Tobacco smoking is a preventable main cause of fatal diseases. Accurate measurements of the effects it has on neurotransmitters are essential in developing new strategies for smoking cessation. Moreover, measurements of neurotransmitter levels can aid in developing drugs that counteract the effects of smoking. The aim of this study is to develop and validate a fast, simultaneous and sensitive method for measuring the levels of neurotransmitters in rat brain after the exposure of tobacco cigarettes. The selected neurotransmitters include dopamine, GABA, serotonin, glutamine and glutamate. The method is based on high-performance liquid chromatography–tandem mass spectrometry. Chromatographic separation was achieved within 3 min using a Zorbax SB C18 column (3.0 × 100 mm, 1.8 μm particle size). The mobile phase consisted of HPLC-grade water and acetonitrile each containing 0.3% heptafluorobutyric acid and 0.5% formic acid at gradient conditions. The linear range was 0.015–0.07, 825–7,218, 140–520, 63.42–160.75 and 38.25 × 103 to 110.35 × 103 ng/ml for dopamine, GABA, serotonin, glutamine and glutamate, respectively. Inter- and intra-run accuracy were in the range 97.82–103.37% with a precision (CV%) of ≤0.90%. The results revealed that 4 weeks of cigarette exposure significantly increased neurotransmitter levels after exposure to tobacco cigarettes in various brain regions, including the hippocampus and the amygdala. This increase in neurotransmitters levels may in turn activate the nicotine dependence pathway. 相似文献

Hydrazones are active compounds having an azomethine –NHN=CH group and are widely studied owing to their ease of preparation and diverse pharmacological benefits. Novel isonicotinic hydrazone derivatives of vanillin aldehyde and salicyl aldehyde were synthesized that had azomethine linkages and were characterized by UV–Visible, FTIR, EI-MS, 1H-NMR and 13C-NMR spectroscopy. The compounds were screened for their antibacterial activity against Staphylococcus aureus, Bacillus subtilus, and Escherichia coli using disc diffusion and minimum inhibitory concentration (MIC) methods. For cytotoxicity, a brine shrimp lethality test was performed to calculate the lethal concentration (LC50). The results demonstrated appreciable antibacterial activities against the applied strains, amongst which the compounds coded NH3 and NH5 showed maximum inhibition and MIC responses. In terms of cytotoxic activity, the maximum effect was observed in compound NH5 and NH6 treatments with minimum survival percentages of 36.10 ± 3.45 and 32.44 ± 2.0, respectively. Breast cancer is one of the most prevalent cancers worldwide usually treated with Tamoxifen. Tamoxifen resistance development is the most challenging issue in an initially responsive breast tumor, and mechanisms of resistance are still under investigation. The objective of this study is to develop and validate a selective, sensitive, and simultaneous high performance liquid chromatography–tandem mass spectrometry method to explore the changes in substrates and metabolites in supernatant media of developed Tamoxifen resistance MCF-7 cells. We focus on the determination of lactate, pyruvate, and L-glutamine which enables the tracking of changes in metabolic pathways as a result of the resistance process. Chromatographic separation was achieved within 3.5 min. using a HILIC column (4.6 × 100 mm, 3.5 µm particle size) and mobile phase of 0.05 M acetic acid–ammonium acetate buffer solution pH 3.0: Acetonitrile (40:60 v/v). The linear range was 0.11–2.25, 0.012–0.227, and 0.02–0.20 mM for lactate, pyruvate, and L-glutamine, respectively. Within- and between-run accuracy was in the range 98.94-105.50% with precision (CV, %) of ≤0.86%. The results revealed a significant increase in both lactate and pyruvate production after acquiring the resistant. An increase in L-glutamine levels was also observed and could be attributed to its over production or decline in its consumption. Therefore, further tracking of genes responsible of lactate, pyruvate, and glutamine metabolic pathways should be performed in parallel to provide in-depth explanation of resistance mechanism. 相似文献

Using each node's degree as a proxy for its importance, the topological hierarchy of a complex network is introduced and quantified. We propose a simple dynamical process used to construct networks which are either maximally or minimally hierarchical. Comparison with these extremal cases as well as with random scale-free networks allows us to better understand hierarchical versus modular features in several real-life complex networks. For random scale-free topologies the extent of topological hierarchy is shown to smoothly decline with gamma, the exponent of a degree distribution, reaching its highest possible value for gamma=2 and quickly approaching zero for gamma>3. 相似文献

