yb3+ have been evaluated. The variation of protonation and stability constants with the inverse of dielectric constant or mole fraction of the solvent was studied. Application of Fuoss expression and consideration of electrosatic and non-electrostatic effects are made to explain the values of the constants.  相似文献

The synthesis and quantitative equilibrium studies of two new azo-pyridazine ligands, namely the pyruvic (PHP) and hippuric (HipHP) acid derivatives are described in detail. Their acid-base and metal-ligand equilibrium constants in 75% dioxan-water solvent at 30 °C have been calculated from potentiometric and spectrophotometric data. The probable structures are inferred from electronic and vibrational spectral results. Evidence is presented for the formation of protonated complexes withPHP.

Zusammenfassung Die Synthese und quantitative Gleichgewichtsuntersuchungen von zwei neuen Azo-pyridazin-Liganden, die von Brenztraubensäure (PHP) bzw. Hippursäure (HipHP) abzuleiten sind, werden im Detail beschrieben. Die entsprechenden Säure-Base- und Metall-Ligand-Gleichgewichtskonstanten in 75% Dioxan-Wasser bei 30 °C wurden aus potentiometrischen und spektrophotometrischen Daten ermittelt. Die wahrscheinlichen Strukturen wurden von Elektronen- und IR-spektroskopischen Daten abgeleitet. Es sind Beweise vorhanden, daß mitPHP protonierte Komplexe gebildet werden.
