Modulation of the PGE2-Mediated Pathway in the Eclosion Blocking Effect of Flumethrin and Terpenoid Subfraction Isolated from Artemesia nilagirica in Rhipicephalus annulatus

Prostaglandins are a group of important cell-signaling molecules involved in the regulation of ovarian maturation, oocyte development, egg laying and associated behaviors in invertebrates. However, the presence of prostaglandin E₂ (PGE₂), the key enzymes for PGE₂ biosynthesis and its interference by drugs were not investigated previously in the ovary of ticks. The present study was undertaken to assess the modulation of the PGE₂-mediated pathway in the eclosion blocking effect of flumethrin and terpenoid subfraction isolated from Artemisia nilagirica in Rhipicephalus annulatus ticks. The acaricidal activities and chemical profiling of the terpenoid subfraction were performed. The localization of the cyclooxygenase1 (COX1) and prostaglandin E synthase (PGES) enzymes and the quantification of PGE₂ in the ovaries of the ticks treated with methanol (control), flumethrin and terpenoid subfraction were also undertaken. In addition, the vitellogenin concentration in hemolymph was also assayed. Both flumethrin and the terpenoid subfraction of A. nilagirica elicited a concentration-dependent inhibition of fecundity and blocking of hatching of the eggs. The COX1 could not be detected in the ovaries of treated and control ticks, while there was no significant difference observed in the concentration of vitellogenin (Vg) in them. The presence of PGES in the oocytes of control ticks was confirmed while the immunoreactivities against PGES were absent in the vitellogenic oocytes of ticks treated with flumethrin and terpenoid subfraction. The levels of PGE₂ were below the detection limit in the ovaries of the flumethrin-treated ticks, while it was significantly lower in the ovaries of the terpenoid subfraction-treated ticks. Hence, the prostaglandin E synthase and PGE₂ were identified as very important mediators for the signaling pathway for ovarian maturation and oviposition in ticks. In addition, the key enzyme for prostaglandin biosynthesis, PGES and the receptors for PGE₂ can be exploited as potential drug targets for tick control. The detection of PGES by immunohistochemistry and quantification of PGE₂ by LC-MSMS can be employed as valuable tools for screening newer compounds for their eclosion blocking acaricidal effects.     

CuSO4/sodium ascorbate catalysed synthesis of benzosuberone and 1,2,3-triazole conjugates: Design,synthesis and in vitro anti-proliferative activity

A novel series of conjugates of benzosuberone and 1,2,3-triazole i.e. 3-(4-phenyl-1H-1,2,3-triazol-1-yl)propyl-9-chloro-2,3-dimethyl-6,7-dihydro-5H-benzo[7]annulene-8-carboxylic acids (8a-j) were synthesized in good to excellent yields catalysed by CuSO₄ under milder reaction conditions and evaluated for their anti-proliferative activity. The structural elucidation of the prepared compounds was carried out using IR, ¹H NMR, ¹³C NMR and Mass spectral analysis. The newly synthesized derivatives (8a-j) were evaluated for their anti-proliferative activity against four human cell lines and the novel derivatives showed moderate to excellent activity. The obtained results suggest that these compounds can be considered as new hits for anti-proliferative drug development programme and further SAR studies can help obtain better anticancer agents.     

Camouflaging Structural Diversity: Co‐crystallization of Two Different Nanoparticles Having Different Cores But the Same Shell

Two ligand‐protected nanoscale silver moieties, [Ag₄₆(SPhMe₂)₂₄(PPh₃)₈](NO₃)₂ and [Ag₄₀(SPhMe₂)₂₄(PPh₃)₈](NO₃)₂ (abbreviated as Ag₄₆ and Ag₄₀, respectively) with almost the same shell but different cores were synthesized simultaneously. As their external structures are identical, the clusters were not distinguishable and become co‐crystallized. The occupancy of each cluster was 50 %. The outer shell of both is composed of Ag₃₂S₂₄P₈, which is reminiscent of fullerenes, and it encapsulates a well‐studied core, Ag₁₄ and a completely new core, Ag₈, which correspond to a face‐centered cube and a simple cube, respectively, resulting in the Ag₄₆ and Ag₄₀ clusters. The presence of two entities (Ag₄₀ and Ag₄₆ clusters) in a single crystal and their molecular formulae were confirmed by detailed electrospray ionization mass spectrometry. The optical spectrum of the mixture showed unique features which were in good agreement with the results from time‐dependent density functional theory (TD‐DFT).