A recent trend is the production of workable microchannel flow cells (MF cells). The nondestructive methods used to assess their reliability are based mainly on output monitoring and do not evaluate internal chemical interactions. We investigate a nondestructive method for evaluating changes in the chemical composition of the inner walls based on evaluation of the extent of alignment of a fluorescent probe in a liquid flowing within MF cells. Two MF cells were built with a 10-microm inner spacing. Their inner walls had four parallel SnO(2) strips, 2.00 mm wide, separated by 0.50-mm-wide glass strips. One cell had strips parallel to the flow and the other perpendicular. Flow-induced intermolecular alignment of rhodamine B in monoethylene glycol was scanned with 28-microm precision by fluorescence depolarization, using polarized-laser-induced fluorescence within induced flows (PLF-FI). No changes of polarization were seen when the flow was stopped. Under flowing conditions, polarization was always 4% lower in the glass region as compared to SnO(2). Glass had a higher solid-liquid interfacial tension (determined by contact angle measurements), thus being more wettable and increasing the drag, which propagates into the liquid flow, decreasing polarization. A method for the determination of salicylamide at concentrations between 25 and 350 ng ml(-1) by use of matrix isopotential synchronous fluorescence spectrometry (MISF) in combination with derivative techniques is proposed. The method allows the determination of compounds in samples with unknown background fluorescence without the need for tedious pre-separation. Synchronous scans are performed along a trajectory that connects points of identical intensity in a three-dimensional fluorescence spectrum. The unknown analytical signal of the serum is suppressed from the MISF spectrum, by calculating its first derivative at lambda(exc)=324 nm and lambda(em)=392 nm. In order to ensure maximum sensitivity and adequate selectivity, the experimental variables affecting the fluorescence intensity of the salicylamide band at lambda(exc)=328 nm and lambda(em)=418 nm were studied. Based on the results, the determination was performed in an aqueous medium at pH 12 that was adjusted with a sodium phosphate/hydrogen phosphate buffer. Calibration graphs were subjected to a comprehensive statistical analysis. The error propagation has been considered in order to calculate the detection limit by the criterium of Clayton. 相似文献

Binding studies of two sulfonamide-functionalized dibenzophenazine-based sensors are reported. Spectroscopic studies showed that both sensors are effective fluorescent turn-on sensors for several anions. Both sensors showed responses to acetate, benzoate, cyanide, and fluoride ions. NMR titrations confirmed the mode of binding of the sensors to be through H-bonding to the sulfonamide groups. 相似文献

The utility of surface-enhanced Raman scattering (SERS) spectrometry for trace determination of various chlorinated pesticides including chlordan, carbophenothion, bromophos, methyl chloropyrifos, dichloran, linuron, and 1-hydroxychlordene is described. Silver-coated substrates consisting of submicron spheres on solid surfaces were used as SERS-active media. The detection limits for these pesticides are at nanogram levels. The sensitivity and spectral selectivity of this technique for toxic chemical agents are discussed. The results indicate that SERS shows promise for the determination of an important class of chlorinated pesticides. 相似文献

Proper management of nonhealing wounds is an imperative clinical challenge. For the effective healing of chronic wounds, suitable wound coverage materials with the capability to accelerate cell migration, cell proliferation, angiogenesis, and wound healing are required to protect the healing wound bed. Biodegradable polymeric meshes are utilized as effective wound coverage materials to protect the wounds from the external environment and prevent infections. Among them, electrospun biopolymeric meshes have got much attention due to their extracellular matrix mimicking morphology, ability to support cell adhesion, and cell proliferation. Herein, electrospun nanocomposite meshes based on polycaprolactone (PCL) and titanium dioxide nanorods (TNR) are developed. TNR incorporated PCL meshes are fabricated by electrospinning technique and characterized by scanning electron microscopy, energy dispersive X‐ray spectroscopy, Fourier transform infrared spectroscopy (FTIR) analysis, and X‐Ray diffraction (XRD) analysis. In vitro cell culture studies, in ovo angiogenesis assay, in vivo implantation study, and in vivo wound healing study are performed. Interestingly, obtained in vitro and in vivo results demonstrated that the presence of TNR in the PCL meshes greatly improved the cell migration, proliferation, angiogenesis, and wound healing. Owing to the above superior properties, they can be used as excellent biomaterials in wound healing and tissue regeneration applications. 相似文献
