Staphylococcal protein A (protein A) is an important protein frequently used in research studies within the fields of biomedicine and biotechnology. Due to some limitations in available protein purification methods which can hold the native structure of the protein A without changing the folding or adding histidine to structure of this protein, its separation in the native form is difficult. In this study, a new cost-effective and powerful technique was introduced for separation of the full-length and truncated forms of recombinant protein A, without any alteration in their 3D structures. Per aqueous liquid chromatography with bare silica gel stationary phase and water:acetonitrile as the mobile phase was proved to be an attractive choice among the range of separation methods. Similar to hydrophilic liquid chromatography, this method employs high percentage of water in mobile phase. The effects of mobile phase composition, pH, and salt concentration on the retention behavior of protein A on bare silica gel stationary phases were investigated. In this method, applying high amounts of aqueous solvent accompanied by a minimum percentage of organic solvent could successfully separate protein A with preservation of folding, and any affinity-tagged group such as histidine has not occurred on its structure. Purity of the fractions obtained by the proposed method was confirmed using SDS-PAGE, western blotting, and matrix-assisted laser desorption ionization time-of-flight mass spectrometry. Cross-linked polystyrene (PS) particles having red blood corpuscle (RBC)-like shape were synthesized by one-pot dispersion polymerization of styrene with ethanol/water mixture and ethylene glycol dimethacrylate (EGDMA) as the reaction medium and cross-linker, respectively. Monitoring of the reaction showed that RBC-like shape forms due to asymmetric shrinkage of a cross-linked network during the phase separation. In addition, three dimensional phase diagram was generated based on the yielded data that showed that the formation of such unique shape extremely depends on the polarity of the medium and injection time of the cross-linker. In situ synthesis of RBC-like particles, as promising biomaterials in targeted drug delivery and a model for the understanding of the cell behavior, via such fast and high solid content approach makes it to be conducive to subsequent scale up, i.e. potential commercial adoption. 相似文献

The present work deals with numerical investigations on heat transfer characteristics and friction factor of aqueous CuO nanofluids flow in a set of four microchannels connected in parallel under laminar regime. For each single phase, volume of fluid, mixture and Eulerian models, a particular computer code is developed to carefully simulate this problem. The three-dimensional steady-state governing equations are solved through finite volume method. The primary aim of this study is to comparatively distinguish the most appropriate and accurate model for numerical studies of nanofluids in microchannels. The results are compared with one another and the data obtained from an experimental work. Regarding the results, an acceptable consistency is observed for all models with the experimental data. The current study truly demonstrates that applying single-phase model to simulate and evaluate the laminar flow of CuO–water nanofluid inside microchannels with uniform wall temperature is more modest, precise and reliable compared with two-phase models.
A novel SBA-15-based fluorescent sensor, SBA-PI: mesoporous SBA-15 structure modified with iminostilbene groups, was designed, synthesized, and characterized by Fourier transform-infrared spectroscopy (FT-IR), ultraviolet–visible spectroscopy, field emission scanning electron microscopy (FESEM), transmission electron microscopy (TEM), energy-dispersive X-ray spectroscopy (EDS), thermogravimetric analysis (TGA), low-angle X-ray diffraction techniques (low-angle XRD), and N2 adsorption–desorption techniques. The SBA-PI as a sensor with a selective behavior for detection of Cu2+ comprises iminostilbene carbonyl as the fluorophore group. The SBA-PI sensor displays an excellent fluorescence response in aqueous solutions and the fluorescence intensity quenches remarkably upon addition of Cu2+. Other common interfering ions even at high concentration ratio showed either no or very small changes in the fluorescence intensity of SBA-PI in the absence of Cu2+. A limit of detection of 8.7 × 10−9 M for Cu2+ indicated that this fluorescence sensor has a high sensitivity and selectivity toward the target copper (II) ion. The fabricated Cu2+ sensor was successfully applied for the determination of the Cu2+ in human blood samples without any significant interference. With the selective analysis of Cu2+ ions down to 0.9 nM in blood, the sensor is a promising and a novel detection candidate for Cu2+ and can be applied in the clinical laboratory. A reversibility and accuracy in the fluorescence behavior of the sensor was found in the presence of I¯ that was described as a masking agent for Cu2+.