Enantioselective synthesis of oseltamivir phosphate

The key steps in the enantioselective synthesis of Tamiflu include an asymmetric Diels-Alder reaction, Mitsunobu inversion using Fukuyama modified Weinreb reagent, carbamate directed epoxidation. Epoxide opening with trimethylsilyl azide furnished a 3:1 mixture of regioisomers that converged to afford the same aziridine. Attempted preparation of the unsaturated ester regioselectively using 2-iodoxybenzoic acid (IBX) following Nicolaou’s protocol failed. The unsaturated ester was prepared by phenylselenylation followed by selenoxide elimination.     

Green chemical multicomponent approach for the synthesis of C3-pyranopyrazole-substituted coumarins

An efficient and rapid method was developed for the synthesis of C3-pyranopyrazole-substituted coumarins from one-pot five-component reaction of β-keto ester, hydrazine, O-hydroxy aromatic aldehydes, 6-methyl, 4-hydroxy pyranone and aromatic aldehyde in the presence of tri-ethylamine in solvent-free conditions. The microwave-assisted method reported herein offers advantageous shorter reaction times, higher yields and cleaner reaction compared with conventional heating methods.     

Influence of pH and microwave calcination on the morphology of KGd(WO<Subscript>4</Subscript>)<Subscript>2</Subscript> particles derived by Pechini Sol–Gel method

KGd(WO₄)₂ (KGW) particles were synthesized at 3.5, 5.5 and 7.5 pH values by Pechini polymeric complex sol–gel method using potassium nitrate, gadolinium nitrate, ammonium paratungstate, citric acid and ethylene glycol as starting materials. Deionized water was used as solvent. Polymeric precursor gel was formed with citric acid as complexing agent and ethylene glycol as binder. Synthesized gel was analyzed by FT-IR spectroscopy. Prepared precursor gels were further annealed using resistive and microwave processes at 550 and 700 °C, respectively. The properties of heat treated samples were characterized by powder XRD, FT-IR, Raman and SEM analysis to understand the crystallinity, organic liberation, tungstate ribbon formation and surface morphology, respectively. The phase formation and different phases of intermediate oxides in pre-fired samples were investigated by powder XRD. Organic liberation in the samples in relation to temperature, and the carbon content in the pre-fired powder was analyzed using FT-IR spectrum. Raman spectrum reveals the formation of tungsten ribbons as well as the quality of the samples. The morphological changes at different synthesis conditions were observed with SEM micrographs.     

Sunlight promoted palladium catalysed Mizoroki-Heck, Suzuki-Miyaura and Sonogashira reactions

The palladium catalysed Mizoroki-Heck, Suzuki-Miyaura and Sonogashira reactions were successfully carried out under irradiation with sunlight. The Heck reaction gives considerable amount of Z product due to photochemical isomerization of initially formed E alkenes. Reaction of methyl 2-iodobenzoate with acrylamide under solar condition furnished 2H-2-benzazepine-1,3-dione rather than the expected derivative of cinnamate while the same reaction with ethyl 2-iodobenzoate gave the desired cinnamide.     

Pd(II) inhibition during hexacyanoferrate(III) oxidation of sugars: A kinetic study

An inhibition effect of PdCl₂ on the rate of oxidation of sugars by alkaline hexacyanoferrate(III) has been observed. The order of reactions in hexacyanoferrate(III) and OH⁻ is zero and unity, respectively, while that in sugars decreases from unity at higher sugar concentration. The kinetic data and spectrophotometric evidence support the formation of {Pd^II − (sugar)} and {Pd^II − sugar)₂} complexes and their resistance to react with Fe(CN)₆³⁻ © 1996 John Wiley & Sons, Inc.
